16分级脂肪抽吸物:是否存在用于脂肪移植的最佳密度显示文摘背景与目的脂肪移植的临床效果是不可预知的。本文中,我们假设离心可以产生“梯度等级密度”的具有不同特性的脂肪,可以影响脂肪抽吸物的持久性和质量。方法将人类女性脂肪抽吸物样本(10mI)以1200g速率离心3rain。弃血液和油脂成分。随后,分离出1.0ml密度最高与密度最低的脂肪,进行脂肪浸润处理及分析。将密度最高与密度最低的脂肪移植于人体FVB小鼠,然后分别于2周和10周获取组织样本,进行短期与长期持久性评价。分别通过流式细胞术和酶联免疫吸附实验,分析祖细胞数量以及血管内皮细胞生长因子(vascularendothelialgrowthfactor,VEGF)、基质细胞来源因子.1(stromalcell—derivedfactor-1,SDF-1)、血小板来源生长因子(platelet—derivedgrowthfactor,PDGF)和脂连蛋白的表达。结果高密度脂肪与低密度脂肪相比,注射后2周[(85.4%±1.9%):(62.3±0.1%),P=0.05]和10周[(60.8%±4.9%):(42.2%±3.9%),P〈0.05]均表现出更高的存活百分率。而且,在高密度的脂肪抽吸物细胞群中,VEGF、基质血管成分(stromalvascularfraction,SVF)、PDGF和脂连蛋白的含量均比低密度的高(分别高出34.4%,P〈0.01;34.6,P〈0.05;52.2%,P〈0.01;45.7%,P〈0.05)。结论与低密度脂肪细胞群相比,随时间增长,高密度脂肪抽吸物细胞群可保持更高的存活百分比。因为高密度脂肪抽吸物细胞群包含更多的祖细胞和更大浓度的多种血管形成介质,所以比低密度脂肪细胞群更有助于血管形成。Robert J.
