1Genome-wide variants of Eurasian facial shape differentiation and a prospective model of DNA based face prediction显示文摘It is a long-standing question as to which genes define the characteristic facial features among different ethnic groups. In this study, we use Uyghurs, an ancient admixed population to query the genetic bases why Europeans and Han Chinese look different. Facial traits were analyzed based on high-dense 3D facial images; numerous biometric spaces were examined for divergent facial features between European and Han Chinese, ranging from inter-landmark distances to dense shape geometrics. Genome-wide association studies(GWAS) were conducted on a discovery panel of Uyghurs. Six significant loci were identified, four of which, rs1868752, rs118078182, rs60159418 at or near UBASH3B, COL23A1, PCDH7 and rs17868256 were replicated in independent cohorts of Uyghurs or Southern Han Chinese. 2A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 3Boosting Lean Electrolyte Lithium-Sulfur Battery Performance with Transition Metals: A Comprehensive Review显示文摘Lithium–sulfur(Li–S) batteries have received widespread attention, and lean electrolyte Li–S batteries have attracted additional interest because of their higher energy densities. This review systematically analyzes the effect of the electrolyte-to-sulfur(E/S) ratios on battery energy density and the challenges for sulfur reduction reactions(SRR) under lean electrolyte conditions. Accordingly, we review the use of various polar transition metal sulfur hosts as corresponding solutions to facilitate SRR kinetics at low E/S ratios(< 10 μL mg~(-1)), and the strengths and limitations of different transition metal compounds are presented and discussed from a fundamental perspective. Subsequently, three promising strategies for sulfur hosts that act as anchors and catalysts are proposed to boost lean electrolyte Li–S battery performance. 4Enhancement and mapping of tolerance to salt stress and 5-fluorocytosine in synthetic yeast strains via SCRaMbLE显示文摘Varied environmental stress can affect cell growth and activity of the cellular catalyst.Traditional path of adaptive evolution generally takes a long time to achieve a tolerance phenotype,meanwhile,it is a challenge to dissect the underlying genetic mechanism.Here,using SCRaMbLE,a genome scale tool to generate random structural variations,a total of 222 evolved yeast strains with enhanced environmental tolerances were obtained in haploid or diploid yeasts containing six synthetic chromosomes.Whole genome sequencing of the evolved strains revealed that these strains generated different structural variants.Notably,by phenotypic-genotypic analysis of the SCRaMbLEd strains,we find that a deletion of gene YFR009W(GCN20)can improve salt tolerance of Saccharomyces cerevisiae,and a deletion of gene YER056C can improve 5-flucytosine tolerance of Saccharomyces cerevisiae.This study shows applications of SCRaMbLE to accelerate phenotypic evolution for varied environmental stress and to explore relationships between structural variations and evolved phenotypes.Jianping Kang Jieyi Li Zhou Guo Sijie Zhou Shuxin Su Wenhai Xiao Yi Wu Yingjin Yuan 2022Synthetic and Systems Biotechnology2022,7,3:2
53D Printing of Well Dispersed Electrospun PLGA Fiber Toughened Calcium Phosphate Scaffolds for Osteoanagenesis显示文摘Although the toughening of Calcium phosphate(CaP)scaffold by the addition of fiber has been well recognized,integrated mechanical,structural and functional considerations have been neglected in the design and fabrication of CaP scaffold implant.The emerging 3D printing provides a promising technique to construct CaP scaffold with precise size and elaborate microstructure.However,the most challenge is to extrude smoothly the CaP paste containing fibers for frequently-used extrusion-based 3D printing.In this study,frozen section and chemical dispersant(Pluronic F127,F127)were employed jointly to prepare non-aggregated polylactic-co-glycolic acid(PLGA)fibers.The injectability of CaP pastes with well dispersed PLGA fibers was more than 90%when the content of PLGA fibers was no more than 3 wt%.Meanwhile rheological property of CAP pastes with well dispersed fibers showed shear thinning,which were both beneficial to extrude CaP paste with well dispersed fibers for 3D printing.Moreover,these CaP scaffolds showed ductile fracture behavior due to the pullout and bridging effect of PLGA fibers.The cell proliferation and alkaline phosphatase(ALP)activity indicated that 3D printed CAP scaffold containing PLGA fibers possesses excellent biocompatibility and facilitate osteogenic differentiation ability.Thus,it was feasible to print CaP pastes with well dispersed PLGA fibers to construct toughening CaP scaffolds with the higher shape fidelity and complex structures,which had significant clinical potentials in osteoanagenesis due to their higher toughness and excellent biocompatibility.Guoru Zhao Rongwei Cui You Chen Sijie Zhou Chen Wang Zhangmei Hu Xiaoke Zheng Maohong Li Shuxin Qu 2020Journal of Bionic Engineering2020,17,4:2

7Dynamics of synthetic yeast chromosome evolution shaped by hierarchical chromatin organization显示文摘Synthetic genome evolution provides a dynamic approach for systematically and straightforwardly exploring evolutionary processes.Synthetic Chromosome Rearrangement and Modification by LoxP-mediated Evolution(SCRaMbLE)is an evolutionary system intrinsic to the synthetic yeast genome that can rapidly drive structural variations.Here,we detect over 260000 rearrangement events after the SCRaMbLEing of a yeast strain harboring 5.5 synthetic yeast chromosomes(synⅡ,synⅢ,synⅤ,circular synⅥ,synⅨR and synⅩ).Remarkably,we find that the rearrangement events exhibit a specific landscape of frequency.We further reveal that the landscape is shaped by the combined effects of chromatin accessibili and spatial contact probability.The rearrangements tend to occur in 3D spatially proximal and chromatin-accessible regions.The enormous numbers of rearrangements mediated by SCRaMbLE provide a driving force to potentiate directed genome evolution,and the investigation of the rearrangement landscape offers mechanistic insights into the dynamics of genome evolution.Sijie Zhou Yi Wu Yu Zhao Zhen Zhang Limin Jiang Lin Liu Yan Zhang Jijun Tang Ying-Jin Yuan 2023National Science Review2023,10,5:1

12Physical and toxicological profiles of human IAPP amyloids and plaques显示文摘Although much has been learned about the fibrillization kinetics, structure and toxicity of amyloid proteins, the properties of amyloid fibrils beyond the saturation phase are often perceived as chemically and biologically inert, despite evidence suggesting otherwise. To fill this knowledge gap, we examined the physical and biological characteristics of human islet amyloid polypeptide(IAPP) fibrils that were aged up to two months. Not only did aging decrease the toxicity of IAPP fibrils, but the fibrils also sequestered fresh IAPP and suppressed their toxicity in an embryonic zebrafish model. The mechanical properties of IAPP fibrils in different aging stages were probed by atomic force microscopy and sonication, which displayed comparable stiffness but age-dependent fragmentation, followed by self-assembly of such fragments into the largest lamellar amyloid structures reported to date. 15Revealing the frequency-dependent oscillations in the nonlinear terahertz response induced by the Josephson current显示文摘Nonlinear responses of superconductors to intense terahertz radiation has been an active research frontier.Using terahertz pump-terahertz probe spectroscopy,we investigate the c-axis nonlinear optical response of a high-temperature superconducting cuprate.After excitation by a single-cycle terahertz pump pulse,the reflectivity of the probe pulse oscillates as the pump-probe delay is varied.Interestingly,the oscillatory central frequency scales linearly with the probe frequency,a fact widely overlooked in pump-probe experiments.By theoretically solving the nonlinear optical reflection problem on the interface,we show that our observation is well explained by the Josephson-type third-order nonlinear electrodynamics,together with the emission coefficient from inside the material into free space.The latter results in a strong enhancement of the emitted signal whose physical frequency is around the Josephson plasma edge.Our result offers a benchmark for and new insights into strong-field terahertz spectroscopy of related quantum materials.Sijie Zhang Zhiyuan Sun Qiaomei Liu Zixiao Wang Qiong Wu Li Yue Shuxiang Xu Tianchen Hu Rongsheng Li Xinyu Zhou Jiayu Yuan Genda Gu Tao Dong Nanlin Wang 2023National Science Review2023,10,11:0
16Rational design of Ruddlesden-Popper perovskite electrocatalyst for oxygen reduction to hydrogen peroxide显示文摘Although the oxygen reduction process to hydrogen peroxide(H_(2)O_(2))is a green option for H_(2)O_(2)generation,the low activity and selectivity hindered the industry's process.In recent years,the electrochemical synthesis of H_(2)O_(2)through a 2e-transfer method of oxygen reduction reaction(ORR)has piqued the interest of both academics and industry.Metal oxide catalysts have emerged as a novel family of electrochemical catalysts due to their unusual physical,chemical,and electrical characteristics.In this work,we first developed a Ruddlesden-Popper perovskite oxide(Pr_(2)NiO_(4+δ))as a highly selective and active catalyst for 2e-ORR to produce H_(2)O_(2).Molybdenum was introduced here to adjust the oxidation states of these transition metals with successful substitution into Ni-site to prepare Pr_(2)Ni1-xMoxO_(4+δ),and the molybdenum substitution improves the H_(2)O_(2)selectivity during the ORR process,in 0.1 M KOH,from 60%of Pr_(2)NiO_(4+δ)to 79%of Pr_(2)Ni_(0.8)Mo_(0.2)O_(4+δ)at 0.55 V versus RHE.A limiting H_(2)O_(2)concentration of_0.24 mM for Pr_(2)NiO_(4+δ)and 0.42 mM for Pr_(2)Ni_(0.8)Mo_(0.2)O_(4+δ)was obtained at a constant current of 10 mA/cm2 using a flow-cell reactor using a gas-diffusion electrode.Ning Han Shihui Feng Wei Guo Omar Martinez Mora Xiaolin Zhao Wei Zhang Sijie Xie Zhenyu Zhou Zhiwei Liu Qiong Liu Kai Wan Xuan Zhang Jan Fransaer 2022SusMat2022,2,4:0
17Development of a novel virus-like particle-based vaccine for preventing tick-borne encephalitis virus infection显示文摘Tick-borne encephalitis virus(TBEV)is an important tick-borne pathogen that poses as a serious public health concern.The coverage and immunogenicity of the currently available vaccines against TBEV are relatively low;therefore,it is crucial to develop novel and effective vaccines against TBEV.The present study describes a novel strategy for the assembly of virus-like particles(VLPs)by co-expressing the structural(core/prM/E)and non-structural(NS2B/NS3Pro)proteins of TBEV.The efficacy of the VLPs was subsequently evaluated in C57BL/6 mice,and the resultant IgG serum could neutralize both Far-Eastern and European subtypes of TBEV.These findings indicated that the VLP-based vaccine elicited the production of cross-subtype reactive antibodies.The VLPs provided protection to mice lacking the type I interferon receptor(IFNAR^(-/-))against lethal TBEV challenge,with undetectable viral load in brain and intestinal tissues.Furthermore,the group that received the VLP vaccine did not exhibit significant pathological changes and the inflammatory factors were significantly suppressed compared to the control group.Immunization with the VLP vaccine induced the production of multiple-cytokine-producing antiviral CD4+T cells in vivo,including TNF-α^(+),IL-2^(+),and IFN-γ^(+)T cells.Altogether,the findings suggest that noninfectious VLPs can serve as a potentially safe and effective vaccine candidate against diverse subtypes of TBEV.Jielin Tang Muqing Fu Chonghui Xu Bao Xue Anqi Zhou Sijie Chen He Zhao Yuan Zhou Jizheng Chen Qi Yang Xinwen Chen 2023Virologica Sinica2023,38,5:0
