| 3 | Constraining Anthropogenic CH_4 Emissions in Nanjing and the Yangtze River Delta,China,Using Atmospheric CO_2 and CH_4 Mixing Ratios显示文摘Methane(CH_4) emissions estimated with the Intergovernmental Panel on Climate Change(IPCC) inventory method at the city and regional scale are subject to large uncertainties. In this study, we determined the CH_4:CO_2emissions ratio for both Nanjing and the Yangtze River Delta(YRD), using the atmospheric CH_4 and CO_2concentrations measured at a suburban site in Nanjing in the winter. The atmospheric estimate of the CH_4:CO_2emissions ratio was in reasonable agreement with that calculated using the IPCC method for the YRD(within 20%), but was 200% greater for the municipality of Nanjing. | 4 | Dissecting conserved cis-regulatory modules of Glu-1 promoters which confer the highly active endosperm-specific expression via stable wheat transformation显示文摘Wheat high-molecular-weight glutenin subunits(HMW-GS) determine dough elasticity and play an essential role in processing quality. HMW-GS are encoded by Glu-1 genes and controlled primarily at transcriptional level, implemented through the interactions between cis-acting elements and trans-acting factors. However, transcriptional mechanism of Glu-1 genes remains elusive. Here we made a comprehensive analysis of cis-regulatory elements within 1-kb upstream of the Glu-1 start codon(-1000 to-1) and identified 30 conserved motifs. Based on motif distribution pattern, three conserved cis-regulatory modules(CCRMs), CCRM1(-300 to-101), CCRM2(-650 to-400), and CCRM3(-950 to-750), were defined, and their functions were characterized in wheat stable transgenic lines transformed with progressive 5′ deletion promoter::GUS fusion constructs. GUS staining, qP CR and enzyme activity assays indicated that CCRM2 and CCRM3 could enhance the expression level of Glu-1, whereas the 300-bp promoter(-300 to-1), spanning CCRM1 and core region(-100 to-1), was enough to ensure accurate Glu-1 initiation at 7 days after flowering(DAF) and shape its spatiotemporal expression pattern during seed development. Further transgenic assays demonstrated that CCRM1-2(-300 to-209) containing Complete HMW Enhancer(-246 to-209) was important for expression level but had no effect on expression specificity in the endosperm. In contrast, CCRM1-1(-208 to-101) was critical for both expression specificity and level of Glu-1. | 5 | A Python Security Analysis Framework in Integrity Verification and Vulnerability Detection显示文摘Python is widely used in web crawler, machine learning, data analysis and so on. However, there is no guarantee that Python scripts are trusted in their whole lifetime because of system insecurity. When the system is attacked, scripts in the computer are likely to be tampered with. Therefore, the trustworthiness of Python scripts needs to be checked through different configuration strategies, including integrity verification and vulnerability detection. In this paper, integrity verification and vulnerability detection are based on two Python scripts, an original Python script and a current Python script, and the original Python script is assumed to has no vulnerabilities. By comparing with the original script, we can find out whether the current script is integrity or not and detect whether there are vulnerabilities if the integrity of the current file is destroyed. Integrity verification with Hash functions is not applied in some cases. In this mode, any changes including blank lines added are considered illegal. So loose integrity verification by combining UNIX diff tool with abstract syntax trees is proposed. The vulnerability detection starts from the premise that the original Python script has no vulnerabilities, and taint analysis is applied on the vulnerability detection framework Bandit to find vulnerabilities. Besides, in order not to change the usage of Python, both integrity verification and vulnerability detection modules are embedded in Python interpreter.
