| 1 | Identification and characterization of a novel bipartite nuclear localization signal in the hepatitis B virus polymerase显示文摘AIM:To characterize the nuclear import of hepatitis B virus(HBV)polymerase(P)and its relevance for the viral life cycle.METHODS:Sequence analysis was performed to predict functional motives within P.Phosphorylation of P was analyzed by in vitro phosphorylation.Phosphorylation site and nuclear localization signal(NLS)were destroyed by site directed mutagenesis.Functionality of the identified NLS was analyzed by confocal fluorescence microscopy and characterizing the karyopherin binding.Relevance of the structural motives for viral life cycle was studied by infection of primary Tupaia hepatocytes with HBV.RESULTS:We identified by sequence alignment and functional experiments a conserved bipartite NLS containing a casein kinaseⅡ(CKⅡ)phosphorylation site located within the terminal protein domain(TP)of the HBV polymerase.Inhibition of CKⅡimpairs the functionality of this NLS and thereby prevents the nuclear import of the polymerase.Binding of the import factor karyopherin-α2 to the polymerase depends on its CKⅡ-mediated phosphorylation of the bipartite NLS.In HBV-infected primary Tupaia hepatocytes CKⅡinhibition in the early phase(post entry phase)of the infection process prevents the establishment of the infection.CONCLUSION:Based on these data it is suggested that during HBV infection the final import of the genome complex into the nucleus is mediated by a novel bipartite NLS localized in the TP domain of HBV polymerase. | Joachim Lupberger Stephanie Schaedler Alexander Peiran Eberhard Hildt | 2013 | World Journal of Gastroenterology2013,19,44: | 8 |

| 19 | Increasing power plant efficiency with clustering methods and Variable Importance Index assessment显示文摘Power plant performance can decrease along with its life span,and move away from the design and commissioning targets.Maintenance issues,operational practices,market restrictions,and financial objectives may lead to that behavior,and the knowledge of appropriate actions could support the system to retake its original operational performance.This paper applies unsupervised machine learning techniques to identify operating patterns based on the power plant’s historical data which leads to the identification of appropriate steam generator efficiency conditions.The selected operational variables are evaluated in respect to their impact on the system performance,quantified by the Variable Importance Index.That metric is proposed to identify the variables among a much wide set of monitored data whose variation impacts the overall power plant operation,and should be controlled with more attention.Principal Component Analysis(PCA)and k-means++clustering techniques are used to identify suitable operational conditions from a one-year-long data set with 27 recorded variables from a steam generator of a 360MW thermal power plant.The adequate number of clusters is identified by the average Silhouette coefficient and the Variable Importance Index sorts nine variables as the most relevant ones,to finally group recommended settings to achieve the target conditions.Results show performance gains in respect to the average historical values of 73.5%and the lowest efficiency condition records of 68%,to the target steam generator efficiency of 76%. | Jéssica Duarte Lara Werncke Vieira Augusto Delavald Marques Paulo Smith Schneider Guilherme Pumi Taiane Schaedler Prass | 2021 | Energy and AI2021,5,3: | 0 |
| 20 | Microcephaly prevalence after the 2015 to 2016 Zika outbreak in Tangará da Serra, Brazil: a population-based study显示文摘Objective::Prenatal infection with the Zika virus (ZIKV) can lead to congenital Zika syndrome (CZS), characterized by microcephaly and brain injury. However, there are questions regarding the prevalence of microcephaly/CZS after the ZIKV outbreak in defined geographic areas. This study aimed to identify adverse outcomes in live births of fetuses exposed in utero to the ZIKV, compared to unexposed births, as well as maternal sociodemographic, delivery, and birth characteristics.Methods::Here, we conducted a cross-sectional observational study to investigate the characteristics of all live births in the city of Tangará da Serra, Mato Grosso, Brazil, in 2016, after the outbreak of ZIKV infection in late 2015. All live births of children to women residing in the municipality of Tangará da Serra between January 1 and December 31, 2016, were evaluated, and head circumference was measured at birth and after 24 hours. Children born with microcephaly or a maternal history of confirmed or suspected prenatal ZIKV infection were evaluated by a multidisciplinary team. The outcomes of the exposed and non-exposed children were compared. Prevalence ratios and their respective 95% confidence intervals were calculated for sociodemographic, delivery, and live birth characteristics.Results::Of 1,441 live births, 106 (7.3%) were from mothers with confirmed or highly probable exposure to ZIKV.
