1Metabolomics characterization of colostrum in three sow breeds and its influences on piglets' survival and litter growth rates显示文摘Background: Colostrum is the first secretion produced by mammary glands during the hours immediately preceding and succeeding parturition. This secretion differs from milk and represents an essential vehicle of passive immunity,prebiotic compounds and growth factors involved in intestinal development. Most of the literature concerning colostrum composition refers mainly to human and cow; and little is known about pig colostrum metabolome and how it varies between pig breeds and different farrowing parity. Thus, the aim of the present research is to provide new information about pig colostrum composition and the associations between metabolites, the sows' breed and the survival and growth rates of their litters.Results: Colostrum samples were gathered from 58 parturitions of sows belonging to three different breeds chosen for their importance in Italian heavy pig production: 31 Large White, 15 Landrace and 12 Duroc respectively. The defatted and ultrafiltered colostrum samples were analysed using1 H–NMR spectroscopy. Principal Components Analysis(PCA) was assessed on the obtained spectra. In addition, using a Stepwise Regression and a Linear Regression analyses the metabolites named after the signals assignment were tested for their associations with piglets' performances.Twenty-five metabolites were identified, comprehending monosaccharides, disaccharides(such as lactose), organic acids(lactate, citrate, acetate and formate), nitrogenous organic acids(such as creatine) and other compounds,including nucleotides. PCA results evidence a clustering due to breed and season effects. Lactose was the main compound determining the assignment of the samples into different clusters according to the sow breed. Furthermore, some metabolites showed to be associated with piglets' performance and survival traits: acetate and taurine were positively related to litter weight gain and piglets' survival rate, respectively, while dimethylamine and cis-aconitate were linked to new-borns' impaired ability to survive.Conclusions: The results obtained suggest that colostrum composition is affected by breed, which, together with environmental conditions, may cause changes in colostrum metabolites content with possible consequences on piglets' performances. 2Search for naive human pluripotent stem cells显示文摘Normal mouse pluripotent stem cells were originally derived from the inner cell mass(ICM) of blastocysts and shown to be the in vitro equivalent of those pre-implantation embryonic cells, and thus were called embryonic stem cells(ESCs). More than a decade later, pluripotent cells were isolated from the ICM of human blastocysts. Despite being called human ESCs, these cells differ significantly from mouse ESCs, including different morphology and mechanisms of control of pluripotency, suggesting distinct embryonic origins of ESCs from the two species. Subsequently, mouse pluripotent stem cells were established from the ICMderived epiblast of post-implantation embryos. These mouse epiblast stem cells(Epi SCs) are morphological and epigenetically more similar to human ESCs. This raised the question of whether cells from the human ICM are in a more advanced differentiation stage than their murine counterpart, or whether the available culture conditions were not adequate to maintain those human cells in their in vivo state, leading to a transition into Epi SC-like cells in vitro. More recently, novel culture conditions allowed the conversion of human ESCs into mouse ESC-like cells called nave(or ground state) human ESCs, and the derivation of nave human ESCs from blastocysts. 4Height-to-diameter ratios with temporal and dendro/morphometric variables for Brazilian pine in south Brazil显示文摘Height-to-diameter ratios(HD)are an important measure of the stability,density and competition of forest stands.It refl ects the vertical growth of the trees,the vulnerability of the forest canopy structure and infl uences volumetric production.HD ratios vary according to tree size,availability of resources for growth,stand density and species composition.Data were taken from 210 trees and a regression technique of generalized linear models for the HD ratio applicable for forest structure conservation was developed.The objective of this study was to model the HD ratios of dominant and co-dominant trees of Araucaria angustifolia according to morphometric,dendrometric,annual diameter increment,stand density,and age variables in three sites in southern Brazil.The results show that the HD ratio decreases with increasing age,crown area and basal area,and increases with stand density and annual diameter increment.Accuracy of the developed equations was demonstrated by the values of deviation,Bayesian and Akaike criteria.The results are of interest to forest managers since they make decisions about silvicultural operations.Growth continuity and forest production indicate that any intervention should be directed at younger trees of smaller sizes,and that one of the main management factors for stand stability and growth is the formation of the stand and its capture of light.Andre Felipe Hess Myrcia Minatti Emanuel Arnoni Costa Luis Paulo Baldissera Schorr Gabriel Teixeira da Rosa Isadora de Arruda Souza Geedre Adriano Borsoi Veraldo Liesenberg Thiago Floriani Stepka Roberta Abatti 2021Journal of Forestry Research2021,32,1:2

15Latent tuberculosis:Risk factors,screening and treatment in liver transplantation recipients from an endemic area显示文摘BACKGROUND Patients undergoing solid organ transplantation,particularly those who live or have lived in tuberculosis(TB)endemic areas,are at a high risk of developing TB.The majority of post-transplantation TB cases are associated with reactivation of latent TB infection(LTBI).Brazil is in a single position with overlapping areas of high TB endemicity and high transplant activity.In liver transplant(LT),one should be aware of the potential hepatotoxicity associated with the treatment regimens for LTBI.AIM To evaluate the frequency of LTBI in LT patients and treatment-related issues.METHODS This was a retrospective analysis of a cohort of cirrhotic patients aged≥18 years,who underwent LT at a high-complexity teaching hospital from January 2005 to December 2012.RESULTS Overall,429 patients underwent LT during the study period.Of these,213(49.7%)underwent the tuberculin skin test(TST)during the pre-transplant period,and 35(16.4%)of them had a positive result.The treatment for LTBI was initiated after LT in 12(34.3%)of the TST-positive patients;in 3(25.0%),treatment was maintained for at least 6 mo.CONCLUSION The prevalence of LTBI was lower than expected.Initiation and completion of LTBI treatment was limited by difficulties in the management of these special patients.Isabela Dias Lauar Luciana Costa Faria Roberta Maia de Castro Romanelli Wanessa Trindade Clemente 2021World Journal of Transplantation2021,11,12:1

20Metabolomics in cancer and cancer-associated inflammatory cells显示文摘Metabolomics is the last frontier of modern molecular biology,and the state-of-the-art technique for studying metabolism.Mass spectrometry and nuclear magnetic resonance spectroscopy are the main analytical approaches in metabolomics.Cellular metabolism plays a pivotal role during cell resting and activation.Immune cells exhibit remarkable metabolic plasticity,fundamental to support their adaptation to inflammatory environments and functional requirements.Cancer is a metabolic and inflammatory disease.A metabolic shift is crucial for oncogenesis,tumor cell survival,invasion,metastasis,and the associated inflammatory process.The tumor microenvironment is mainly orchestrated by immune-inflammatory cells and essential for the neoplastic process.Inflammatory cells from tumor stroma adapt to different metabolic pathways during tumor progression,and this metabolic reprogramming affects macrophages,neutrophils,T cells,and others.Targeting the metabolism of tumor and immune cells may lead to important therapeutic implications in cancer.Thus,understanding the metabolic changes that drive the interactions between tumor and stromal cells is a promising avenue advances in cancer diagnostics and therapies,leading to more accurate guidance.In this review,we discuss the most recent metabolomics approaches in cancer studies on the tumor-associated inflammatory microenvironment.Gilson Costa dos Santos Roberta Saldanha-Gama Natália Mesquita de Brito Mariana Renovato-Martins Christina Barja-Fidalgo 2021Journal of Cancer Metastasis and Treatment2021,7,1:0
