1A dual-pass data assimilation scheme for estimating surface fluxes with FY3A-VIRR land surface temperature显示文摘In this work, a dual-pass data assimilation scheme is developed to improve predictions of surface flux. Pass 1 of the dual-pass data assimilation scheme optimizes the model vegetation parameters at the weekly temporal scale, and Pass 2 optimizes the soil moisture at the daily temporal scale. Based on ensemble Kalman filter(EnKF), the land surface temperature(LST) data derived from the new generation of Chinese meteorology satellite(FY3A-VIRR) are assimilated into common land model(CoLM) for the first time. Six sites, Daman, Guantao, Arou, BJ, Miyun and Jiyuan, are selected for the data assimilation experiments and include different climatological conditions. The results are compared with those from a dataset generated by a multi-scale surface flux observation system that includes an automatic weather station(AWS), eddy covariance(EC) and large aperture scintillometer(LAS). 2Regional and Global Land Data Assimilation Systems: Innovations,Challenges, and Prospects显示文摘Since the North American and Global Land Data Assimilation Systems(NLDAS and GLDAS) were established in2004, significant progress has been made in development of regional and global LDASs. National, regional, projectbased, and global LDASs are widely developed across the world. This paper summarizes and overviews the development, current status, applications, challenges, and future prospects of these LDASs. We first introduce various regional and global LDASs including their development history and innovations, and then discuss the evaluation, validation, and applications(from numerical model prediction to water resources management) of these LDASs. More importantly, we document in detail some specific challenges that the LDASs are facing: quality of the in-situ observations, satellite retrievals, reanalysis data, surface meteorological forcing data, and soil and vegetation databases; land surface model physical process treatment and parameter calibration; land data assimilation difficulties; and spatial scale incompatibility problems. 3Efficient hepatic delivery and protein expression enabled by optimized mRNA and ionizable lipid nanoparticle显示文摘mRNA is a novel class of therapeutic modality that holds great promise in vaccination,protein replacement therapy,cancer immunotherapy,immune cell engineering etc.However,optimization of mRNA molecules and efficient in vivo delivery are quite important but challenging for its broad application.Here we present an ionizable lipid nanoparticle(iLNP)based on iBL0713 lipid for in vitro and in vivo expression of desired proteins using codon-optimized mRNAs.mRNAs encoding luciferase or erythropoietin(EPO)were prepared by in vitro transcription and formulated with proposed iLNP,to form iLP171/mRNA formulations.It was revealed that both luciferase and EPO proteins were successfully expressed by human hepatocellular carcinoma cells and hepatocytes.The maximum amount of protein expression was found at 6 h post-administration.The expression efficiency of EPO with codon-optimized mRNA was significantly higher than that of unoptimized mRNA.Moreover,no toxicity or immunogenicity was observed for these mRNA formulations.Therefore,our study provides a useful and promising platform for mRNA therapeutic development.Tongren Yang Chunhui Li Xiaoxia Wang Deyao Zhao Mengjie Zhang Huiqing Cao Zicai Liang Haihua Xiao Xing-Jie Liang Yuhua Weng Yuanyu Huang 2020Bioactive Materials2020,5,4:7

5Changes in Global Cloud Cover Based on Remote Sensing Data from 2003 to 2012显示文摘As is well known,clouds impact the radiative budget,climate change,hydrological processes,and the global carbon,nitrogen and sulfur cycles.To understand the wide-ranging effects of clouds,it is necessary to assess changes in cloud cover at high spatial and temporal resolution.In this study,we calculate global cloud cover during the day and at night using cloud products estimated from Moderate Resolution Imaging Spectroradiometer(MODIS)data.Results indicate that the global mean cloud cover from 2003 to 2012 was 66%.Moreover,global cloud cover increased over this recent decade.Specifically,cloud cover over land areas(especially North America,Antarctica,and Europe)decreased(slope=–0.001,R^2=0.5254),whereas cloud cover over ocean areas(especially the Indian and Pacific Oceans)increased(slope=0.0011,R^2=0.4955).Cloud cover is relatively high between the latitudes of 36°S and 68°S compared to other regions,and cloud cover is lowest over Oceania and Antarctica.The highest rates of increase occurred over Southeast Asia and Oceania,whereas the highest rates of decrease occurred over Antarctica and North America.The global distribution of cloud cover regulates global temperature change,and the trends of these two variables over the 10-year period examined in this study(2003–2012)oppose one another in some regions.These findings are very important for studies of global climate change.MAO Kebiao YUAN Zijin ZUO Zhiyuan XU Tongren SHEN Xinyi GAO Chunyu 2019Chinese Geographical Science2019,29,2:5
6Anomalous pancreaticobiliary junction: image analysis and treatment principles显示文摘BACKGROUND: Anomalous pancreaticobiliary junction is often associated with biliary tract carcinoma and acute pan- creatitis. We assessed the value of image analysis in the diag- nosis of patients with anomalous pancreaticobiliary junction (APBJ) and the principles for the treatment of APBJ. METHODS: Sixty-four patients with APBJ were subjected to ultrasound imaging, endoscopic retrograde cholangio- pancreatography (ERCP) and magnetic resonance cholan- giopancreatography (MRCP) before surgery. The diagnos- tic accuracy of image analysis and their surgical outcomes were evaluated retrospectively. RESULTS: On ERCP and MRCP, the length of the com- mon channel was calculated to be 15 mm or longer in all patients, and the angle of the junction was more than 75° in 49 (76.6%) of the 64 patients. Of the 64 patients, 28 were defined of pancreatic duct type (P-C) (28/64, 43.75%), 32 bile duct type (C-P) (32/64, 50%), and 4 common chan- nel type (4/64, 6.25%). CONCLUSIONS: Patients with APBJ are often associated with biliary tract and pancreatic diseases, and early detec- tion and correct surgical treatment could avoid serious complications. 7Protective effect of liver ischemic preconditioning on rat hepatocytes显示文摘Ischemic preconditioning (IPC) protects liver graft function following ischemia in liver transplantation and liver resection. The aim of this study was to assess the advantages and any potential disadvantages of liver IPC prior to isolation for rat hepatocytes during isolation and cryopreservation. After isolating and thawing the cryopreserved hepatocytes after 14 and 28 d,cell viability,efficiency,and lactate dehydrogenase (LDH) levels in preserve solution were examined for every group. 8Retrieval of Land-surface Temperature from AMSR2 Data Using a Deep Dynamic Learning Neural Network显示文摘It is more difficult to retrieve land surface temperature(LST) from passive microwave remote sensing data than from thermal remote sensing data, because the emissivities in the passive microwave band can change more easily than those in the thermal infrared band. Thus, it is very difficult to build a stable relationship. Passive microwave band emissivities are greatly influenced by the soil moisture, which varies with time. This makes it difficult to develop a general physical algorithm. This paper proposes a method to utilize multiple-satellite, sensors and resolution coupled with a deep dynamic learning neural network to retrieve the land surface temperature from images acquired by the Advanced Microwave Scanning Radiometer 2(AMSR2), a sensor that is similar to the Advanced Microwave Scanning Radiometer Earth Observing System(AMSR-E). The AMSR-E and MODIS sensors are located aboard the Aqua satellite. The MODIS LST product is used as the ground truth data to overcome the difficulties in obtaining large scale land surface temperature data. The mean and standard deviation of the retrieval error are approximately 1.4° and 1.9° when five frequencies(ten channels, 10.7, 18.7, 23.8, 36.5, 89 V/H GHz) are used. This method can effectively eliminate the influences of the soil moisture, roughness, atmosphere and various other factors. An analysis of the application of this method to the retrieval of land surface temperature from AMSR2 data indicates that the method is feasible. 9Ionizable lipid-assisted efficient hepatic delivery of gene editing elements for oncotherapy显示文摘CRISPR/Cas9-based gene editing has emerged as a powerful biotechnological tool,that relies on Cas9 protein and single guided RNA(sgRNA)to edit target DNA.However,the lack of safe and efficient delivery carrier is one of the crucial factors restricting its clinical transformation.Here,we report an ionizable lipid nanoparticle(iLP181,pKa=6.43)based on iLY1809 lipid enabling robust gene editing in vitro and in vivo.The iLP181 effectively encapsulate psgPLK1,the best-performing plasmid expressing for both Cas9 protein and sgRNA targeting Polo-like kinase 1(PLK1).The iLP181/psgPLK1 nanoformulation showed uniformity in size,regular nanostructure and nearly neutral zeta potential at pH 7.4.The nanoformulation effectively triggered editing of PLK1 gene with more than 30%efficiency in HepG2-Luc cells.iLP181/psgPLK1 significantly accumulated in the tumor for more than 5 days after a single intravenous injection.In addition,it also achieved excellent tumor growth suppression compared to other nucleic acid modalities such as siRNA,without inducing adverse effects to the main organs including the liver and kidneys.This study not only provides a clinically-applicable lipid nanocarrier for delivering CRISPR/Cas system(even other bioactive molecules),but also constitutes a potential cancer treatment regimen base on DNA editing of oncogenes.Chunhui Li Tongren Yang Yuhua Weng Mengjie Zhang Deyao Zhao Shuai Guo Bo Hu Wanxuan Shao Xiaoxia Wang Abid Hussain Xing-Jie Liang Yuanyu Huang 2022Bioactive Materials2022,7,3:2
10Gradual upregulation of OCI-5 expression during occurrence and progression of rat hepatocellular carcinoma显示文摘BACKGROUND: OCI-5, the rat homologene of human glypican 3 ( GPC3), is confirmed upregulated in hepatocellular carcinoma (HCC). The present study was undertaken to detect gene expression change of OCI-5 during occurrence and progression of rat HCC. METHODS: Male Sprague-Dawley rats were given diethyl-nitrosamine ( DENA) to induce HCC. Three DENA-induced rats and one control rat were sacrificed every week for 18 weeks during the development of HCC. Tissues specimens were snap-frozen in liquid nitrogen and total RNA was isolated. Sk-Hepl cells were treated with DENA at different concentrations. The gene expression levels of OCI-5 and GPC3 were detected with the RT-PCR method. RESULTS: OCI-5 was not expressed in normal rat liver tissues. When HCC occurred and aggravated, OCI-5 expression was gradually elevated to a very high level. GPC3 was not expressed in the DENA-treated Sk-Hepl cells. CONCLUSIONS: OCI-5 was not expressed in normal rat liver tissues but in rat HCC tissues. High-expression of OCI-5 in DENA-induced rat HCC model was the gene expression change of HCC not the DENA-induced gene expression. 20Retinoblastoma in a young adult mimicking Coats'disease显示文摘Retinoblastoma is the most common childhood primary intraocular malignancy, with the majority of cases being diagnosed before 5 years of age. Retinoblastoma in adults is extremely rare. Here, we report the case of a 20-year-old man who presented with a 3 year history of blurred vision in the right eye. Imaging did not reveal the typical presentation of retinoblastoma. After considering Coats' disease, a diagnosis of late-presenting retinoblastoma was made through cytological analysis.
