| 1 | Simultaneous determination of amlodipine,valsartan and hydrochlorothiazide by LC-ESIMS/MS and its application to pharmacokinetics in rats显示文摘Polypill is a fixed-dose combination that contains three or more active ingredients used as a single daily pill to achieve a large effect in preventing cardiovascular disease with minimal adverse effects.A novel and accurate liquid chromatography tandem mass spectrometry method using electrospray ionization mode has been developed and validated for the simultaneous determination of amlodipine(AMD),valsartan(VAL) using losartan(LOS) as an internal standard(IS),and hydrochlorothiazide(HCT) using furosemide(FSD) as an IS.The separation was carried on Aquasil C_(18)(50 mm×2.1 mm,5μm) reversed phase column using acetonitrile and water containing 0.1%formic acid(50:50,v/v) as the mobile phase.The method was validated in terms of linearity,accuracy and precision over the concentration range of 1-1000 ng/mL.The intra and inter-day precision and accuracy,stability and extraction recoveries of all the analytes were in the acceptable range.This method can be successfully applied to the pharmacokinetic study of AMD,VAL and HCT when given as a polypill. | Shankar Ganesh Gadepalli Pragney Deme Madhusudana Kuncha Ramakrishna Sistla | 2014 | Journal of Pharmaceutical Analysis2014，4，6： | 5 |
| 2 | Antioxidant and hepatoprotective effects of Boswellia ovalifoliolata bark extracts显示文摘Paracetamol(PCM) hepatotoxicity is related to reactive oxygen species(ROS) formation and excessive oxidative stress; natural antioxidant compounds have been tested as an alternative therapy. This study evaluated the hepatoprotective activity of an alcoholic extract of Boswellia ovalifoliolata(BO) bark against PCM-induced hepatotoxicity. BO extract also demonstrated antioxidant activity in vitro, as well as scavenger activity against 2, 2-diphenyl-1-picrylhydrazyl. Administration of PCM caused a significant increase in the release of transaminases, alkaline phosphatase, and lactate dehydrogenase in serum. Significant enhancement in hepatic lipid peroxidation and marked depletion in reduced glutathione were observed after parac intoxication with severe alterations in liver histology. BO treatment was able to mitigate hepatic damage induced by acute intoxication of PCM and showed a pronounced protective effect against lipid peroxidation, deviated serum enzymatic variables, and maintained glutathione status toward control. | 4 | Poly(ADP-ribose) polymerase inhibition reveals a potential mechanism to promote neuroprotection and treat neuropathic pain显示文摘Neuropathic pain is triggered by the lesions to peripheral nerves which alter their structure and function. Neuroprotective approaches that limit the pathological changes and improve the behavioral outcome have been well explained in different experimental models of neuropathy but translation of such strategies to clinics has been disappointing. Experimental evidences revealed the role of free radicals, especially peroxynitrite after the nerve injury. They provoke oxidative DNA damage and consequent over-activation of the poly(ADP-ribose) polymerase(PARP) upregulates pro-inflammatory pathways, causing bioenergetic crisis and neuronal death. Along with these changes, it causes mitochondrial dysfunction leading to neuronal apoptosis. In related preclinical studies agents that neutralize the free radicals and pharmacological inhibitors of PARP have shown benefits in treating experimental neuropathy. | 5 | Direct injection HILIC-MS/MS analysis of darunavir in rat plasma applying supported liquid extraction显示文摘A novel bioanalytical method was developed and validated for the quantitative determination of darunavir(DRV) in rat plasma by employing hydrophilic interaction chromatography and tandem mass spectrometry(HILIC-MS/MS) with supported liquid extraction(SLE).lrbesartan(IRB) was used as an internal standard(IS).The analyte in rat plasma(200 μL) was isolated through SLE using ethyl acetate as the eluting solvent.The chromatographic separation was achieved on Luna-HILIC(250 mm × 4.6 mm,5 μm)column with a mobile phase of 0.1% of formic acid in water:acetonitrile(5:95,v/v),at a constant flow rate of 1.0mL/min.The MS/MS ion transitions for DRV(548.1→392.0) and IS(429.2→207.1) were monitored on an ion trap mass spectrometer,operating in the multiple reaction monitoring(MRM) mode.The lower limit of quantitation(LLOQ) was 0.2 ng/mL and quantitation range was 0.2-5000 ng/mL.The method was validated for its selectivity,sensitivity,carryover,linearity,precision,accuracy,recovery,matrix effect and stability.The method was successfully applied to pharmacokinetic study in rats. | Bokka Ramesh Nemali Manjula Sistla Ramakrishna Potturi Sita Devi | 2015 | Journal of Pharmaceutical Analysis2015，5，1： | 2 |
