2Elevated Kir2.1/nuclear N2ICD defines a highly malignant subtype of non-WNT/SHH medulloblastomas显示文摘Medulloblastoma(MB)is one of the most common childhood malignant brain tumors(WHO grade IV),traditionally divided into WNT,SHH,Group 3,and Group 4 subgroups based on the transcription profiles,somatic DNA alterations,and clinical outcomes.Unlike WNT and SHH subgroup MBs,Group 3 and Group 4 MBs have similar transcriptomes and lack clearly specific drivers and targeted therapeutic options.The recently revised WHO Classification of CNS Tumors has assigned Group 3 and 4 to a provisional non-WNT/SHH entity.In the present study,we demonstrate that Kir2.1,an inwardly-rectifying potassium channel,is highly expressed in non-WNT/SHH MBs,which promotes tumor cell invasion and metastasis by recruiting Adam10 to enhance S2 cleavage of Notch2 thereby activating the Notch2 signaling pathway.Disruption of the Notch2 pathway markedly inhibited the growth and metastasis of Kir2.1-overexpressing MB cell-derived xenograft tumors in mice.Moreover,Kir2.1^(high)/nuclear N2ICD^(high)MBs are associated with the significantly shorter lifespan of the patients.Thus,Kir2.1^(high)/nuclear N2ICD^(high)can be used as a biomarker to define a novel subtype of non-WNT/SHH MBs.Our findings are important for the modification of treatment regimens and the development of novel-targeted therapies for non-WNT/SHH MBs.Yan-Xia Wang Haibo Wu Yong Ren Shengqing Lv Chengdong Ji Dongfang Xiang Mengsi Zhang Huimin Lu Wenjuan Fu Qing Liu Zexuan Yan Qinghua Ma Jingya Miao Ruili Cai Xi Lan Bin Wu Wenying Wang Yinhua Liu Dai-Zhong Wang Mianfu Cao Zhicheng He Yu Shi Yifang Ping Xiaohong Yao Xia Zhang Peng Zhang Ji Ming Wang Yan Wang Youhong Cui Xiu-Wu Bian 2022Signal Transduction and Targeted Therapy2022,7,4:0
3NDRG1 promotes endothelial dysfunction and hypoxia-induced pulmonary hypertension by targeting TAF15显示文摘Background:Pulmonary hypertension(PH)represents a threatening pathophysiologic state that can be induced by chronic hypoxia and is characterized by extensive vascular remodeling.However,the mechanism underlying hypoxia-induced vascular remodeling is not fully elucidated.Methods and Results:By using quantitative polymerase chain reactions,western blotting,and immunohistochemistry,we demon-strate that the expression of N-myc downstream regulated gene-1(NDRG1)is markedly increased in hypoxia-stimulated endothelial cells in a time-dependent manner as well as in human and rat endothelium lesions.To determine the role of NDRG1 in endothelial dysfunction,we performed loss-of-function studies using NDRG1 short hairpin RNAs and NDRG1 over-expression plasmids.In vitro,silencing NDRG1 attenuated proliferation,migration,and tube formation of human pulmonary artery endothelial cells(HPAECs)un-der hypoxia,while NDRG1 over-expression promoted these behaviors of HPAECs.Mechanistically,NDRG1 can directly interact with TATA-box binding protein associated factor 15(TAF15)and promote its nuclear localization.Knockdown of TAF15 abrogated the effect of NDRG1 on the proliferation,migration and tube formation capacity of HPAECs.Bioinformatics studies found that TAF15 was involved in regulating PI3K-Akt,p53,and hypoxia-inducible factor 1(HIF-1)signaling pathways,which have been proved to be PH-related pathways.In addition,vascular remodeling and right ventricular hypertrophy induced by hypoxia were markedly alleviated in NDRG1 knock-down rats compared with their wild-type littermates.Conclusions:Taken together,our results indicate that hypoxia-induced upregulation of NDRG1 contributes to endothelial dysfunction through targeting TAF15,which ultimately contributes to the development of hypoxia-induced PH.Chengwei Li Junzhu Lv Gulinuer Wumaier Yu Zhao Liang Dong Yuzhen Zeng Ning Zhu Xiujuan Zhang Jing Wang Jingwen Xia Shengqing Li 2023Precision Clinical Medicine2023,6,4:0
4Ecological health analysis of wetlands in the middle reaches of Yangtze River under changing environment显示文摘Changes in wetland ecosystems have a critical impact on the local ecology and species diversity.Different development scenarios and policies are key factors influencing their changes.Therefore,we studied changes of wetlands in the middle Yangtze River basin(MYRB)in 2001-2020,and a patch-generated land use simulation(PLUS)model and random forest(RF)method were applied to predict and analyze the changes under different scenarios in the MYRB in the future(i.e.2035-2095).The results indicated that:(1)The regions with high wetland proportions were concentrated in the central and eastern MYRB in 2001-2020,with a 1.5%decrease in overall wetland area;(2)The RF could simulate the future ecological quality with training and testing accuracies of 0.98 and 0.92,respectively;(3)Remote Sensing Ecological Index(RSEI)less than 0.5 in the central and eastern regions and 13.3%reduction in the northwest in the SSP245 scenario.In general,the study provides a basis for future regional studies of ecosystem quality and provides data to support wetland conservation and management.Shengqing Zhang Xiaoyan Zhai Peng Yang Jun Xia Sheng Hu Libo Zhou Cai Fu 2023International Journal of Digital Earth2023,16,1:0
