3Dihydroartemisinin induces ER stress-dependent apoptosis of Echinococcus protoscoleces in vitro显示文摘In this study,we investigated the effect of dihydroartemisinin on Echinococcus protoscoleces and explored the role of endoplasmic reticulum stress in this process.Echinococcus protoscoleces were collected and cultured in RPMI 1640 medium.Changes in the expressions of glucose-regulated protein 78(GRP-78),caspase-12,and C/EBP homologous protein(CHOP)were assessed through confocal immunofluorescence and western blot analysis.Cell viability and morphological changes were observed under a light microscope.The ultrastructure of protoscoleces was observed by scanning electron microscopy and transmission electron microscopy.Caspase-3 activity was detected using an enzyme assay kit.After dihydroartemisinin treatment,the protoscoleces showed loss of viability,and morphological changes including soma contraction,blebs formation,hooks loss,microtrichia destruction,and development of lipid droplets was observed.The levels of caspase-12 and CHOP were increased within 2 days of dihydroartemisinin treatment.However,the levels of GRP-78,caspase-12,and CHOP were decreased in 4 days.Furthermore,caspase-3 activity was increased after treatment with different concentrations of dihydroartemisinin.Dihydroartemisinin can induce apoptosis in protoscoleces via the ER stress-caspase-3 apoptotic pathway in vitro.These results indicate that dihydroartemisinin is a potentially valuable therapeutic agent against echinococcosis.Rongji Ma Wenjuan Qin Yuanmao Xie Ziwei Han Shuojie Li Yufeng Jiang Hailong Lv 2020Acta Biochimica et Biophysica Sinica2020,52,10:1

5Two-photon live imaging of direct glia-to-neuron conversion in the mouse cortex显示文摘Over the past decade,a growing number of studies have reported transcription factor-based in situ reprogramming that can directly conve rt endogenous glial cells into functional neurons as an alternative approach for n euro regeneration in the adult mammalian central ne rvous system.Howeve r,many questions remain regarding how a terminally differentiated glial cell can transform into a delicate neuron that forms part of the intricate brain circuitry.In addition,concerns have recently been raised around the absence of astrocyte-to-neuron conversion in astrocytic lineage-tra cing mice.In this study,we employed repetitive two-photon imaging to continuously capture the in situ astrocyte-to-neuron conversion process following ecto pic expression of the neural transcription factor NeuroD1 in both prolife rating reactive astrocytes and lineage-tra ced astrocytes in the mouse cortex.Time-lapse imaging over several wee ks revealed the ste p-by-step transition from a typical astrocyte with numero us short,tapered branches to a typical neuro n with a few long neurites and dynamic growth cones that actively explored the local environment.In addition,these lineage-converting cells were able to migrate ra dially or to ngentially to relocate to suitable positions.Furthermore,two-photon Ca2+imaging and patch-clamp recordings confirmed that the newly generated neuro ns exhibited synchronous calcium signals,repetitive action potentials,and spontaneous synaptic responses,suggesting that they had made functional synaptic connections within local neural circuits.In conclusion,we directly visualized the step-by-step lineage conversion process from astrocytes to functional neurons in vivo and unambiguously demonstrated that adult mammalian brains are highly plastic with respect to their potential for neuro regeneration and neural circuit reconstruction.Zongqin Xiang Shu He Rongjie Chen Shanggong Liu Minhui Liu Liang Xu Jiajun Zheng Zhouquan Jiang Long Ma Ying Sun Yongpeng Qin Yi Chen Wen Li Xiangyu Wang Gong Chen Wenliang Lei 2024Neural Regeneration Research2024,19,8:0

7An assessment Protocal of Ammonia Removal Efficacy for Air Purification Products显示文摘A device is designed to assess the ammonia removal effect of passive air purification products which derived the QB/T 2761-2006 Methods for determination of purificatory effect of indoor environment decontamination product.The device is composed of three parts:ammonia generator,ammonia removal device and ammonia collection device,which is made up of common laboratory equipment.The reagents used are common acid and alkali reagents in the laboratory.During the experiment,5.0mL of 25%concentrated ammonia water is added to the ammonia generator as the ammonia source,and filter paper is used as the sample carrier.Take 3.0g sample and add it to the filter paper,after the ammonia is absorbed by the filter paper with the sample,the residual ammonia is collected and measured by acid-base titration.The ammonia removal efficacy of the sample is calculated by the ammonia absorption amount of the blank sample and the sample to be tested.This method has the characteristics of simple equipment,small area,toxic or harmful substance free,and fabulous accuracy.Four different samples were tested,and the results matched the standard method.Li Rongjie Ma Jiaoying Zhang Tianyi 2023China Detergent & Cosmetics2023,8,3:0
8Homogenization Function of Microchannel on Heat Absorber with Compound Parabolic Concentrator显示文摘Microchannels offer unique advantages on heat transfer performance. In this paper, microchannels are applied to the compound parabolic concentrator(CPC) system. A multi-physical field coupling model based on Finite Element Method is proposed to investigate the homogenization effect of the microchannel heat absorber on the CPC non-uniform concentration. The energy conversion process from optics to heat is simulated using TracePro software, and the heat transfer processes in the microchannel are computed by Fluent using user defined functions(UDF). It is found that the microchannels behave well on weakening the influence of the nonuniformity solar heat flux on the performance of the CPC. The temperature nonuniformity of the outlet section is less than 10^(-3) in the direction of fluid flow caused by the microchannel, although the maximum surface heat flux inhomogeneity of the microchannel reaches 2.3. The peak value of the heat flux on the surface of the absorber changes from double peak to single peak, and moves to the edge, resulting in more uneven heat flux distribution with the increase of the incident angle within the acceptance semi-angle of the CPC. The result of TracePro clearly shows that when the concentration ratio is less than 5, the heat flux nonuniformity on the surface of the absorber decreases with the increase in concentration ratio. It was interestingly found that the temperature distribution of the heat transfer fluid has weak sensitivity to the changes of truncation ratio.
