1Fibronectin: Functional character and role in alcoholic liver disease显示文摘Fibronectins are adhesive glycoproteins that can be found in tissue matrices and circulating in various fluids of the body. The variable composition of fibronectin molecules facilitates a diversity of interactions with cell surface receptors that suggest a role for these proteins beyond the structural considerations of the extracellular matrix. These interactions implicate fibronectin in the regulation of mechanisms that also determine cell behavior and activity. The two major forms, plasma fibronectin (pFn) and cellular fibronectin (cFn), exist as balanced amounts under normal physiological conditions. However, during injury and/or disease, tissue and circulating levels of cFn become disproportionately elevated. The accumulating cFn, in addition to being a consequence of prolonged tissue damage, may in factstimulate cellular events that promote further damage. In this review, we summarize what is known regarding such interactions between fibronectin and cells that may influence the biological response to injury. We elaborate on the effects of cFn in the liver, specifically under a condition of chronic alcohol-induced injury. Studies have revealed that chronic alcohol consumption stimulates excess production of cFn by sinusoidal endothelial cells and hepatic stellate cells while impairing its clearance by other cell types resulting in the build up of this glycoprotein throughout the liver and its consequent increased availability to influence cellular activity that could promote the development of alcoholic liver disease. We describe recent findings by our laboratory that support a plausible role for cFn in the promotion of liver injury under a condition of chronic alcohol abuse and the implications of cFn stimulation on the pathogenesis of alcoholic liver disease. These findings suggest an effect of cFn in regulating cell behavior in the alcohol-injured liver that is worth further characterizing not only to gain a more comprehensive understanding of the role this reactive glycoprotein plays in the progression of injury but also for the insight further studies could provide towards the development of novel therapies for alcoholic liver disease.Razia S Aziz-Seible Carol A Casey 2011World Journal of Gastroenterology2011,17,20:5
2An evaluation of OSOM BV blue test in the diagnosis of bacterial vaginosis显示文摘Objective:To determine the prevalence of bacterial vaginosis(BV) in patients with vaginal discharge and evaluate the efficacy of OSOM BV blue test in diagnosis.Methods:OSOM BV blue test,a rapid diagnostic test based on the principle of detection of bacterial sialidase activity in vaginal fluid specimens was conducted.A total of 405 patients in the reproductive age group(15- 45 years) having vaginal discharge were included in the study along with 10 healthy age-matched controls.Two high vaginal swabs were collected aseptically from each patient.One swab was used to make smear for gram staining,and the other was for OSOM BV blue test.Amine test and vaginal pH test were taken as well.Results:The prevalence of bacterial vaginosis was 60.7%. OSOM BV blue test showed good efficacy,as compared with gram staining in diagnosing BV.The sensitivity and specificity of OSOM BV blue test were 97.6%and 97.5%respectively.Amsel’s criteria diagnosed 180(44.4%) cases of BV and had sensitivity and specificity of 67.1%and 90.6%respectively.Thus the performance of OSOM BV blue was better than the methods based on Amsel’s criteria.Conclusions:OSOM BV blue test is an efficacious bed side test,helpful in rapidly making an accurate diagnosis of BV in setups lacking laboratory facilities or expert microbiologists.Fatima Shujatullah Haris M Khan Razia Khatoon Tamkin Rabbani Abida Malik 2010Asian Pacific Journal of Tropical Medicine2010,3,7:3
316S rDNA-Based Phylogeny of Non-Symbiotic Bacteria of Entomopathogenic Nematodes from Infected Insect Cadavers显示文摘Using 16S rDNA gene sequencing technique, three different species of non-symbiotic bacteria of entomopatho-genic nematodes (EPNs) (Steinernema sp.and Heterorhabditis sp.) were isolated and identified from infected insect cadavers(Galleria mellonella larvae) after 48-hour post infections.Sequence similarity analysis revealed that the strains SRK3, SRK4 and SRK5 belong to Ochrobactrum cytisi,Schineria larvae and Ochrobactrum anthropi,respectively.The isolates O.anthropi and S.larvae were found to be associated with Heterorhabditis indica strains BDU-17 and Yer-136,respectively,whereas O.cytisi was associated with Steinernema siamkayai strain BDU-87. Phenotypically, temporal EPN bacteria were fairly related to symbiotic EPN bacteria (Photorhabdus and Xenorhabdus genera). The strains SRK3 and SRK5 were phylogeographically similar to several non-symbionts and contaminated EPN bacteria isolated in Germany(LMG3311T) and China (X-14),while the strain SRK4 was identical to the isolates of S.larvae (L1/57,L1/58, L1/68 and L2/11) from Wohlfahrtia magnifica in Hungary.The result was further confirmed by RNA secondary structure and minimum energy calculations of aligned sequences.This study suggested that the non-symbionts of these nematodes are phylogeographically diverged in some extent due to phase variation.Therefore,these strains are not host-dependent, but environment-specific isolates.M. 4Cellular fi bronectin stimulates hepatocytes to produce factors that promote alcohol-induced liver injury显示文摘AIM:To examine the consequences of cellular f ibronectin(cFn)accumulation during alcohol-induced injury,and inv estigate whether increased cFn could have an effect on hepatocytes(HCs)by producing factors that could cont ribute to alcohol-induced liver injury.METHODS:HCs were isolated from rats fed a control or ethanol liquid diet for four to six weeks.Exogenous c Fn(up to 7.5 μg/mL)was added to cells cultured for 20 h,and viability(lactate dehydrogenase),apoptosis(caspase activity)and se cretion of proinflammat-ory cytokines(tumor ne c rosis fac tor alpha,TNF-α and interleukin 6,IL-6),mat rix metalloproteinases(MMPs)and their inhibitors(tissue inhibitors of metall-oproteinases,TIMPs)was det ermined.Degrad ation of iodinated cFn was det ermined over a 3 h time period in the preparations.RESULTS:cFn degradation is impaired in HCs isolated from ethanol-fed animals,leading to its accumulation in the matrix.Addition of exogenous cFn did not affect viability of HCs from control or ethanolfed animals,and apoptosis was affected only at the higher concentration.Sec retion of MMPs,TIMPs,TNF-α and IL-6,however,was increased by exogenously added cFn,with HCs from ethanolfed animals showing increased susceptibility compared to the controls.CONCLUSION:These results suggest that the elevated amounts of cFn observed in alcoholic liver injury can stimulate hepatocytes to produce factors which promote further tissue damage.Razia S Aziz-Seible Benita L McVicker Kusum K Kharbanda Carol A Casey 2011World Journal of Hepatology2011,3,2:2
