| 1 | bHLH121 Functions as a Direct Link that Facilitates the Activation of FIT by bHLH IVc Transcription Factors for Maintaining Fe Homeostasis in Arabidopsis显示文摘Iron(Fe)deficiency is prevalent in plants grown in neutral or alkaline soil.Plants have evolved sophisticated mechanisms that regulate Fe homeostasis,ensuring survival.In Arabidopsis,FER-LIKE IRON DEFICIENCY-INDUCED TRANSCRIPTION FACTOR(FIT)is a crucial regulator of Fe-deficiency response.FIT is activated indirectly by basic helix-loop-helix(bHLH)IVc transcription factors(TFs)under Fed eficiency;how ever,it remains unclear which protein(s)act as the linker to mediate the activation of FIT by bHLH IVc TFs.In this study,we characterize the functions of bHLH121 and demonstrate that it directly associates with the FIT promoter.We found that loss-of-function mutations of bHLH121 cause severe Fedeficiency symptoms,reduced Feaccum ulation,and disrupted expression of genes associated with Fehomeostasis.Genetic analysis showed that FIT is epistatic to bHLH121 and FIT overexpression partially rescues the bhlh121 mutant.Further investigations revealed that bHLH IVc TFs interact with and promote nuclear accumulation of bHLH121.We demonstrated that bHLH121 has DNA-binding activity and can bind the prom oters of the FIT and bHLHlb genes,but we did not find that it has either direct transcriptional activation or repression activity tow ard these genes.Meanw hile,we found that bHLH121 functions downstream of and is a direct target of bHLH IVc TFs,and its expression is induced by Fe deficiency in a bHLH IV c-dependent manner.Taken together,these results establish that bHLH121 functions together with bHLH IVc TFs to positively regulate the expression of FIT and thus plays a pivotal role in maintaining Fe homeostasis in Arabidopsis. | Rihua Lei Yang Li Yuerong Cai Chenyang Li Mengna Pu Chengkai Lu Yujie Yang Gang Liang | 2020 | Molecular Plant2020,13,4: | 14 |
| 2 | A sulfur dioxide polymer prodrug showing combined effect with doxorubicin in combating subcutaneous and metastatic melanoma显示文摘Melanoma,as the most aggressive and treatment-resistant skin malignancy,is responsible for about 80%of all skin cancer mortalities.Prone to invade into the dermis and form distant metastases significantly reduce the patient survival rate.Therefore,early treatment of the melanoma in situ or timely blocking the deterioration of metastases is critical.In this study,a sulfur dioxide(SO_(2))polymer prodrug was designed as both an intracellular glutathione(GSH)-responsive SO_(2) generator and a carrier of doxorubicin(DOX),and used for the treatment of subcutaneous and metastatic melanoma.Firstly,chemical conjugation of 4-N-(2,4-dinitrobenzenesulfonyl)-imino-1-butyric acid(DIBA)onto the side chains of methoxy poly(ethylene glycol)grafted dextran(mPEG-g-Dex)resulted in the synthesis of the amphiphilic polymer prodrug of SO_(2),mPEG-g-Dex(DIBA).The obtained mPEG-g-Dex(DIBA)could self-assemble into stable micellar nanoparticles and exhibited a glutathione-responsive SO_(2) release behavior.Subsequently,DOX was encapsulated into the core of mPEG-g-Dex(DIBA)micelles to form DOX-loaded nanoparticles(PDDN-DOX).The formed PDDN-DOX could be internalized by B16F10 cells and synchronously release DOX and SO_(2) into the tumor cells.As a result,PDDN-DOX exerted synergistic anti-tumor effects in B16F10 melanoma cells because of the oxidative damage properties of SO_(2) and toxic effects of DOX.Furthermore,in vivo experiments verified that PDDN-DOX had great potential for the treatment of subcutaneous and metastasis melanoma.Collectively,our present work demonstrates that the combination of SO_(2)-based gas therapy and chemotherapeutics offers a new avenue for inhibiting melanoma progression and metastases. | Lin An Peng Zhang Wei Shen Xuan Yi Weitian Yin Rihua Jiang Chunsheng Xiao | 2021 | Bioactive Materials2021,6,5: | 3 |

| 4 | Comparative proteomic analysis of the effects of high-concentrate diet on the hepatic metabolism and inflammatory response in lactating dairy goats显示文摘Background: To understand the impact of feeding a high-concentrate diet to mid-lactating goats for a long time on liver metabolism and inflammatory response, two dimensional polyacrylamide gel electrophoresis(2-DE) and real-time PCR method were employed to detect proteins differentially expressed in liver and their m RNAs expression in goats fed high concentrate diet(HC) or low concentrate diet(LC). Twelve lactating dairy goats were randomly assigned to either a HC diet group(65 % concentrate of dry matter; n = 6) or a LC diet group(35 % concentrate of dry matter; n = 6) for 10 wk.Results: Twenty differentially expressed spots(≥2.0-fold changes) in the hepatic tissues were excised and successfully identified using MALDI TOF/TOF. Of these, 8 proteins were up-regulated, while the rest 12 proteins were down-regulated in HC goats compared to LC. Differential expressed proteins including alpha enolase 1(ENO1), glutamate dehydrogenase 1(GLUD1), glutathione S-transferase A1(GSTA1), ATP synthase subunit 5β(ATP5β), superoxide dismutase [Cu-Zn](SOD1), cytochrom c oxidase subunit Via(COX6A1) and heat shock protein 60(HSP60) were further verified by real-time PCR and/or western blot at m RNA or protein expression level. Consistent with the 2-DE results, a significant decrease of β-actin protein expression and SOD enzyme activity was observed in liver of HC goats(P < 0.05), while ENO1 protein expression was significantly up-regulated in HC compared to LC goats(P < 0.05). However, western blot analysis did not show a significant difference of hepatic HSP60 protein between HC and LC group, which did not match the decrease of HSP60 content detected by 2-DE analysis. Real-time PCR showed that glutathione S-transferase P1(GSTP1) and SOD1 m RNA expression was significantly decreased in liver of HC goats, while cytochrom c oxidase(COX3) and ATPase 8(ATP8) m RNAs expression were markedly increased compared to LC(P < 0.05). | 16 | Experimental Study of Rotor Flow Separation Control using a New Type of Dielectric Barrier Discharge Plasma Actuator显示文摘Flow separation occurring on rotor blades is an important limiting factor for helicopter performance. This paper presents a new type of dielectric barrier discharge(DBD) plasma actuator for rotor blade flow separation control called bipolar DBD plasma actuator. The bipolar DBD plasma actuator's connection mode differs from traditional plasma actuators and eliminates reverse discharge between electrodes. The experiments in this work were carried out by smoke-wire flow visualization and PIV technology in the open test section of a low speed wind tunnel, and solved the problem of high voltage electricity supplied to the plasma actuator while the rotor rotated. In this experiment, the rotor, camera, and laser were synchronized to obtain results. Smoke-wire results and PIV results illustrated that the flow separation weakened with increasing rotor speed; the separated flow area was large at a rotating speed of 300 r/min and gradually became smaller at the rotating speed of 600 r/min, the flow separation disappeared at the rotating speed of 1200 r/min.
