18-bromo-7-methoxychrysin inhibits properties of liver cancer stem cells via downregulation of β-catenin显示文摘AIM:To evaluate whether 8-bromo-7-methoxychrysin(BrMC),a synthetic analogue of chrysin,inhibits the properties of cancer stem cells derived from the human liver cancer MHCC97 cell line and to determine the potential mechanisms.METHODS:CD133+cells were sorted from the MHCC97 cell line by magnetic activated cell sorting,and amplified in stem cell-conditioned medium to obtain the enriched CD133+sphere forming cells(SFCs).The stem cell properties of CD133+SFCs were validated by the tumorsphere formation assay in vitro and the xenograft nude mouse model in vivo,and termed liver cancer stem cells(LCSCs).The effects of BrMC on LCSCs in vitro were evaluated by MTT assay,tumorsphere formation assay and transwell chamber assay.The effects of BrMC on LCSCs in vivo were determined using a primary and secondary xenograft model in Balb/c-nu mice.Expressions of the stem cell markers,epithelialmesenchymal transition(EMT)markers andβ-catenin protein were analyzed by western blotting or immunohistochemical analysis.RESULTS:CD133+SFCs exhibited stem-like cell properties of tumorsphere formation and tumorigenesis capacity in contrast to the parental MHCC97 cells.We found that BrMC preferentially inhibited proliferation and self-renewal of LCSCs(P<0.05).Furthermore,BrMC significantly suppressed EMT and invasion of LCSCs.Moreover,BrMC could efficaciously eliminate LCSCs in vivo.Interestingly,we showed that BrMC decreased the expression ofβ-catenin in LCSCs.Silencing ofβ-catenin by small interfering RNA could synergize the inhibition of self-renewal of LCSCs induced by BrMC,while Wnt3a treatment antagonized the inhibitory effects of BrMC.CONCLUSION:BrMC can inhibit the functions and characteristics of LCSCs derived from the liver cancer MHCC97 cell line through downregulation ofβ-catenin expression.Mei-Fang Quan Li-Hong Xiao Zhi-Hong Liu Hui Guo Kai-Qun Ren Fei Liu Jian-Guo Cao Xi-Yun Deng 2013World Journal of Gastroenterology2013,19,43:22

4Panretinal photocoagulation versus panretinal photocoagulation plus intravitreal bevacizumab for high-risk proliferative diabetic retinopathy显示文摘AIM: To evaluate the effects of panretinal photocoagulation(PRP) compared with PRP plus intravitreal bevacizumab(IVB) in patients with high-risk proliferative diabetic retinopathy(PDR) according to the Early Treatment Diabetic Retinopathy Study criteria.· METHODS: The data were collected retrospectively from the eyes of high-risk PDR patients, which were divided into two groups. After treated with standard PRP,the eyes were randomly assigned to receive only PRP(PRP group) or PRP plus intravitreal injection of 1.25 mg of bevacizumab(PRP-Plus group). Patients underwent complete ophthalmic evaluation, including best corrected visual acuity(BCVA), intraocular pressure(IOP), and new vessel size in fluorescein angiography(FA) and optical coherence tomography for the assessment of central subfield macular thickness(CSMT) at baseline and at weeks 12(±2), 16(±2), 24(±2) and 48(±2). Main outcome measures also included vitreous clear-up time and neovascularization on the disc(NVD) regression time.Adverse events associated with intravitreal injection were investigated.·RESULTS: Thirty consecutive patients(n =36 eyes)completed the 48-week follow-up. There was no significant difference between the PRP and PRP-Plus groups with respect to age, gender, type or duration of diabetes, area of fluorescein leakage from active neovascularizations(NVs), BCVA or CSMT at baseline.The mean vitreous clear-up time was 12.1 ±3.4wk after PRP and 8.4 ±3.5wk after PRP combined with IVB. The mean time interval from treatment to complete NVD regression on FA examination was 15.2 ±3.5wk in PRP group and 12.5±3.1wk in PRP-Plus group. No significant difference in CSMT was observed between the groups throughout the study period. However, the total area of actively leaking NVs was significantly reduced in the PRP-Plus group compared with the PRP group(P <0.05).Patients received an average of 1.3 injections(range:1-2).Ten eyes(27.8%) underwent 2 injections. Two eyes had ocular complication of PDR progression to dense vitreous hemorrhage(VH). No major adverse events were identified.·CONCLUSION: The adjunctive use of IVB with PRP is associated with a greater reduction in the area of active leaking NVs than PRP alone in patients with high-risk PDR. Short-term results suggest combined IVB and PRP achieved rapid clearance of VH and regression of retinal NV in the treatment of high-risk PDR. 5SbHKT1;4, a member of the high‐affinity potassium transporter gene family from Sorghum bicolor, functions to maintain optimal Na^+/K^+ balance under Na^+ stress显示文摘In halophytic plants, the high‐affinity potassium transporter HKT gene family can selectively uptake Kt in the presence of toxic concentrations of Nat. This has so far not been well examined in glycophytic crops. Here, we report the characterization of SbHKT1;4, a member of the HKT gene family from Sorghum bicolor. Upon Nat stress, SbHKT1;4 expression was more strongly upregulated in salt‐tolerant sorghum accession, correlating with a better balanced Nat/ Kt ratio and enhanced plant growth. Heterogeneous expression analyses in mutants of Saccharomyces cerevisiae and Arabidopsis thaliana indicated that overexpressing SbHKT1;4 resulted in hypersensitivity to Nat stress, and such hypersensitivity could be alleviated with the supply of elevated levels of Kt, implicating that SbHKT1;4 may mediate Kt uptake in the presence of excessive Nat. Further electrophysiological evidence demonstrated that SbHKT1;4 could transport Nat and Kt when expressed in Xenopus laevis oocytes. 6Origin and accumulation of high-maturity oil and gas in deep parts of the Baxian Depression, Bohai Bay Basin, China显示文摘Great quantities of light oil and gas are produced from deep buried hill reservoirs at depths of 5,641 m to 6,027 m and 190 °C to 201 °C in the Niudong-1 Well, representing the deepest and hottest commercial hydrocarbons discovered in the Bohai Bay Basin in eastern China. This discovery suggests favorable exploration prospects for the deep parts of the basin. However, the discovery raises questions regarding the genesis and accumulation of hydrocarbons in deep reservoirs. Based on the geochemical features of the hydrocarbons and characteristics of the source rocks as well as thermal simulation experiments of hydrocarbon generation, we conclude that the oil and gas were generated from the highly mature Sha-4 Member (Es4 ) source rocks instead of thermal cracking of crude oils in earlier accumulations. The source kitchen with abnormal pressures and karsted carbonate reservoirs control the formation of high-maturity hydrocarbon accumulations in the buried hills (i.e., Niudong-1) in conjunction with several structural-lithologic traps in the Es4 reservoirs since the deposition of the upper Minghuazhen Formation. 7Formability,microstructure evolution and mechanical properties of wire arc additively manufactured AZ80M magnesium alloy using gas tungsten arc welding显示文摘Wire arc additive manufacturing(WAAM)technology has been used to fabricate the multi-layer single-pass deposited wall of AZ80M magnesium(Mg)alloy by gas tungsten arc welding.The formability,thermal cycles,microstructural evolution and mechanical properties of the WAAM AZ80M Mg alloy were investigated.The results show that there was significant difference in the temperature variation and the geometries between the original several layers and the subsequent deposited layers.Owing to the arc energy input,the interpass temperature rised rapidly and then stabilized at 150℃.As a result,the width of the deposited wall increased and then kept stable.There were obvious differences in the microstructure of the WAAM AZ80M Mg alloy among the top zone,intermediate zone and bottom zone of deposited wall.During the arc deposition process,theβphase of the WAAM AZ80M Mg alloy redissolved due to the cyclic heat accumulation,and then precipitated in the grain boundary.The cyclic heat accumulation also led to weakening of dendrite segregation.From the substrate to the top zone,the hardness of the deposited wall decreased gradually,and the intermediate zone which was the main body of deposited wall had relatively uniform hardness.The tensile properties of the WAAM AZ80M Mg alloy were different between the vertical direction and the horizontal direction.And the maximum ultimate tensile strength of the WAAM AZ80M Mg alloy was 308 MPa which was close to that of the as-extruded AZ80M Mg alloy.Yangyang Guo Gaofeng Quan Yinglong Jiang Lingbao Ren Lingling Fan Houhong Pan 2021Journal of Magnesium and Alloys2021,9,1:10
8Yu Gan Long Ameliorates Hepatic Fibrosis by Inhibiting PI3K/AKT,Ras/ERK and JAK1/STAT3 Signaling Pathways in CCl4-induced Liver Fibrosis Rats显示文摘Yu Gan Long(YGL)is a Chinese traditional herbal formula which has been reported to attenuate liver fibrosis for many years and we have explored its anti-fibrotic mechanism through blocking transforming growth factor(TGF-β)in the previous study.But the mechanisms associated with platelet-derived growth factor(PDGF)-BB remain obscure.In this study,we further investigated the mechanism of YGL reducing carbon tetrachloride(CCl4)-induced liver fibrosis in rats.Our results showed that YGL suppressed CCl4-induced upregulation of collagen IV(Col IV),type HI precollagen(PCHI),hyaluronuc acid(HA)and laminin(LN),which are implicated in liver fibrosis.Also,YGL reduced theα-smooth muscle actin(α-SMA)expression,which acts as the indicator of liver fibrosis.Furthermore,YGL decreased the serum levels of hepatic stellate cell(HSC)mitogen PDGF-BB and inflammation cytokines,including TNF-α,IL-1β,IL-6.Markers involved in liver fibrosis,such as Ras,p-Raf-1,p-ERK1/2,p-JNK,p-P38,p-PI3K,p-AKT,p-JAKl,p-STAT3 were downregulated significantly after treatment with YGL.Our results indicated that YGL ameliorated CCl4-induced liver fibrosis by reducing inflammation cytokines production,and suppressing Ras/ERK,PI3K/AKT,and JAK1/STAT3 signaling pathways,which provided further evidence towards elucidation of the anti-fibrotic mechanism of YGL.Hou-gang LI Peng-tao YOU Yu XI Yu CAI Yi-jun TU Meng-heng WANG Wan-ci SONG Tai-min QUAN Hui-ying REN Yan-wen LIU Han-xiong DAN Shi-qing XU 2020Current Medical Science2020,40,3:9
9Synergistic effects of biochar/microbial inoculation on the enhancement of pig manure composting显示文摘With the aim to comparatively investigate the effect of the addition of additives such as biochar(BC),microbial inoculation(MI)and biochar/microbial inoculation(BCMI)on composting enhancement,nitrogen conservation,greenhouse gas emissions,the quality improvement of pig manure compost were comparatively investigated in a 42-day aerobic pig manure composting experiment.The results showed that the duration of the thermophilic stage,the degradation of organic matter and the detoxification of the compost were enhanced in the BC,MI and BCMI treatments compared with those in the control(without additive).Moreover,the content of total Kjeldahl nitrogen in the BC,MI and BCMI treatments was increased by 38.1,48.9 and 59.0%,respectively,through the reduction of NH_(3) volatilization and N_(2)O production,which were higher than those of the control(31.8%).A reduction in CH_(4) release during composting was not observed with the MI and BCMI treatments.Following 42 days of composting,the final product of the three treatments exhibited acceptable potential for use as a fertilizer in agriculture;the BCMI treatment showed an especially synergistic effect on pig manure composting enhancement.Zhineng Tu Xiuna Ren Junchao Zhao Sanjeev Kumar Awasthi Quan Wang Mukesh Kumar Awasthi Zengqiang Zhang Ronghua Li 2019Biochar2019,1,1:8
10Radial propagation of magnetospheric substorm-injected energetic electrons observed using a BD-IES instrument and Van Allen Probes显示文摘In cases where substorm injections can be observed simultaneously by multiple spacecraft,they can help elucidate the potential mechanisms of particle transport and energization,of great importance to understanding and modeling the magnetosphere.In this paper,using data returned from the BeiDa-IES(BD-IES) instrument onboard a satellite in an inclined(55°) geosynchronous orbit(IGSO),in combination with two geo-transfer orbiting(GTO) satellite Van Allen Probes(A and B),we analyze a substorm injection event that occurred on the 16 th of October 2015.During this substorm injection,the IGSO onboard BD-IES was outbound,while both Van Allen Probe satellites(A and B) were inbound,a configuration of multiple trajectories that provides a unique opportunity to simultaneously investigate both the inward and outward radial propagation of substorm injection.Indicated by AE/AL indices,this substorm was closely related to an IMF/solar wind discontinuity that showed a sharp change in IMF Bz direction to the north.The innermost signature of this substorm injection was detected by Van Allen Probes A and B at L-3.7,while the outermost signature was observed by the onboard BD-IES instrument at L~\0.These data indicate that the substorm had a global,rather than just local,effect.Finally,we suggest that electric fields carried by fast-mode compressional waves around the substorm injection are the most likely candidate mechanism for the electron injection signatures observed in the inner- and outermost inner magnetosphere.ZONG QiuGang HAO YiXin ZOU Hong FU SuiYan ZHOU XuZhi REN Jie WANG LingHua YUAN ChongJing LIU ZhiYang JIA XiangHong QUAN Lin LIU Ying WANG YongFu 2016Science China Earth Sciences2016,59,7:8
11Abundance and composition of ammonia-oxidizing bacteria and archaea in different types of soil in the Yangtze River estuary显示文摘Tidal flats are soil resources of great significance.Nitrification plays a central role in the nitrogen cycle and is often a critical first step in nitrogen removal from estuarine and coastal environments.We determined the abundance as well as composition of ammonia-oxidizing bacteria(AOB) and ammonia-oxidizing archaea(AOA) in different soils during land reclamation process.The abundance of AOA was higher than that of AOB in farm land and wild land while AOA was not detected in tidal flats using real-time polymerase chain reaction(PCR).The different abundances of AOB and AOA were negatively correlated with the salinity.The diversities of AOB and AOA were also investigated using clone libraries by amplification of amoA gene.Among AOB,nearly all sequences belonged to the Nitrosomonas lineage in the initial land reclamation process,i.e.,tidal flats,while both Nitrosomonas and Nitrosospira lineages were detected in later and transition phases of land reclamation process,farm land and wild land.The ratio of the numbers of sequences of Nitrosomonas and Nitrosospira lineages was positively correlated with the salinity and the net nitrification rate.As for AOA,there was no obvious correlation with the changes in the physicochemical properties of the soil.This study suggests that AOB may be more import than AOA with respect to influencing the different land reclamation process stages.Xiao-ran LI Yi-ping XIAO Wen-wei REN Zeng-fu LIU Jin-huan SHI Zhe-xue QUAN 2012Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2012,13,10:6
12Docking control for probe-drogue refueling: An additive-state-decomposition-based output feedback iterative learning control method显示文摘Designing a controller for the docking maneuver in Probe-Drogue Refueling(PDR) is an important but challenging task, due to the complex system model and the high precision requirement.In order to overcome the disadvantage of only feedback control, a feedforward control scheme known as Iterative Learning Control(ILC) is adopted in this paper.First, Additive State Decomposition(ASD) is used to address the tight coupling of input saturation, nonlinearity and the property of Non Minimum Phase(NMP) by separating these features into two subsystems(a primary system and a secondary system).After system decomposition, an adjoint-type ILC is applied to the Linear Time-Invariant(LTI) primary system with NMP to achieve entire output trajectory tracking, whereas state feedback is used to stabilize the secondary system with input saturation.The two controllers designed for the two subsystems can be combined to achieve the original control goal of the PDR system.Furthermore, to compensate for the receiverindependent uncertainties, a correction action is proposed by using the terminal docking error,which can lead to a smaller docking error at the docking moment.Simulation tests have been carried out to demonstrate the performance of the proposed control method, which has some advantages over the traditional derivative-type ILC and adjoint-type ILC in the docking control of PDR.Jinrui REN Quan QUAN Cunjia LIU Kai-Yuan CAI 2020Chinese Journal of Aeronautics2020,33,3:5

14Paleoclimatic indication of X-ray fluorescence core-scanned Rb/Sr ratios: A case study in the Zoige Basin in the eastern Tibetan Plateau显示文摘The X-ray fluorescence(XRF)core-scanned Rb/Sr ratios of lake sediments have been widely used as a proxy for chemical weathering intensity and past climate change.However,some factors could affect Rb/Sr ratios,causing misinterpretation and limiting its application.In this study,we present a high-resolution XRF core-scanned Rb/Sr record of core ZB13-C1 from the Zoige Basin in the eastern Tibetan Plateau.To validate its application,we correlated this record with the chemical index of alteration(CIA)and other paleoclimatic proxies.Our results showed that(1)the core-scanned Rb/Sr ratios and CIA were reliable proxies of chemical weathering intensity in fine-grained sedimentary sequences;(2)the low values of corescanned Rb/Sr ratios and CIA were significantly correlated with high total organic carbon content,arboreal pollen content,carbonate content,and C/N ratios,confirming its reliability as a proxy for the Asian summer monsoon intensity;(3)the corescanned Rb/Sr ratios at core depths of 25–0.3 and 56–47 m were unable to reliably reflect chemical weathering intensity due to both the grain-size effect and the low test accuracy.Our study highlights the need for mutual verification of multiple indicators before accurately applying Rb/Sr as a paleoclimatic proxy in other similar study areas.Hanfei YANG Yan ZHAO Qiaoyu CUI Weihe REN Quan LI 2021Science China Earth Sciences2021,64,1:5

16Effect of heat treatment on the microstructure and mechanical properties of AZ80M magnesium alloy fabricated by wire arc additive manufacturing显示文摘To maximize the benefits of wire arc additive manufacturing(WAAM)processes,the effect of post-deposition heat treatment on the microstructure and mechanical properties of WAAM AZ80M magnesium(Mg)alloy was investigated.Three different heat treatment procedures(T4,T5 and T6)were performed.According to the results,after T4 heat treatment,the microsegregation of alloying elements was improved with the eutectic structure dissolved.Samples after T5 heat treatment inherited the net-like distribution of secondary phases similar to the as-deposited sample,where the eutectic structure covering the interdendritic regions and theβ-phase precipitated around the eutectic structure.After T6 heat treatment,the tinyβ-phases re-precipitated from the matrix and distributed in inner and outer of the grains.The hardness distribution of the samples went through T4 and T6 heat treatment was more uniform in comparison to that of T5 heat treated samples.The tensile test showed that the T6 heat treatment improved the strength and ductility,and the anisotropy between horizontal and vertical can be eliminated.Moreover,T4 treated samples exhibited highest ductility.Yangyang Guo Gaofeng Quan Mert Celikin Lingbao Ren Yuhang Zhan Lingling Fan Houhong Pan 2022Journal of Magnesium and Alloys2022,10,7:5
17Ultraviolet-to-microwave room-temperature photodetectors based on three-dimensional graphene foams显示文摘Highly sensitive broadband photodetection is of critical importance for many applications.However,it is a great challenge to realize broadband photodetection by using a single device.Here we report photodetectors(PDs)based on three-dimensional(3 D)graphene foam(GF)photodiodes with asymmetric electrodes,which show an ultra-broadband photoresponse from ultraviolet to microwave for wavelengths ranging from 10~2 to 10~6 nm.Moreover,the devices exhibit a high photoresponsivity of 10~3 A·W^-1,short response time of 43 ms,and3 d B bandwidth of 80 Hz.The high performance of the devices can be attributed to the photothermoelectric(PTE,also known as the Seebeck)effect in 3 D GF photodiodes.The excellent optical,thermal,and electrical properties of 3 D GFs offer a superior basis for the fabrication of PTE-based PDs.This work paves the way to realize ultra-broadband and high-sensitivity PDs operated at room temperature.YIFAN LI YATING ZHANG YU YU ZHILIANG CHEN QINGYAN LI TENGTENG LI JIE LI HONGLIANG ZHAO QUAN SHENG FENG YAN ZHEN GE YUXIN REN YONGSHENG CHEN JIANQUAN YAO 2020Photonics Research2020,8,3:4
18Combining WV-2 images and tree physiological factors to detect damage stages of Populus gansuensis by Asian longhorned beetle (Anoplophora glabripennis) at the tree level显示文摘Background:Anoplophora glabripennis(Motschulsky),commonly known as Asian longhorned beetle(ALB),is a wood-boring insect that can cause lethal infestation to multiple borer leaf trees.In Gansu Province,northwest China,ALB has caused a large number of deaths of a local tree species Populus gansuensis.The damaged area belongs to Gobi desert where every single tree is artificially planted and is extremely difficult to cultivate.Therefore,the monitoring of the ALB infestation at the individual tree level in the landscape is necessary.Moreover,the determination of an abnormal phenotype that can be obtained directly from remote-sensing images to predict the damage degree can greatly reduce the cost of field investigation and management.Methods:Multispectral WorldView-2(WV-2)images and 5 tree physiological factors were collected as experimental materials.One-way ANOVA of the tree’s physiological factors helped in determining the phenotype to predict damage degrees.The original bands of WV-2 and derived vegetation indices were used as reference data to construct the dataset of a prediction model.Variance inflation factor and stepwise regression analyses were used to eliminate collinearity and redundancy.Finally,three machine learning algorithms,i.e.,Random Forest(RF),Support Vector Machine(SVM),Classification And Regression Tree(CART),were applied and compared to find the best classifier for predicting the damage stage of individual P.gansuensis.Results:The confusion matrix of RF achieved the highest overall classification accuracy(86.2%)and the highest Kappa index value(0.804),indicating the potential of using WV-2 imaging to accurately detect damage stages of individual trees.In addition,the canopy color was found to be positively correlated with P.gansuensis’damage stages.Conclusions:A novel method was developed by combining WV-2 and tree physiological index for semi-automatic classification of three damage stages of P.gansuensis infested with ALB.The canopy color was determined as an abnormal phenotype that could be directly assessed using remote-sensing images at the tree level to predict the damage degree.These tools are highly applicable for driving quick and effective measures to reduce damage to pure poplar forests in Gansu Province,China.Quan Zhou Xudong Zhang Linfeng Yu Lili Ren Youqing Luo 2021Forest Ecosystems2021,8,3:3
