1Changing patterns and survival improvements of young breast cancer in China and SEER database, 1999-2017显示文摘Objective: Breast cancer in young females was usually considered more aggressive and requires aggressive therapy. We investigated whether early detection and improved treatments changed the patterns of characteristics,management and outcomes of young breast cancer patients over time.Methods: Females under 40 years of age diagnosed with breast cancer during the periods 1999-2017 and1999-2015 were identified in the Fudan University Shanghai Cancer Center(FUSCC) and the population-based Surveillance, Epidemiology, and End Results(SEER) registry, respectively. Clinicopathologic characteristics and treatment information were collected. Patients diagnosed before 2013 were followed up.Results: The proportions of young breast cancer patients were 15.0% and 5.3% in the FUSCC and SEER cohorts, respectively. In the FUSCC cohort, there was a significant increase in the proportion of ductal carcinoma in situ(DCIS)(from 8.8% to 16.9%;P<0.0001) and it remained stable in SEER cohort. The proportion of T1-stage tumors increased dramatically in the FUSCC cohort(from 35.3% to 41.9%;P=0.008), whereas it decreased in SEER cohort(from 42.4% to 33.0%;P<0.0001). The percentage of estrogen receptor(ER)-positive cancers was consistently increased in both the invasive ductal carcinoma(IDC) and DCIS patients in the two cohorts. Breastconserving surgery and immediate implant reconstruction after mastectomy both exhibited increased use over time in the FUSCC cohort. Both the FUSCC and SEER cohorts showed a significantly better prognosis in the recent time period.Conclusions: With the increased early-stage and ER-positive diseases in young patients as well as better systemic treatment strategies, improved survival has been observed in recent years. 2Simvastatin inhibits apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax显示文摘BACKGROUND: Many studies have showed that apoptosis of endothelial cells plays a curial role in the progress of sepsis. But the role of simvastatin in apoptosis of endothelial cells induced by sepsis is not clear. The present study aimed to investigate the role of simvastatin in apoptosis of endothelial cells induced by sepsis and its mechanism.METHODS: Human umbilical vein endothelial cells(HUVECs) were randomly divided into three groups: control group, sepsis serum intervention group(sepsis group) and simvastatin+sepsis serum intervention group(simvastatin group). After 24-hour incubation with corresponding culture medium, the relative growth rate of HUVECS in different groups was detected by MTT assay; the apoptosis of HUVECs was detected by Hoechst33258 assay and fl ow cytometry; and the expression of the Bcl-2 and Bax genes of HUVECs was detected by PCR.RESULTS: Compared with the sepsis group, HUVECs in the simvastatin group had a higher relative growth rate. Apoptotic HUVECs decreased significantly in the simvastatin group in comparison with the sepsis group. Expression of the Bcl-2 gene in HUVECs decreased obviously, but the expression of the Bax gene increased obviously after 24-hour incubation with sepsis serum; however, the expression of the Bcl-2 and Bax genes was just the opposite in the simvastatin group.CONCLUSIONS: Our study suggests that simvastatin can inhibit apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax. 3Palaeogeomorphology and its control on the development of sequence stratigraphy and depositional systems of the Early Silurian in the Tarim Basin显示文摘The Silurian in the Tarim Basin was deposited on the basement deformed by the Caledonian tectonic movements at the end of the Late Ordovician. The development and distribution of sedimentary sequences of the Early Silurian have been clearly controlled by the palaeogeomorphology of the Late Ordovician. Based on unconformity characteristics and distribution of erosion, several zones can be differentiated including a high uplifted erosion zone, a transitional slope zone and a depression zone. The central and west Tabei Uplift zones show high angular unconformity and intense erosion. The Tarim Basin in the late Ordovician shows characteristics of higher in the west, lower in the east while higher in the south, lower in the north. The Early Silurian mainly developed transgressive and highstand systems tracts on the whole, while the lowstand systems tract only developed partly below the slope break. The palaeogeomorphology controlled the clastic source supply and deposit distribution. Braided delta system and tidal flat–estuary system were deposited. The duration of uplifting of the Tazhong paleouplift was longer than that of the Tabei paleo-uplift, and deposition was later. This led to the lower and middle members of the Kepingtage Formation missing in that area. As a large-scale transgression occurred during the deposition period of the upper member of the Kepingtage Formation, sediment from the west of the basin was transported and deposited by tides and waves, forming tidal–marine debris systems above the uplift. Proximal alluvial fan and fan delta coarse clastic deposits developed in proximal uplift zone in the east and southeast of the basin, and braided delta put forward to the transitional zone between the edge of uplift and the sea. Large-scale tidal channel, sub-distributary channel and mouth bar of the delta front can form favorable reservoirs, and they are primary targets for oil and gas exploration. 7Rescue of premature aging defects in Cockayne syndrome stem cells by CRISPR/Cas9-mediated gene correction显示文摘Cockayne syndrome(CS)is a rare autosomal recessive inherited disorder characterized by a variety of clinical features,including increased sensitivity to sunlight,progressive neurological abnormalities,and the appearance of premature aging.However,the pathogenesis of CS remains unclear due to the limitations of current disease models.Here,we generate integration-free induced pluripotent stem cells(iPSCs)from fibroblasts from a CS patient bearing mutations in CSB/ERCC6 gene and further derive isogenic genecorrected CS-iPSCs(GC-iPSCs)using the CRISPR/Cas9 system.CS-associated phenotypic defects are recapitulated in CS-iPSC-derived mesenchymal stem cells(MSCs)and neural stem cells(NSCs),both of which display increased susceptibility to DNA damage stress.Premature aging defects in CS-MSCs are rescued by the targeted correction of mutant ERCC6.We next map the transcriptomic landscapes in CS-iPSCs and GC-iPSCs and their somatic stem cell derivatives(MSCs and NSCs)in the absence or presence of ultraviolet(UV)and replicative stresses,revealing that defects in DNA repair account for CS pathologies.Moreover,we generate autologous GC-MSCs free of pathogenic mutation under a cGMP(Current Good Manufacturing Practice)-compliant condition,which hold potential for use as improved biomaterials for future stem cell replacement therapy for CS.Collectively,our models demonstrate novel disease features and molecular mechanisms and lay a foundation for the development of novel therapeutic strategies to treat CS.Si Wang Zheying Min Qianzhao Ji Lingling Geng Yao Su Zunpeng Liu Huifang Hu Lixia Wang Weiqi Zhang Keiichiro Suzuiki Yu Huang Puyao Zhang Tie-Shan Tang Jing Qu Yang Yu Guang-Hui Liu Jie Qiao 2020Protein & Cell2020,11,1:7

9Surveys in Areas of High Risk of Iodine Deficiency and Iodine Excess in China, 2012-2014: Current Status and Examination of the Relationship between Urinary Iodine Concentration and Goiter Prevalence in Children Aged 8-10 Years显示文摘Objective We aimed to evaluate goiter prevalence and iodine nutritional status in areas with high levels of water iodine;to monitor the prevalence of iodine deficiency disorders(IDD)in areas at high risk of IDD;and to compare the prevalence of goiter and urine iodine(UI)concentrations between children living in the two areas.Methods Based on surveillance from 2012-2014,we analyzed the concentration of UI and prevalence of goiter in 8-10-year-old children from 12 high-risk IDD provinces,and from 8 provinces and municipalities with excessive water iodine.We calculated goiter prevalence for each UI level according to World Health Organization(WHO)standards and constructed predictive prevalence curves.Results The goiter prevalence and median UI of children from areas with high water iodine were not optimal,being above the WHO standards(5%and 100-199μg/L,respectively),whereas those in high-risk areas fell within the standard.UI and goiter prevalence exhibited a U-shaped relationship in high-risk endemic areas and a parabolic relationship in areas of iodine excess.Conclusion Iodine surplus in high-iodine areas leads to high goiter prevalence and UI.However,in high-risk areas,UI was optimal and goiter prevalence met the national criteria for IDD elimination.CUI Si Lu LIU Peng SU Xiao Hui LIU Shou Jun 2017Biomedical and Environmental Sciences2017,30,2:6
10s Early-onset in Major Depression a Predictor of Specific Clinical Features with More Impaired Social Function?显示文摘Background: Early-onset major depressive disorder (MDD) (EOD) is often particularly malignant due to its special clinical features, accompanying impaired social function, protracted recovery time, and frequent recurrence. This study aimed to observe the effects of age onset on clinical characteristics and social function in MDD patients in Asia. Methods: In total, 547 out-patients aged 18-5 years who were from 13 study sites in five Asian countries were included. These patients had MDD diagnose according to the Diagnostic and Statistical Manual of Mental Disorders, 4^th Edition criteria. Clinical features and social function were assessed using Symptom Checklist-90-revised (SCL-90-R) and Sheehan Disability Scale (SDS). Quality of life was assessed by a 36-item Short-form Health Survey (SF-36). Analyses were performed using a continuous or dichotomous (cut-off: 30 years) age-of-onset indicator. Results: Early-onset MDD (EOD, 〈30 years) was associated with longer illness (P - 0.003), unmarried status (P 〈 0.001), higher neuroticism (P 〈 0.002) based on the SCL-90-R, and more limited social function and mental health (P = 0.006, P = 0.007) based on the SF-36 and SDS. The impairment of social function and clinical severity were more prominent at in-patients with younger onset ages. Special clinical features and more impaired social function and quality of life were associated with EOD, as in western studies. Conclusions: EOD often follows higher levels of neuroticism. 11Identification and Evolution of Groundwater Chemistry in the Ejin Sub-Basin of the Heihe River, Northwest China显示文摘Hydro-chemical characteristics of groundwater and their changes as affected by human activities were studied in the Ejin Sub-Basin of the Inner Mongolia Autonomous Region, China, to understand the groundwater evolution, to identify the predominant geochemical processes taking place along the horizontal groundwater flow path, and to characterize anthropogenic factors affecting the groundwater environment based on previous data. The concentrations of major ions and total dissolved solids (TDS) in the groundwater showed a great variation, with 62.5% of the samples being brackish (TDS≥1000 mg L-1). The groundwater system showed a gradual hydro-chemical zonation composed of Na+-HCO3-, Na+-Mg2+-SO42-Cl-, and Na+-Cl-. The relationships among the dissolved species allowed identification of the origin of solutes and the processes that generated the observed water compositions. The dissolution of halite, dolomite, and gypsum explained, in part, the presence of Na+, K+, Cl-, SO42-, and Ca2+, but other processes, such as mixing, Na+ exchange for Ca2+ and Mg2+, and calcite precipitation also contributed to the composition of water. Human activity, in particular large-scale water resources development associated with dramatic population growth in the last 50 years, has led to tremendous changes in the groundwater regime, which reflected in surface water runoff change, decline of groundwater table and degeneration of surface water and groundwater quality. 12Dihydroartemisinin suppresses renal fibrosis in mice by inhibiting DNA-methyltransferase 1 and increasing Klotho显示文摘Renal fibrosis is an unavoidable end result of all forms of progressive chronic kidney diseases(CKD).Discovery of efficacious drugs against renal fibrosis is in crucial need.In a preliminary study we found that a derivative of artemisinin,dihydroartemisinin(DHA),exerted strong renoprotection,and reversed renal fibrosis in adenine-induced CKD mouse model.In this study we investigated the anti-fibrotic mechanisms of DHA,particularly its specific target in renal cells.Renal fibrosis was induced in mice by unilateral ureteral obstruction(UUO)or oral administration of adenine(80 mg·kg^(-1)),the mice received DHA(30 mg·kg^(-1)·d^(-1),i.g.)for 14 or 21 days,respectively.We showed that DHA administration markedly attenuated the inflammation and fibrotic responses in the kidneys and significantly improved the renal function in both the renal fibrosis mouse models.In adenine-treated mice,DHA was more effective than 5-azacytidine against renal fibrosis.The anti-fibrotic effects of DHA were also observed in TGF-β1-treated HK-2 cells.In order to determine the target protein of DHA,we conducted pull-down technology coupled with shotgun proteomics using a small-molecule probe based on the structure of DHA(biotin-DHA).As a results,DNA methyltransferase 1(DNMT1)was identified as the anti-fibrotic target of DHA in 3 different types of renal cell lines(HK-2,HEK293 and 3T3).We demonstrated that DHA directly bound to Asn 1529 and Thr 1528 of DNMT1 with a Kd value of 8.18μM.In primary mouse renal tubular cells,we showed that DHA(10μM)promoted DNMT1 degradation via the ubiquitin–proteasome pathway.DHA-reduced DNMT1 expression effectively reversed Klotho promoter hypermethylation,which led to the reversal of Klotho protein loss in the kidney of UUO mice.This subsequently resulted in inhibition of the Wnt/β-catenin and TGF-β/Smad signaling pathways and consequently conferred renoprotection in the animals.Knockdown of Klotho abolished the renoprotective effect of DHA in UUO mice.Our study reveals a novel pharmacological activity for DHA,i.e.,renoprotection.DHA exhibits this effect by targeting DNMT1 to reverse Klotho repression.This study provides an evidence for the possible clinical application of DHA in the treatment of renal fibrosis.Wei Zhou Min-min Chen Hui-ling Liu Zi-lin Si Wen-hui Wu Hong Jiang Lin-xiao Wang Nosratola D.Vaziri Xiao-fei An Ke Su Cheng Chen Ning-hua Tan Zhi-hao Zhang 2022Acta Pharmacologica Sinica2022,43,10:4
13Microclimatic characteristics of the Heihe oasis in the hyperarid zone of China显示文摘The microclimate of a desert oasis in hyperarid zone of China was monitored using micrometeorological methods and compared with those of areas adjacent to forested land. Differences in ground-level photosynthetically active radiation (PAR) on clear, cloudy and dust storm days and their subtending causes are analysed and discussed. Desert oases serve the ecological functions of altering solar radiation, adjusting near-ground and land surface temperatures, reducing soil temperature differences, lowering wind velocity, and increasing soil and atmospheric humidity. The total solar radiation in the interior of the oasis was roughly half of that outside a forest canopy. During the growing season, air temperatures in Populus euphratica Oliv. (poplar) and Tamarix ramosissima Ledeb. (tamarisk) forests were 1.62 °C and 0.83 °C lower respectively than those in the areas around the forests. Furthermore, the taller the forest cover, the greater the temperature drops; air temperatures in the upper storey were greater than those in the lower storey, i.e., air temperature rose with increasing height. Over the growing season, the relative humidities of the air in the poplar and tamarisk forests were 8.5% and 4.2% higher respectively than those in areas around the forests. Mean wind velocity in poplar-forested lands was 0.33 m s-1, 2.31 m s-1 lower than that in the surrounding area. 14The origin of Mosuo people as revealed by mtDNA and Y chromosome variation显示文摘The Mosuo, living in the Lugu Lake area in northwest Yunnan Province, China, is the only matriarchal population in China. The Mosuo was officially identified as Naxi nationality although its relationship with Naxi remains controversial. We studied the genetic relationship between the Mosuo and five other ethnic groups currently residing in northwest Yunnan, i.e. Naxi, Tibetan, Bai, Yi and Pumi, by typing the genetic variations in mtDNA HVS1 and 21 Y chromosome markers (13 SNPs & 8 STR markers). We showed that the maternal lineages of the Mosuo bear the strongest resemblance with those found in Naxi while its paternal lineages are more similar to those that are prevalent in Yunnan Tibetan. 15Progress and challenges in research of the mechanisms of anhedonia in major depressive disorder显示文摘There is an increasing heavy disease burden of major depressive disorder(MDD)globally.Both high diagnostic heterogeneity and complicated pathological mechanisms of MDD pose significant challenges.There is much evidence to support anhedonia as a core feature of MDD.In the Diagnostic and Statistical Manual of Mental Disorders,Fifth Edition,anhedonia is further emphasised as a key item in the diagnosis of major depression with melancholic features.Anhedonia is a multifaceted symptom that includes deficits in various aspects of reward processing,such as anticipatory anhedonia,consummatory anhedonia,and decision-making anhedonia.Anhedonia is expected to become an important clinicopathological sign for predicting the treatment outcome of MDD and assisting clinical decision making.However,the precise neurobiological mechanisms of anhedonia in MDD are not clearly understood.In this paper,we reviewed(1)the current understanding of the link between anhedonia and MDD;(2)the biological basis of the pathological mechanism of anhedonia in MDD;and(3)challenges in research on the pathological mechanisms of anhedonia in MDD.A more in-depth understanding of anhedonia associated with MDD will improve the diagnosis,prediction,and treatment of patients with MDD in the future.Yun-Ai Su Tianmei Si 2022General Psychiatry2022,35,1:2
16Mindfulness-based online intervention on mental health and quality of life among COVID-19 patients in China: an intervention design显示文摘Background:COVID-19 can lead to increased psychological symptoms such as post-traumatic stress disorder(PTSD),depression,and anxiety among patients with COVID-19.Based on the previous mindfulness-based interventions proved to be effective,this protocol reports a design of a randomized controlled trial aiming to explore the efficacy and possible mechanism of a mindful living with challenge(MLWC)intervention developed for COVID-19 survivors in alleviating their psychological problems caused by both the disease and the pandemic.Methods:In April 2021,more than 1600 eligible participants from Hubei Province of China will be assigned 1:1 to an online MLWC intervention group or a waitlist control group.All participants will be asked to complete online questionnaires at baseline,post-program,and 3-month follow-up.The differences of mental health status(e.g.PTSD)and physical symptoms including fatigue and sleeplessness between the COVID-19 survivors who receiving the online MLWC intervention and the control group will be assessed.In addition,the possible mediators and moderators of the link between the MLWC intervention and target outcomes will be evaluated by related verified scales,such as the Five Facets Mindfulness Questionnaire.Data will be analyzed based on an intention-to-t「eat approach,and SPSS software will be used to perform statistical analysis.Discussion:The efficacy and potential mechanism of MLWC intervention in improving the quality of life and psychological status of COVID-19 survivors in China are expected to be reported.Findings from this study will shed light on a novel and feasible model in improving the psychological well-being of people during such public health emergencies.Trial registration Chinese ClinicalTrial Registry(ChiCTR),ChiCTR2000037524;Registered on August 29,2020,http://www.chictr.org.cn/showproj.aspx?proj=60034.Ming-Yu Si Wei-Jun Xiao Chen Pan Hao Wang Yi-Man Huang Jun Lian Winnie W.S.Mak Zhi-Wei Leng Xiao-You Su Qiu-PingTang Yu Jiang Lu-Zhao Feng Wei-Zhong Yang Chen Wang 2021Infectious Diseases of Poverty2021,10,3:2
17Cenozoic ultrahigh-temperature metamorphism in pelitic granulites from the Mogok metamorphic belt, Myanmar显示文摘Ultrahigh-temperature(UHT)metamorphism is critical for understanding the most extreme thermal evolution of continental crust.However,UHT metamorphism predominantly occurred in the Precambrian and is rarely observed in the modern Earth.Here,we report the discovery of~25 Ma UHT granulites from the Mogok metamorphic belt(MMB)in Myanmar via a combined study of petrology and geochronology.The studied pelitic granulites well preserve a peak mineral assemblage of garnet+sillimanite+plagioclase(antiperthite)+K-feldspar+quartz+Ti-rich biotite+rutile+ilmenite.Pressure(P)-temperature(T)pseudosections and conventional geothermobarometry data only constrain the P-T conditions of the peak stage to<12 kbar and 780–890°C.However,high Zr contents in the matrix rutile(3005–4308 ppm)and high Ti contents(up to 9.2 wt% TiO_(2))in the biotite demonstrate that the Mogok granulites may have experienced UHT metamorphism.The Zr-in-rutile thermometer and X_(Grs) isopleth in the pseudosections yield peak P-T conditions of~12 kbar and>900°C.In situ SIMS and LAICP-MS U-Pb dating and trace element analyses show that both metamorphic zircon cores and rims have flat heavy rare earth element(HREE)patterns with negative Eu anomalies.The metamorphic zircon rims show the lowest HREE contents and yield ^(206)Pb/^(238)U ages of 24.9±0.5 and 25.4±0.6 Ma,respectively,representing the timing of UHT metamorphism.Our results indicate that the central MMB underwent~25 Ma UHT metamorphism,which is possibly induced by continental rifting along the thinned orogenic lithosphere.Our data,as well as reported Cenozoic UHT events,further suggest that UHT metamorphism can be produced in the modern plate tectonic regime by lithospheric extension.Si CHEN Yi CHEN Yibing LI Bin SU Qinghua ZHANG Me Me AUNG Kyaing SEIN 2021Science China Earth Sciences2021,64,11:2
18Prefrontal Nectin3 Reduction Mediates Adolescent Stress-Induced Deficits of Social Memory,Spatial Working Memory,and Dendritic Structure in Mice显示文摘Chronic stress may disrupt the normal neurodevelopmental trajectory of the adolescent brain(especially the prefrontal cortex) and contribute to the pathophysiology of stress-related mental illnesses,but the underlying molecular mechanisms remain unclear.Here,we investigated how synaptic cell adhesion molecules(e.g.,nectin3)are involved in the effects of adolescent chronic stress on mouse medial prefrontal cortex(mPFC).Male C57BL/6N mice were subjected to chronic social instability stress from postnatal days 29 to 77.One week later,the mice exposed to chronic stress exhibited impaired social recognition and spatial working memory,simplified dendritic structure,and reduced spine density in the mPFC.Membrane localization of nectin3 was also altered,and was significantly correlated with behavioral performance.Furthermore,knocking down mPFC nectin3 expression by adeno-associated virus in adolescent mice reproduced the stress-induced changes in behavior and mPFC morphology.These results support the hypothesis that nectin3 is a potential mediator of the effects of adolescent chronic stress on prefrontal structural and functional abnormalities.Hong-Li Wang Ji-Tao Li Han Wang Ya-Xin Sun Rui Liu Xiao-Dong Wang Yun-Ai Su Tian-Mei Si 2020Neuroscience Bulletin2020,36,8:2
19Resource and trajectory optimization in UAV-powered wireless communication system显示文摘The unmanned aerial vehicle(UAV) is a promising enabler of Internet of Things(IoT) owing to its highly flexible features. Combined with wireless power transfer(WPT) techniques, a UAV can provide energy for IoT nodes, which can extend the lifetime of energy constrained communication systems. This paper studies resource and trajectory optimization in UAV-powered wireless communication systems, which consists of two UAVs and two ground nodes(GNs). The system works in a way that the two UAVs alternately charge the two GNs through wireless power transfer and two GNs also alternately send their information to the corresponding UAV with the harvested energy, which can effectively reduce the interference while receiving the information of GNs. Aiming to maximize the minimum throughput of two GNs, wireless resource and UAVs’ trajectories are jointly optimized with the constraints of UAV collision avoidance, flying speed, and transmit power. Successive convex programming(SCP) and block coordinate descent(BCD) are utilized to solve the optimization problem. 20Sap-flow measurement and scale transferring from sample trees to entire forest stand of Populus euphratica in desert riparian forest in extreme arid region显示文摘Understanding how the transpiration of this vegetation type responds to environmental stress is important for determining the wa-ter-balance dynamics of the riparian ecosystem threatened by groundwater depletion. Transpiration and sap flow were measured using the heat-pulse technique. The results were then projected up to the stand level to investigate the stand's water-use in relation to climate forcing in the desert riparian forest in an extreme arid region. This study took place from April through October 2003 and from May through October 2004. The experimental site was selected in the Populus euphratica Forest Reserve (101o10' E, 41o59' N) in Ejina county, in the lower Heihe River basin, China. The sapwood area was used as a scalar to extrapolate the stand-water consumption from the whole trees' water consumption measured by the heat-pulse velocity recorder (HPVR). Scale transferring from a series of individual trees to a stand was done according to the existing natural variations between trees under given environmental conditions. The application of the biometric parameters available from individual tree and stand levels was proved suitable for this purpose. A significant correlation between the sapwood area and tree diameter at breast height (DBH) was found. The prediction model is well fitted by the power model. On the basis of the prediction model, the sapwood area can be cal-culated by DBH.
