4Genetic variants at 10q 23.33 are associated with plasma lipid levels in a Chinese population显示文摘Plasma lipid abnormalities are implicated in the pathogenic process of type 2 diabetes.The IDE-KIF11-HHEX gene cluster on chromosome 10q23.33 has been identified as a susceptibility locus for type 2 diabetes.We hypothesized that genetic variants at 10q23.33 may be associated with plasma lipid concentrations.Seven tagging single nucleotide polymorphisms(SNPs:rs7923837,rs2488075,rs947591,rs11187146,rs5015480,rs4646957 and rs1111875) at 10q23.33 were genotyped in 3,281 subjects from a Han Chinese population,using the TaqMan OpenArray and Sequenom MassARRAY platforms.Multiple linear regression analyses showed that SNP rs7923837 in the 3'-flanking region of HHEX was significantly associated with triglyceride levels(P = 0.019,0.031 mmol/L average decrease per minor G allele) and that rs2488075 and rs947591 in the downstream region of HHEX were significantly associated with total cholesterol levels(P = 0.041,0.058 mmol/L average decrease per minor C allele and P = 0.018,0.063 mmol/L average decrease per minor A allele,respectively).However,the other four SNPs(rs11187146,rs5015480,rs4646957 and rs1111875) were not significantly associated with any plasma lipid concentrations in this Chinese population.Our data suggest that genetic variants in the IDE-KIF11-HHEX gene cluster at 10q23.33 may partially explain the variation of plasma lipid levels in the Han Chinese population.Further studies are required to confirm these findings in other populations.Sijun Liu Yun Qian Feng Lu Meihua Dong Yudi Lin Huizhang Li Chong Shen Juncheng Dai Yue Jiang Guangfu Jin Zhibin Hu Hongbing Shen 2014The Journal of Biomedical Research2014,28,1:1

7Concurrent validity of evaluating knee kinematics using Kinect system during rehabilitation exercise显示文摘Squats,ascending and descending stairs,and lunges are three functional movements popularly used in clinical rehabilitation.This study aimed to analyze the validity of the Kinect v2 for assessing the kinematics of three functional knee movements.An intelligent program was developed for real-time kinematic evaluation of the knee joint using the Kinect v2 system.Based on image recognition technology,images from the Kinect v2 depth camera were used to develop a kinematic evaluation program for knee joint movements.The kinematics of 30 healthy volunteers during stair ascent and descent,squatting,and lunging movements were collected synchronously with both VICON and Kinect systems.The peak knee angles in the sagittal and coronal planes were extracted and analyzed.A paired samples t-test and the relative coefficient within the groups were used for statistical analysis of the validity of the Kinect compared to the VICON.There was no significant difference(p>0.05)in bilateral knee flexion angle when lunging.No significant difference(p>0.05)was found in left knee flexion during squatting exercises.Although the Kinect generally underestimated knee flexion and valgus,it displayed excellent consistency and good to excellent agreement with the VICON motion analysis system.The results demonstrated that the Kinect based measurement tool for analyzing knee kinematics has the potential to be an effective clinical training device for maintaining knee alignment during exercise.Guojiong Hu Wenli Wang Bin Chen Hongping Zhi Yudi Li Yixin Shen Kuan Wang 2021Medicine in Novel Technology and Devices2021,,3:0
8Hidden Genetic Regulation of Human Complex Traits via Brain Isoforms显示文摘Alternative splicing exists in most multi-exonic genes,and exploring these complex alternative splicing events and their resultant isoform expressions is essential.However,it has become conventional that RNA sequencing results have often been summarized into gene-level expression counts mainly due to the multiple ambiguous mapping of reads at highly similar regions.Transcript-level quantification and interpretation are often overlooked,and biological interpretations are often deduced based on combined transcript information at the gene level.Here,for the most variable tissue of alternative splicing,the brain,we estimate isoform expressions in 1,191 samples collected by the Genotype-Tissue Expression(GTEx)Consortium using a powerful method that we previously developed.We perform genome-wide association scans on the isoform ratios per gene and identify isoform-ratio quantitative trait loci(irQTL),which could not be detected by studying gene-level expressions alone.By analyzing the genetic architecture of the irQTL,we show that isoform ratios regulate edu-cational attainment via multiple tissues including the frontal cortex(BA9),cortex,cervical spinal cord,and hippocampus.These tissues are also associated with different neuro-related traits,including Alzheimer’s or dementia,mood swings,sleep duration,alcohol intake,intelligence,anxiety or depression,etc.Mendelian randomization(MR)analysis revealed 1,139 pairs of isoforms and neuro-related traits with plausible causal relationships,showing much stronger causal effects than on general diseases measured in the UK Biobank(UKB).Our results highlight essential transcript-level biomarkers in the human brain for neuro-related complex traits and diseases,which could be missed by merely investigating overall gene expressions.Lu Pan Chenqing Zheng Zhijian Yang Yudi Pawitan Trung Nghia Vu Xia Shen 2023Phenomics2023,3,3:0
