1Protective effects of Phyllanthus acidus(L.) Skeels leaf extracts on acetaminophen and thioacetamide induced hepatic injuries in Wistar rats显示文摘Objective:To investigate and compare the hepatoprotective effects of crude ethanolic and aqueous extracts of Phyllanthus acidus(L.) Skeels(P.acidus) leaves on acetaminophen(APAP) and thioacetamide(TAA) induced liver toxicity in wistar rats.Silymarin was the reference hepatoprotective agent.Methods:In two different sets of experiments,the P.acidus extracts (200 and 400 mg/kg,body weight) and silymarin(100 mg/kg,body weight) were given orally for 7 days and a single dose of APAP(2 g/kg,per oral) or TAA(100 mg/kg,subcutaneous) were given to rats.The level of serum aspartate transaminase(AST),alanine transaminase(ALT),alkaline phosphatase(ALP),total bilirubin and total protein were monitored to assess hepatotoxicity and hepatoprotection.Results:APAP or TAA administration caused severe hepatic damage in rats as evident from significant rise in serum AST,ALT,ALP,total bilirubin and concurrent depletion in total serum protein.The P.acidus extracts and silymarin prevented the toxic effects of APAP or TAA on the above serum parameters indicating the hepatoprotective action.The aqueous extract was found to be more potent than the corresponding ethanolic extract against both toxicants.The phenolic and flavonoid content(175.02±4.35 and 74.68±1.28,respectively) and 2,2-diphenyl-1- picrylhydrazil(DPPH)[IC50=(33.2±0.31)μg/mL]scavenging potential was found maximum with aqueous extract as compared to ethanolic extract.Conclusions:The results of present study suggests that the aqueous extract of P.acidus leaves has significant hepatoprotective activity on APAP and TAA induced hepatotoxicity,which might be associate with its high phenolic and flavonoid content and antioxidant properties.Nilesh Kumar Jain Abhay K Singhai 2011Asian Pacific Journal of Tropical Medicine2011,4,6:6
2Preliminary pharmacological evaluation of Martynia annua Linn leaves for wound healing显示文摘Objective:To evaluate the wound healing potential of fractions from ethanol extract of Martynin annua(M.annua) Linn leaves.Methods:Ethanol extract of Af.annua Linn leaves was fractionate into three different fractions(MAF-A,MAF-B and MAF-C) which were screened for wound healing potential using two models:excision and incision on rats.The thin layer chromatography(TLC) profile of all fractions were analysed and TLC of luteolin was also done.The PovidoneIodine Ointment was used as reference for comparision.Excision and incision wounds were created on dorsal portion of rats for study.Wound contraction,biochemical parameters(protein level and hydroxyproline level) and histopathological study were performed in excision wound model whereas incision model was used for determination of tensile strength.Results:The wound contraction and tensile strength of skin tissues were observed significantly greater in MAF-C fraction treated group than other two fractions(P<0.01).In excision wound method(on day 18) protein content and hydroxyproline were found significantly higher in MAF-C group than control group(P<0.01).Histopathological study also showed better angiogenesis,matured collagen fibres and fibroblast cells as compared with the control group.Conclusions:In conclusion, our findings suggest that fiaction MAF-C from ethanol extract of M.annua leaves is found most effective in wound healing.Santram Lodhi Singhai AK 2011Asian Pacific Journal of Tropical Biomedicine2011,1,6:3
3Clinicohistopathological implications of phosphoserine 9 glycogen synthase kinase-3β/β-catenin in urinary bladder cancer patients显示文摘BACKGROUND Aberrant activation of phosphorylated form of glycogen synthase kinase-3β[pS9 GSK-3β(Serine 9 phosphorylation)] is known to trigger Wnt/β-catenin signal cascade but its clinicohistopathological implications in bladder carcinogenesis remain unknown.AIM To investigate the diagnostic and prognostic relevance of expressions of pS9 GSK-3β, β-catenin and its target genes in the pathobiology of bladder cancer.METHODS Bladder tumor tissues from ninety patients were analyzed for quantitative expression and cellular localization of pS9 GSK-3β by immunohistochemical(IHC) staining. Real time-quantitative polymerase chain reaction and IHC were done to check the expression of β-catenin, Cyclin D1, Snail and Slug at transcriptome and protein level respectively. Clinicohistopathological variables were obtained from histology reports, follow up and OPD visits of patients.Expressions of the markers were statistically correlated with these variables to determine their significance in clinical setting. Results were analysed using SPSS20.0 software.RESULTS Aberrant(low or no membranous/high nuclear/high cytoplasmic) expression of pS9 GSK-3β was noted in 51% patients and found to be significantly associated with tumor stage and tumor grade(P = 0.01 and 0.04; Mann Whitney U test).Thirty one percent tumors exhibited aberrant co-expression of pS9 GSK-3β andβ–catenin proteins and showed strong statistical association with tumor stage,tumor type, smoking/tobacco chewing status(P = 0.01, 0.02 and 0.04, MannWhitney U test) and shorter overall survival probabilities of patients(P = 0.02;Kaplan Meier test). Nuclear immunostaining of Cyclin D1 in tumors with altered pS9 GSK-3β/β–catenin showed relevance with tumor stage, grade and type.CONCLUSIONβ–catenin and pS9 GSK-3β proteins are identified as markers of diagnostic/prognostic significance in disease pathogenesis. 6CNTs mediated CD44 targeting;a paradigm shift in drug delivery for breast cancer显示文摘The breast cancer is one of the most common cancer affecting millions of lives worldwide.Though the prevalence of breast cancer is worldwide;however,the developing nations are having a comparatively higher percentage of breast cancer cases and associated complications.The molecular etiology behind breast cancer is complex and involves several regulatory molecules and their downstream signaling.Studies have demonstrated that the CD44 remains one of the major molecule associated not only in breast cancer but also several other kinds of tumors.The complex structure and functioning of CD44 posed a challenge to develop and deliver precise anti-cancerous drugs against targeted tissue.There are more than 20 isoforms of CD44 reported till date associated with several kinds of tumor in the using breast cancer.The success of any anti-cancerous therapy largely depends on the precise drug delivery system,and in modern days nanotechnology-based drug delivery vehicles are the first choice not only for cancer but several other chronic diseases as well.The Carbon nanotubes(CNTs)have shown tremendous scope in delivering the drug by targeting a particular receptor and molecules.Functionalized CNTs including both SWCNTs and MWCNTs are a pioneer in drug delivery with higher efficacy.The present work emphasized mainly on the potential of CNTs including both SWCNTs and MWCNTs in drug delivery for anti-cancerous therapy.The review provides a comprehensive overview of the development of various CNTs and their validation for effective drug delivery.The work focus on drug delivery approaches for breast cancer,precisely targeting CD44 molecule.Nidhi Jain Singhai Suman Ramteke 2020Genes & Diseases2020,7,2:1
