1Interferon-induced Transmembrane Protein 3 Prevents Acute Influenza Pathogenesis in Mice显示文摘Objective Interferon-induced transmembrane protein 3(IFITM3)is an important member of the IFITM family.However,the molecular mechanisms underlying its antiviral action have not been completely elucidated.Recent studies on IFITM3,particularly those focused on innate antiviral defense mechanisms,have shown that IFITM3 affects the body’s adaptive immune response.The aim of this study was to determine the contribution of IFITM3 proteins to immune control of influenza infection in vivo.Methods We performed proteomics,flow cytometry,and immunohistochemistry analysis and used bioinformatics tools to systematically compare and analyze the differences in natural killer(NK)cell numbers,their activation,and their immune function in the lungs of Ifitm3-/-and wild-type mice.Results Ifitm3-/-mice developed more severe inflammation and apoptotic responses compared to wild-type mice.Moreover,the NK cell activation was higher in the lungs of Ifitm3-/-mice during acute influenza infection.Conclusions Based on our results,we speculate that the NK cells are more readily activated in the absence of IFITM3,increasing mortality in Ifitm3-/-mice.SUN Qiang LEI Na LU Jian GAO Rong Bao LI Zi LIU Li Qi SUN Ying GUO Jun Feng WANG Da Yan SHU Yue Long 2020Biomedical and Environmental Sciences2020,33,5:4
2COVID-19 epidemic prediction and the impact of public health interventions: A review of COVID-19 epidemic models显示文摘The coronavirus disease outbreak of 2019(COVID-19)has been spreading rapidly to all corners of the word,in a very complex manner.A key research focus is in predicting the development trend of COVID-19 scientifically through mathematical modelling.We conducted a systematic review of epidemic prediction models of COVID-19 and the public health intervention strategies by searching the Web of Science database.55 studies of the COVID-19 epidemic model were reviewed systematically.It was found that the COVID-19 epidemic models were different in the model type,acquisition method,hypothesis and distribution of key input parameters.Most studies used the gamma distribution to describe the key time period of COVID-19 infection,and some studies used the lognormal distribution,the Erlang distribution,and theWeibull distribution.The setting ranges of the incubation period,serial interval,infectious period and generation time were 4.9-7 days,4.41-8.4 days,2.3-10 days and 4.4-7.5 days,respectively,and more than half of the incubation periods were set to 5.1 or 5.2 days.Most models assumed that the latent period was consistent with the incubation period.Some models assumed that asymptomatic infections were infectious or pre-symptomatic transmission was possible,which overestimated the value of R0.For the prediction differences under different public health strategies,the most significant effect was in travel restrictions.There were different studies on the impact of contact tracking and social isolation,but it was considered that improving the quarantine rate and reporting rate,and the use of protective face mask were essential for epidemic prevention and control.The input epidemiological parameters of the prediction models had significant differences in the prediction of the severity of the epidemic spread.Therefore,prevention and control institutions should be cautious when formulating public health strategies by based on the prediction results of mathematical models.Yue Xiang Yonghong Jia Linlin Chen Lei Guo Bizhen Shu Enshen Long 2021Infectious Disease Modelling2021,6,1:3
3Experimental study on the pyrolysis of Shenhua coal to produce hydrogen显示文摘Based on thermogravimetric analysis, kinetic parameters of Shenhua coal, such as activation energy and frequency factor have been got from the TG / DTG results. It showed that activation energy is the smallest at the temperature from 850℃ to 930℃ with heating rate of 15℃·min-1 . Pyrolysis was further carried out on a fixed-bed reactor with different heating rate. The production of hydrogen has been studied in particular. It gave the highest hydrogen production at the heating rate of 15℃·min-1 . And it also found the increase of hydrogen production with the raise of temperature. Furthermore, catalytic pyrolysis of coal was done at adding 2% of transition-metal oxides to discover the impact of catalysts upon coal pyrolysis at heating rate of 15℃·min-1 . It indicated that both N-type and P-type oxide enhanced hydrogen production in coal pyrolysis. 4A Novel Reassortant H2N3 Influenza Virus Isolated from China显示文摘Objective To analyze the genetic composition of a novel H2N3 virus isolate identified from a duck cage swab in a live poultry market (LPM) in 2009 in Guangdong province of China.Methods PCR-positive specimens were inoculated into embryonated chicken eggs and subtyped by conventional RT-PCR. All segments of the virus A/environment/Guangdong/2/2009 were sequenced,and phylogenetic trees were constructed and analyzed.Results The genes of this virus belong to Eurasian-lineage avian viruses. The virus is a reassortant withthe HA gene from an H2N2 virus and the NA gene from an H5N3 virus. The PB1, PB2, and NP genes were from an H4N6 virus, the PA was from an H3N8 virus, the M gene was from an H1N3 virus, and the NS gene was from an H10N6 virus.Conclusion A novel avian-origin reassortant H2N3 influenza virus was detected in a live poultry market. 5Synthesis of Hydroxypropyl-β-cyclodextrin Bonded Silica-gel and its Application to Resolution显示文摘In order to set up a simple and effective method for resolution of optical isomers, hydroxypropyl-β-cyclodextrin was bonded to silica-gel, which can be used for preparation of thin-layer chromatography plates. Resolution of clenbuterol and propranolol were investigated on these thin-layer chromatography plates using different combinations of solvent systems at ambient temperature. The best simultaneous resolution was achieved in solvent system of acetonitrile- n-butanol (50:50, v/v). Rst values of resolution of clenbuterol hydrochloride and propranolol hydrochloride are 3.6 and 4.3, respectively. The spots of different enantiomers are separated clearly. The results showed that hydroxypropyl-β-cyclodextrin bonded silica-gel could be successful in resolution of chiral adrenergic drugs. 6Phylogenetic and Molecular Analysis of an H7N7 Avian Influenza Virus Isolated in East Dongting Lake in 2012显示文摘Objective In March 2012, an H7N7 subtype avian influenza virus(AIV) named A/wild goose/Dongting/PC0360/2012(H7N7)(DT/PC0360) was recovered from a wild goose in East Dongting Lake. We performed whole-genome sequencing of the isolate, and analyzed the phylogenetic and molecular characterization. Methods RNA was extracted from environment samples(including fecal samples from wild bird or domestic ducks, and water samples) for detecting the presence of Influenza A Virus targeting Matrix gene, using realtime RT-PCR assay. The positive samples were performed virus isolation with embryonated eggs. The subtype of the isolates were identified by RT-PCR assay with the H1-H16 and N1-N9 primer set. The whole-genome sequencing of isolates were performed. Phylogenetic and molecular characterizations of the eight genes of the isolates were analyzed. Results Our results suggested that all the eight gene segments of DT/PC0360 belonged to the Eurasian gene pool, and the HA gene were belonged to distinct sublineage with H7N9 AIV which caused outbreaks in China's Mainland in 2013. The hemagglutinin cleavage site of HA of DT/PC0360 showed characterization of low pathogenic avian influenza virus. 7A Novel Reassortant H3N8 Influenza Virus Isolated from Drinking Water for Duck in a Domestic Duck Farm in Poyang Lake Area显示文摘Objective To conduct a full genome sequence analysis for genetic characterization of an H3N8 influenza virus isolated from drinking water of a domestic duck farm in Poyang Lake area in 2011. Methods The virus was cultivated by specific pathogen free (SPF) chicken embryo eggs and was subtyped into hemagglutinin (HA) and neuraminidase (NA) by real-time PCR method. Eight gene segments were sequenced and phylogenetic analysis was conducted. Results The NA gene of this virus belongs to North American lineage; other seven genes belong to Eurasian lineage. Compared with the viruses containing NA gene, the PB2 and PB1 gene came from different clades. And this indicates that the virus was a novel reassortant genotype. The HA receptor binding preference was avian-like and the cleavage site sequence showed a low pathogenic feature. There was no drug resistancemutation of M2 protein. Themutations of Asn30Asp, and Thr215Ala of the M1 protein implied the potential of pathogenicity increase in mice. 8H5N1 Avian Influenza Pre-pandemic Vaccine Strains in China显示文摘Objective To prepare the 4 candidate vaccine strains of H5N1 avian influenza virus isolated in China. Methods Recombinant viruses were rescued using reverse genetics. Neuraminidase(NA) and hemagglutinin(HA) segments of the A/Xinjiang/1/2006, A/Guangxi/1/2009, A/Hubei/1/2010, and A/Guangdong/1/2011 viruses were amplified by RT-PCR. Multibasic amino acid cleavage site of HA was removed and ligated into the pCIpolI vector for virus rescue. The recombinant viruses were evaluated by trypsin dependent assays. Their embryonate survival and antigenicity were compared with those of the respective wild-type viruses. Results The 4 recombinant viruses showed similar antigenicity compared with wild-type viruses, chicken embryo survival and trypsin-dependent characteristics. 9Identification of Dual Receptor-binding Specific Strains of Human H5N1 Viruses in China显示文摘Objective Both the 2,6 linkage and its topology on target cells are critical for the recognition by human influenza virus.The binding preference of avian flu virus H5N1 HA to the 2,3-linked sialylated glycans is considered the major factor limiting its efficient infection and transmission in humans.To monitor potential adaptation of H5N1 virus in human population,the surveillance of receptor-binding specificity was undertaken in China.Methods The binding specificity of 32 human H5N1 virus strains isolated from 2003 to 2009 was tested by 2,3-specific sialidase-treated chicken red blood cell (CRBC) agglutination assay and a solid-phase direct binding assay with synthetic sialylglycopolymers.Results Dual binding preference to 2,3 and 2,6-glycans were found in two strains:A/Guangdong/1/06 (A/GD/1/06) and A/Guangxi/1/08 (A/GX/1/08).Thoughminor effect of short-2,6-binding was detected in A/GX/1/08 at a low virus titer,both showed high affinity to the oligosaccharide at a high load.Notably both are of the long-2,6-recognition,with the same topology as that of human H1N1 and H3N2 viruses.Conclusion The findings suggest that human H5N1 virus in China likely acquired the potential human-adaptation ability.Further research and surveillance on receptor-binding specificity of H5N1 viruses are required.ZHOU Jian Fang ZOU Shu Mei LI Zi WANG Min DONG Jie GUO Jun Feng WEI He Jiang WEN Le Ying XU Hong SHU Yue Long 2012Biomedical and Environmental Sciences2012,25,1:0
10Evaluation of A Single-reaction Method for Whole Genome Sequencing of Influenza A Virus using Next Generation Sequencing显示文摘Objective To evaluate a single-reaction genome amplification method, the multisegment reverse transcription-PCR(M-RTPCR), for its sensitivity to full genome sequencing of influenza A virus, and the ability to differentiate mix-subtype virus, using the next generation sequencing(NGS) platform. Methods Virus genome copy was quantified and serially diluted to different titers, followed by amplification with the M-RTPCR method and sequencing on the NGS platform. Furthermore, we manually mixed two subtype viruses to different titer rate and amplified the mixed virus with the M-RTPCR protocol, followed by whole genome sequencing on the NGS platform. We also used clinical samples to test the method performance. Results The M-RTPCR method obtained complete genome of testing virus at 125 copies/reaction and determined the virus subtype at titer of 25 copies/reaction. Moreover, the two subtypes in the mixed virus could be discriminated, even though these two virus copies differed by 200-fold using this amplification protocol. The sensitivity of this protocol we detected using virus RNA was also confirmed with clinical samples containing low-titer virus.
