1金龙胶囊对胃癌细胞MGC-803和BGC-823侵袭转移能力的影响显示文摘目的:探讨金龙胶囊对人胃癌MGC-803,BGC-823细胞侵袭和迁移能力的影响并对其具体机制进行探讨。方法:体外培养胃癌MGC-803,BGC-823细胞,分为金龙胶囊组(0.1,0.2,0.4,0.8 g·L-1),空白组和5-氟尿嘧啶(5-FU)组,采用噻唑蓝(MTT)比色法分别检测金龙胶囊对MGC-803,BGC-823细胞增殖的抑制作用,计算半数抑制率(IC50);采用细胞侵袭(transwell)小室法和划痕实验检测金龙胶囊处理MGC-803和BGC-823细胞24 h后,细胞侵袭和迁移能力发生改变;蛋白免疫印迹法(Western blot)检测金龙胶囊处理MGC-803,BGC-823细胞24 h后E-钙黏素(E-cadherin),基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)蛋白表达情况。结果:金龙胶囊能明显抑制MGC-803,BGC-823细胞的增殖,呈一定的浓度及时间依赖性;与空白组比较,金龙胶囊能明细减少MGC-803,BGC-823细胞的穿膜细胞数目,可明显影响2种胃癌细胞的侵袭与迁移能力,并呈明显浓度依赖性(P〈0.05);金龙胶囊组的划痕距离较空白组明显降低,影响MGC-803,BGC-823细胞迁移,并呈浓度依赖性(P〈0.05);与空白组比较,金龙胶囊组能够提高MGC-803和BGC-823细胞中E-cadherin蛋白表达,降低MMP-2,MMP-9蛋白表达水平,且呈一定的量效关系(P〈0.05)。结论:金龙胶囊能够抑制人胃癌MGC-803,BGC-823细胞的侵袭和迁移能力,其作用机制可能与上调E-cadherin表达,抑制MMP-2,MMP-9表达水平有关。本研究证实了金龙胶囊对胃癌MGC-803,BGC-823细胞抗侵袭转移的部分机制。李丹 金凤 陶丽 倪腾洋 王海波 冯俊 朱光 钱亚云 丁岩冰 Masataka Sunagawa 刘延庆 2018中国实验方剂学杂志2018,24,19:7
