| 1 | Spatial and temporal variation of fish assemblages in a subtropical small stream of the Huangshan Mountain显示文摘Spatial and temporal variation of fish assemblages were investigated seasonally from May 2007 to February 2008across 11 study sites in a subtropical small stream, the Puxi Stream, of the Huangshan Mountain. Along the longitudinal gradientfrom headwater to downstream, fish species richness and abundance increased gradually, but then decreased significantly at thelower reaches. The highest species richness and abundance were observed in August and the lowest in February. Based on analysisof similarities (ANOSIM), fish assemblages were significantly different in spatial variation but not in temporal variation. Althoughdifferences were observed both among sites and among stream orders, the lower R value in order-variation suggestedstream order was not the optimal factor explaining the spatial variation of fish assemblages. In addition, dam construction did notsignificantly alter fish assemblages in the sites adjacent to and immediately downstream to dams. Using cluster analysis andnon-metric Multi Dimensional Scaling analysis (NMS), assemblages were separated into three groups at a Bray-Curtis similarityvalue of 42%: the upper, middle and lower groups. Following analysis of similarity percentages of species contributions (SIMPER),shifts in occurrence or abundance of S. curriculus, Z. platypus, R. bitterling and A.fasciatus contributed most to the differencesamongst the three groups. Standard Deviation Redundancy Analysis (RDA) suggested that habitat structure (such as elevation,substrate, and flow velocity) contributed to the spatial and temporal pattern of fish assemblages in the Puxi Stream. In conclusion,the fish assemblages in Puxi Stream presented significant spatial but not temporal variation. Human disturbance has perhapsinduced the decrease in species diversity in the lower reaches. | 2 | Changes in Chilled Chicken Quality and Correlation Analysis during Different Shelf Life显示文摘[Objective] The paper was to study the change rule and correlation analysis of quality indexes of chilled chicken within 7 d shelf life.[Method] Six pieces of fresh breast muscle were packaged in polyethylene(PE) food storage bags,labeled,and refrigerated at 4 ℃.The p H,water loss rate,tenderness,color value and inosinic acid content of chicken were measured at different time points of preservation(cold storage for 0,1,2,3,6,7 d).[Result] With the prolongation of shelf life,the p H of chilled chicken first increased then tended to be stable;the water loss rate first increased then stabilized;the shear force first decreased then increased.There was little difference in brightness value(L*) of chilled chicken dur-ing different shelf life(P>0.05).The red degree value(a*) decreased with the prolongation of shelf life;the yellow degree value(b*) increased with the extension of shelf life.The inosinic acid content decreased with the prolongation of shelf life.The p H of chilled chicken in different shelf life had significant positive correlation with water loss rate(P<0.05) and extremely significant negative correlation with shear force(P<0.01).[Conclusion]The shelf life of 7 d has impact on meat color and inosinic acid of chilled chicken.Cold fresh preservation of chilled chicken should be less than 6 d;p H has certain correlation with water loss rate and shear force. | Ju Xiaojun Tu Yunjie Zou Jianmin Zhang Ming Ji Gaige Shan Yanju Liu Yifan Shu Jingting | 2018 | Animal Husbandry and Feed Science2018,10,2: | 2 |
| 3 | Culture and Identification of Myoblasts Isolated from Duck Embryos显示文摘[Objective] Using embryonic myoblasts to research the formation and developmental mechanisms of skeletal muscle is becoming a research hotspot. This study aimed to establish a method of isolation, culture and identification of myoblasts in duck embryos. [Method] Pectoral and leg muscle samples were isolated from the embryos of Gaoyou duck at 13 d of hatching, then disassociated with collagenase and trypsin and purified via differential adhesion. The isolated cells were cultured in vitro and detected for the expression of Pax7 protein using immunofluorescence technique. [Result] Myoblasts were obtained successfully both from pectoral and leg muscles in duck embryos and these cells proliferated strongly and differentiated well. Immunofluorescence staining showed that more than 95% cells could express Pax7 protein. | 4 | A New Detecting Method for the Sequence Difference of CHD1 Gene on Sex Chromosomes Z and W in Domesticated Ducks of China显示文摘A new method for detecting the sequence differences of duck Chromo-helicase-DNA-Binding 1( CHD1) genes on sex chromosomes Z and W was established by amplifying DNA fragment length poly morphism,which aimed to solve some technical problems,such as the sex identification with nondamage,the misclassifc rate of artificial sex identification during embryonic period,and the injury caused by anal reversal identification method during neonatal period. The improved sex identification method was developed by DNA amplified fragment length polymorphism by using CHD1 gene sequence in bird. The sex identification PCR primers YPF/YPR were used,and the PCR product was cloned,sequenced and blasted; and the effectiveness and accuracy of this method were verified through living examples. 2% agarose gel electrophoresis was used to distinguish 495 bp CHD1-Z and 351 bp CHD1-W obtained by primer YPF/YPR. The female( ZW) displayed two visible bands; while male( ZZ) had only one single band.
