2Construction of an electronic physical map of Magnaporthe oryzae using genomic position-ready SSR markers显示文摘Magnaporthe oryzae is a model for plant pathogenic filamentous fungi. We have assembled a simple sequence repeat (SSR)-based physical map of the species, using in silico sequence data. A set of 120 SSR markers was developed from the genomic sequence of the reference isolate 70-15. These markers were readily amplified from the genomic DNA of other isolates, and high levels of allelic variation characterised the parental isolates of the two crosses tested. All the markers were locatable to one of the seven M. oryzae chromosomes. An SSR-based physical in silico map was constructed, and pre-existing SSR and RFLP loci were integrated into the map, along with 23 Avr (avirulence) genes and two other genes of importance to the plant/pathogen interaction. 3Genetic and physical mapping of AvrPi7, a novel avirulence gene of Magnaporthe oryzae using physical position-ready markers显示文摘Rice blast, caused by the fungal pathogen Magnaporthe oryzae, is one of the most devastating crop diseases worldwide. The avirulence gene corresponding to rice blast resistance gene Pi7 in field isolate CHL346 was inherited as a single gene, designated AvrPi7, in a segregating population consisting of 189 ascospore progenies derived from a cross between field isolates CHL346 and CHL42. In order to determine the chromosomal location of the AvrPi7 locus, a total of 121 simple sequence repeat (SSR) markers were developed based on the whole-genome sequence of reference isolate 70-15 of M. oryzae. Linkage analysis of the locus with these SSR markers showed that eight SSR markers on chromosome 1 were linked to the locus, among which the closest flanking markers MS1-9 and MS1-15 were 3.2 and 16.4 cM from the locus, respectively. For fine mapping, additional PCR-based makers including eight SSR markers and three candidate avirulence gene (CAG) markers were developed in the region flanking both markers. The AvrPi7 locus was genetically delimited within a 1.6-cM region flanked by markers MS1-21 and MS1-22, and co-segregated with the marker CAG2. To construct a physical map of the AvrPi7 locus, molecular markers linked to the Avr gene were mapped on the supercontigs of the ref-erence isolate 70-15 through bioinformation analysis (BIA). Consequently, the AvrPi7 locus was delim-ited to a 75-kb interval flanked by markers MS1-21 and MS1-22 based on the reference sequence. 6Development of a Rapid Test Bar for Quantitative Detection of Chloride Ions in Fresh Concrete显示文摘Due to the shortage of natural raw materials,many practitioners proposed to make concrete from desalinated sea sand.To reduce the risk of abuse of uncleaned sea sand,we proposed a new rapid test bar to detect chloride ions in fresh concrete quantitatively.The test bars were prepared by mixing silver chromate with cotton fiber.By virtue of the high liquid-absorbent capacity of cotton fiber,the chloride-contained liquid in fresh concrete will rapidly enter into the test bar and react with the silver chromate to finally show a certain coloring height,which corresponds to the actual chloride content.In order to optimize the preparation process,the color rendering,testing time and stability of test rods under different mixing ratios and conditions were investigated.The relationship between color height and chloride content was established,and an auxiliary card is prepared for user-friendly calculation of chloride content.The results showed that the best performance of the test bar was obtained under the following conditions:mass concentration of silver chromate 5.0g/L,soaking time 1 min,drying temperature 50℃and content of discolored silica gel 0.5g/root.Yingjie ZHANG Shujie WEI Wen PAN Xuping JI Zijian SONG 2021Research and Application of Materials Science2021,3,1:0
7Quantum dot mode-locked frequency comb with ultra-stable 25.5 GHz spacing between 20℃ and 120℃显示文摘Semiconductor mode-locked lasers(MLLs)are promising frequency comb sources for dense wavelength-divisionmultiplexing(DWDM)data communications.Practical data communication requires a frequency-stable comb source in a temperature-varying environment and a minimum tone spacing of 25 GHz to support high-speed DWDM transmissions.To the best of our knowledge,however,to date,there have been no demonstrations of comb sources that simultaneously offer a high repetition rate and stable mode spacing over an ultrawide temperature range.Here,we report a frequency comb source based on a quantum dot(QD)MLL that generates a frequency comb with stable mode spacing over an ultrabroad temperature range of 20–120℃.The two-section passively mode-locked In As QD MLL comb source produces an ultra-stable fundamental repetition rate of 25.5 GHz(corresponding to a 25.5 GHz spacing between adjacent tones in the frequency domain)with a variation of 0.07 GHz in the tone spacing over the tested temperature range.By keeping the saturable absorber reversely biased at-2 V,stable mode-locking over the whole temperature range can be achieved by tuning the current of the gain section only,providing easy control of the device.At an elevated temperature of 100℃,the device shows a 6 d B comb bandwidth of 4.81 nm and 31 tones with>36 d B optical signal-to-noise ratio.The corresponding relative intensity noise,averaged between 0.5 GHz and 10 GHz,is-146 d Bc∕Hz.Our results show the viability of the In As QD MLLs as ultra-stable,uncooled frequency comb sources for low-cost,large-bandwidth,and low-energy-consumption optical data communications.SHUJIE PAN JIANOU HUANG ZICHUAN ZHOU ZHIXIN LIU LALITHA PONNAMPALAMZIZHUO LIU MINGCHU TANG MU-CHIEH LO ZIZHENG CAO KENICHI NISHI KEIZO TAKEMASA MITSURU SUGAWARA RICHARD PENTY IAN WHITE ALWYN SEEDS HUIYUN LIU SIMING CHEN 2020Photonics Research2020,8,12:0
8Highly efficient 1D p-Te/2D n-Bi_(2)Te_(3) heterojunction self-driven broadband photodetector显示文摘Broadband photodetectors with self-driven functions have attracted intensive scientific interest due to their low energy consumption and high optical gain.However,high-performance broadband self-driven photodetectors are still a significant challenge due to the complex fabrication processes,environmental toxicity,high production costs of traditional 3D semiconductor materials and sharply raised contact resistance,severe interfacial recombination of 2D materials and 2D/3D mixed dimension heterojunction.Here,1D p-Te/2D n-Bi_(2)Te_(3) heterojunctions are constructed by the simple and low-cost hydrothermal method.1D p-Te/2D n-Bi_(2)Te_(3) devices are applied in photoelectrochemical(PEC)photodetectors,with their high performance attributed to the good interfacial contacts reducing interface recombination.The device demonstrated a broad wavelength range(365–850 nm)with an Iph/Idark as high as 377.45.The R_(i),D^(*),and external quantum efficiency(EQE)values of the device were as high as 12.07 mA/W,5.87×10^(10) Jones,and 41.05%,respectively,which were significantly better than the performance of the prepared Bi_(2)Te_(3) and Te devices.A comparison of the freshly fabricated device and the device after 30 days showed that 1D p-Te/2D n-Bi_(2)Te_(3) had excellent stability with only 18.08%decay of photocurrent.It is anticipated that this work will provide new emerging material for future design and preparation of a high-performance self-driven broadband photodetector.Chenchen Zhao Dongbo Wang Jiamu Cao Zhi Zeng Bingke Zhang Jingwen Pan Donghao Liu Sihang Liu Shujie Jiao Tianyuan Chen Gang Liu Xuan Fang Liancheng Zhao Jinzhong Wang 2024Nano Research2024,17,3:0
9A chromosome-level genome assembly for Erianthus fulvus provides insights into its biofuel potential and facilitates breeding for improvement of sugarcane显示文摘Erianthus produces substantial biomass,exhibits a good Brix value,and shows wide environmental adaptability,making it a potential biofuel plant.In contrast to closely related sorghum and sugarcane,Erianthus can grow in degraded soils,thus releasing pressure on agricultural lands used for biofuel production.However,the lack of genomic resources for Erianthus hinders its genetic improvement,thus limiting its potential for biofuel production.In the present study,we generated a chromosome-scale reference genome for Erianthus fulvus Nees.The genome size estimated by flow cytometry was 937 Mb,and the assembled genome size was 902 Mb,covering 96.26%of the estimated genome size.A total of 35065 proteincoding genes were predicted,and 67.89%of the genome was found to be repetitive.A recent wholegenome duplication occurred approximately 74.10 million years ago in the E.fulvus genome.Phylogenetic analysis showed that E.fulvus is evolutionarily closer to S.spontaneum and diverged after S.bicolor.Three of the 10 chromosomes of E.fulvus formed through rearrangements of ancestral chromosomes.Phylogenetic reconstruction of the Saccharum complex revealed a polyphyletic origin of the complex and a sister relationship of E.fulvus with Saccharum sp.,excluding S.arundinaceum.On the basis of the four amino acid residues that provide substrate specificity,the E.fulvus SWEET proteins were classified as monoand disaccharide sugar transporters.Ortho-QTL genes identified for 10 biofuel-related traits may aid in the rapid screening of E.fulvus populations to enhance breeding programs for improved biofuel production.The results of this study provide valuable insights for breeding programs aimed at improving biofuel production in E.fulvus and enhancing sugarcane introgression programs.Ling Kui Aasim Majeed Xianhong Wang Zijiang Yang Jian Chen Lilian He Yining Di Xuzhen Li Zhenfeng Qian Yinming Jiao Guoyun Wang Lufeng Liu Rong Xu Shujie Gu Qinghui Yang Shuying Chen Hongbo Lou Yu Meng Linyan Xie Fu Xu Qingqing Shen Amit Singh Karl Gruber Yunbing Pan Tingting Hao Yang Dong Fusheng Li 2023Plant Communications2023,4,4:0
