| 6 | Youjing granules ameliorate spermatogenesis in rats through regulating the prolifereation of spermatogonial stem cells显示文摘Male infertility has evolved from a common reproductive system disease to a major social issue.Youjing granule(YG)is a Chinese medicinal material used as a therapy method for tonifying the kidneys and removing dampness due to its pathogenic characteristics.YG has been shown to regulate sperm quality in clinical trials,but the underlying mechanism is not fully understood.The present study was aimed to explore the protective effects and mechanism of action of YG on male reproductive system damage caused by methyl methane sulfonate(MMS).We first established an infertility model of rats through oral administration of MMS and then treated with YG.To determine the effect of YG,spermatogenesis,microvascular density,and secretory function of Leydig cells and Sertoli cells in rats were assessed.Spermatogonial stem cells(SSCs)were co-cultured with mouse embryo fibroblast(MEF)cells as an in vitro cell model before exposure to serum containing YG.Furthermore,the proliferation and apoptosis of SSCs were measured.Results indicated that YG increased the expression of self-renewal and proliferation-related molecules such as glial cell line derived neurotrophic factor(GDNF)and fibroblast growth factor-2(FGF2),and improved the quality of sperm and the proliferation of SSCs.In conclusion,YG may protect spermatogenetic function of rats through regulating the proliferation and self-renewal of SSCs. | JIANG Xu-Ping SUN Yao-Xiang QIAO Bin ZHU Wen-jiao CHU Yu-Jiao SUN Miao-Miao ANG Ruije MA Tie-Liang ANG Zhi-An | 2022 | Chinese Journal of Natural Medicines2022,20,8: | 1 |

| 11 | Click CAR-T cell engineering for robustly boosting cell immunotherapy in blood and subcutaneous xenograft tumor显示文摘The adoptive transfer of chimeric antigen receptor-T(CAR-T)cells has shown remarkable clinical responses in hematologic malignancies.However,unsatisfactory curative results and side effects for tumor treatment are still unsolved problems.Herein we develop a click CAR-T cell engineering strategy via cell glycometabolic labeling for robustly boosting their antitumor effects and safety in vivo.Briefly,paired chemical groups(N3/BCN)are separately incorporated into CAR-T cell and tumor via nondestructive intrinsic glycometabolism of exogenous Ac4GalNAz and Ac4ManNBCN,serving as an artificial ligand-receptor.Functional groups anchored on cell surface strengthen the interaction of CAR-T cell and tumor via bioorthogonal click chemistry,further enhancing specific recognition,migration and selective antitumor effects of CAR-T cells.In vivo,click CAR-T cell completely removes lymphoma cells and minimizes off-target toxicity via selective and efficient bioorthogonal targeting in blood cancer.Surprisingly,compared to unlabeled cells,artificial bioorthogonal targeting significantly promotes the accumulation,deep penetration and homing of CAR-T cells into tumor tissues,ultimately improving its curative effect for solid tumor.Click CAR-T cell engineering robustly boosts selective recognition and antitumor capabilities of CAR T cells in vitro and in vivo,thereby holding a great potential for effective clinical cell immunotherapy with avoiding adverse events in patients. | Hong Pan Wenjun Li Ze Chen Yingmei Luo Wei He Mengmeng Wang Xiaofan Tang Huamei He Lanlan Liu Mingbin Zheng Xin Jiang Ting Yin Ruijing Liang Yifan Ma Lintao Cai | 2021 | Bioactive Materials2021,6,4: | 0 |
| 12 | Stability and band gap engineering of silica-confined lead halide perovskite nanocrystals under high pressure显示文摘SiO_(2)is the major mineral substance in the upper mantle of the earth.Therefore,studies of the silica-coated materials under high-pressure are essential to explore the physical and chemical properties of the upper mantle.The silica-confined CsPbBr_(3)nanocrystals(NCs)have recently attracted much attention because of the improved photoluminescence(PL)quantum yield,owing to the protection of silica shell.However,it remains considerable interest to further explore the relationship between optical properties and the structure of CsPbBr_(3)@SiO_(2)NCs.We systemically studied the structural and optical properties of the CsPbBr_(3)@SiO_(2)NCs under high pressure by using diamond anvil cell(DAC).The discontinuous changes of PL and absorption spectra occurred at~1.40 GPa.Synchrotron X-ray diffraction(XRD)studies of CsPbBr_(3)@SiO_(2)NCs under high pressure indicated an isostructural phase transformation at about 1.36 GPa,owing to the pressure-induced tilting of the Pb-Br octahedra.The isothermal bulk moduli for two phases are estimated about 60.0 GPa and 19.2 GPa by fitting the equation of state.Besides,the transition pressure point of CsPbBr_(3)@SiO_(2)NCs is slightly higher than that of pristine CsPbBr_(3)NCs,which attributed to the buffer effect of coating silica shell.The results indicate that silica shell is able to enhance the stabilization without changing the relationship between optical properties and structure of CsPbBr_(3)NCs.Our results were fascinated to model the rock metasomatism in the upper mantle and provided a new‘lithoprobe’for detecting the upper mantle. | Ruijing Fu Yaping Chen Lingrui Wang Zhiwei Ma Pengfei Lv Ying Song Songrui Yang Guanjun Xiao Bo Zou | 2021 | Geoscience Frontiers2021,12,2: | 0 |
