| 3 | Prospect of China's ambient air quality standards显示文摘Ambient air quality standards are the core strategic goal of ambient air quality management.Countries worldwide have given importance to research on the development of ambient air quality standards.To understand the history of the development of China’s ambient air quality standards,this study analyzed the background associated with all previous formulations and revisions of the standards,classification of functional areas,standard grading,pollutants,and evolution of the standard limits over the past 40 years.The results show that since the initial release of the“Ambient Air Quality Standard”by China in 1982,it has been supplemented once,revised twice,and modified twice.The first ambient air quality standard specified the standard limits of six pollutants commonly found in ambient air.With the development of ambient air quality management,the number of pollutants has increased to ten.Since the release of the“Ambient Air Quality Standards”in 2012,the ambient air quality in China has significantly improved.However,the proportion of cities meeting these standards is still low.At present,China is suitably positioned to make the standards associated with 24 hr SO2concentrations more stringent such that it meets the values defined in the World Health Organization(WHO)interim target-2 and the WHO air quality guideline(AQG).We further suggested that the SO2standard should be revised promptly.Simultaneously,regions with a relatively high proportion of cities meeting the standard are encouraged to introduce more stringent interim target limits in due course to manage the local ambient air quality. | Zongshuang Wang Yufei Tan Min Guo Miaomiao Cheng Yanyue Gu Siyu Chen Xuefang Wu Fahe Chai | 2023 | Journal of Environmental Sciences2023,,1: | 2 |

| 5 | 4-Phenylbutyric acid accelerates rehabilitation of barrier function in IPEC-J2 cell monolayer model显示文摘As the first line of defence against pathogens and endotoxins crossing the intestine-blood barrier,the intestinal epithelial barrier plays a determinant role in pigs’health and growth.4-Phenylbutyric acid(4-PBA),an aromatic fatty acid,was reported to benefit homeostasis of endoplasmic reticulum and protein synthesis.However,whether 4-PBA affects intestinal epithelial barrier function in pigs is unknown.This study aimed to explore the effects of 4-PBA on the intestinal barrier function,using in vitro models of well-differentiated intestinal porcine epithelial cell(IPEC-J2)monolayers in the transwell plates.Cell monolayers with or without 4-PBA(1.0 mmol/L)treatment were challenged with physical scratch,deoxynivalenol(DON,2.0μg/mL,48 h),and lipopolysaccharide(LPS,5.0μg/mL,48 h),respectively.Transepithelial electrical resistance(TEER)and fluorescein isothiocyanate-dextran(FD-4)permeability were measured to indicate barrier integrity and permeability.Real-time PCR and Western blot were conducted to determine relative gene and protein expressions of tight junction proteins.As expected,physical scratch,DON,and LPS challenges decreased TEER and increased FD-4 permeability.4-PBA treatment accelerated cell mitigation and rehabilitation of the physical scratch-damaged intestinal epithelial barrier but did not alleviate DON or LPS induced barrier damage.However,once 48-h DON and LPS challenges were removed,rehabilitation of the epithelial barrier function of IPEC-J2 monolayer was accelerated by the 4-PBA treatment.Also,the relative gene and protein expressions of zonula occludens-1(ZO-1),occludin,and claudin-1 were further upregulated by the 4-PBA treatment during the barrier rehabilitation.Taken together,4-PBA accelerated the IPEC-J2 cell monolayer barrier recovering from physical scratch,DON-,and LPS-induced damage,via enhancing cell mitigation and expressions of tight junction proteins. | Qian Jiang Jie Yin Jiashun Chen Xiaokang Ma Miaomiao Wu Xilong Li Kang Yao Bi'e Tan Yulong Yin | 2021 | Animal Nutrition2021,,4: | 2 |

| 7 | Targeting m6A modification inhibits herpes virus 1 infection显示文摘The latent infection by herpes virus type 1(HSV-1)may be lifelong in trigeminal ganglia and a suspected cause of Alzheimer's Disease(AD)and Amyotrophic lateral sclerosis(ALS).Whether and how N6-methyladenosine(m6A)modification of viral RNAs affects virus infection are poorly understood.Here,we report that HSV-1 infection enhanced the expression of m6A writers(METTL3,METTL14)and readers(YTHDF1/2/3)at the early infection stage and decreased their expression later on,while suppressed the erasers'(FTO,ALBKH5)expression immediately upon infection to facilitate viral replication.Inhibiting m6A modification by 3-deazaadenosine(DAA)significantly decreased viral replication and reduced viral reproduction over 1000 folds.More interestingly,depleting the writers and readers by siRNAs inhibited virus replication and reproduction;whereas depleting the erasers promoted viral replication and reproduction.Silencing YTHDF3 strikingly decreased viral replication by up to 90%,leading to reduction of up to 10-fold viral replication and over 100-fold virus reproduction,respectively.Depletion of m6A initiator METTL3(by 60%–70%)by siRNA correlatedly decreased viral replication 60%–70%,and reduced virus yield over 30-fold.Consistently,ectopic expression of METTL3 largely increased virus yield.METTL3 knockdown suppressed the HSV-1 intermediate early and early genes(ICP0,ICP8 and UL23)and late genes(VP16,UL44,UL49 and ICP47);while ectopic expression of METTL3 upregulated these gene expression.Results from our study shed the lights on the importance for m6A modification to initiate HSV-1 early replication.The components of m6A modification machinery,particularly m6A initiator METTL3 and reader YTHDF3,would be potential important targets for combating HSV-1 infections. | Zhuoying Feng Fanghang Zhou Miaomiao Tan Tingting Wang Ying Chen Wenwen Xu Bin Li Xin Wang Xin Deng Ming-Liang He | 2022 | Genes & Diseases2022,9,4: | 1 |

| 9 | Development of a novel conductance-based technology for environmental bacterial sensing显示文摘In this study, a simple impedance based technology for measuring bacterial concentrations was developed. The measurement system includes the signal amplification, copper probes and a sample loader. During the experiments, the conductance of Bacillus subtilis var niger, Pseudomonas fluorescens, and Escherichia coli were measured using the combination of a pre-amplifier and a lock-in amplifier. The conductance data were modeled verses the bacterial concentrations. Results indicated that the relationship between the conductance of bacterial suspensions and their concentrations follows a generic model: Y=C1+C2×e ( X/C3 ) , where Y is the conductance (S), X is the bacterial concentration (Number/mL: abbreviated to N/mL) for all species tested, and C1 3 are constants. Gram negative P. fluorescens and E. coli assumed similar conductance curves, which were flatter than that of gram positive B. subtilis var niger. For P. fluorescens and E. coli the culturing technique resulted in higher concentration levels (statistically significant) from 2 to 4 times that measured by the impedance based technology. For B. subtilis var niger, both methods resulted in similar concentration levels. These differences might be due to membrane types, initial culturability and the obtained conductance curves. | 13 | Establishment of organoid models for pancreatic ductal adenocarcinoma and screening of individualized therapy strategy显示文摘Background:Patients with pancreatic ductal adenocarcinoma(PDAC)who undergo surgical resection and receive effective chemotherapy have the best chance for longterm survival.Unfortunately,because of the heterogeneity of pancreatic cancer,it is difficult to find a personalized treatment strategy for patients.Organoids are ideal preclinical models for personalized medicine.Therefore,we explore the cultivation conditions and construction methods of PDAC organoid models to screen the individualized therapy strategy.Methods:Fresh PDAC tissues from surgical resection were collected and digested with digestive enzymes;then the tumor cells were embedded in Matrigel with a suitable medium to establish the PDAC organoid models.The genetic stability of the organoids was analyzed using whole exon sequencing;hematoxylin and eosin staining and immunohistochemistry of organoids were performed to analyze their consistency with the pathological morphology of the patient's tumor tissue;After 2 days of organoid culture,we selected four commonly used clinical chemotherapy drugs for single or combined treatment to analyze drug sensitivity.Results:Two cases of PDAC organoid models were successfully established,and the results of their pathological characteristics and exome sequencing were consistent with those of the patient's tumor tissue.Both PDAC organoids showed more sensitivity to gemcitabine and cisplatin,and the combined treatment was more effective than monotherapy.Conclusion:Both organoids better retained the pathological characteristics,genomic stability,and heterogeneity with the original tumor.Individual PDAC organoids exhibited different sensitivities to the same drugs.Thus,this study provided ideal experimental models for screening individualized therapy strategy for patients with PDAC. | Miaomiao Gong Han Meng Dengxu Tan Peng Li Jing Qin Qingling An Changhong Shi Jiaze An | 2023 | Animal Models and Experimental Medicine2023,6,5: | 0 |

| 15 | Optimizing feeding regimen of replacement gilts to improve their reproductive performance and retention rate of their first 2 parities显示文摘The effects of age,body weight(BW),and backfat thickness(BF)of replacement gilts at first estrus and first mating on their subsequent reproductive performance and retention of their first 2 parities as sows was evaluated.A total of 3,025 Danish replacement gilts were categorized by farm(allocated to 4 farms),cross combination,age,BW and BF at first estrus and first mating,estrous cycle number at first mating,and flush feeding before first mating.The result shows that all the factors mentioned above were significantly associated with reproductive performance and retention rates of the first 2 parities.Farm 3 had more piglets born alive per litter(BA)(P<0.05).Farms 3 and 4 had more healthy piglets per litter(HP)(P<0.05).Farm 4 had the most piglets weaned per litter(PW)(P<0.05).Landrace×Landrace×Yorkshire(L×L×Y)replacement gilts had the most total piglets born per litter(TB),BA,HP,PW and a higher retention rate of the 2 parities than Landrace×Yorkshire(L×Y)replacement gilts(P<0.05).In addition,flush feeding before first mating had the most TB,BA,HP,PW,and a higher retention rate of the 2 parities than no flush feeding(P<0.05).Because the effects of replacement gilts rearing parameters on reproductive performance traits differed,we used 100 replacement gilts as a unit and the total number of weaned piglets from the first 2 parities as a new index.Replacement gilts undergoing their first estrus between 180 and 210 d of age at 115 to 124.9 kg BW and 14 to 15 mm BF had significantly higher reproductive indexes for their first 2 parities per 100 replacement gilts.Replacement gilts that mated between 210 and 230 d of age at 140 to 149.9 kg BW and 15 to 16 mm BF had optimal reproductive indexes.These results provide a new insight into the complex relationships among these reproductive performance traits and may help guide successful management of replacement gilts as a pivotal starting point for future fertility and longevity of rearing herds. | Jiajian Tan Miaomiao Wang Haiqing Sun Chao Wang Hongkui Wei Siwen Jiang Yuanfei Zhou Jian Peng | 2023 | Animal Nutrition2023,,1: | 0 |
