1Sporopollen Assemblages from the Cretaceous Yimin Formation of the Hailar Basin,Inner Mongolia,China显示文摘Three sporopollen assemblages are recognized for the first time from the Cretaceous Yimin Formation in the Hailar Basin of eastern Inner Mongolia. They are (in ascending order): the Impardecispora-Aequitriradites-Clavatipollenites assemblage; the Triporoletes-Pilosisporites-Asteropollis assemblage; and the Appendicisporites-Asteropollis-Tricolpites assemblage, distributed in Members 1, 2 and 3 of the Yimin Formation respectively. Recognition of this biostratigraphic sequence is very important for the division and correlation of the Yimin Formation in the basin. 2Predicting the Dynamic Behavior of Asphalt Concrete Using Three-dimensional Discrete Element Method显示文摘A user-defined three-dimensional(3D)discrete element model was presented to predict the dynamic modulus and phase angle of asphalt concrete(AC).The 3D discrete element method(DEM)model of AC was constructed employing a user-defined computer program developed using the 'Fish' language in PFC3D.Important microstructural features of AC were modeled,including aggregate gradation,air voids and mastic.The irregular shape of aggregate particle was modeled using a clump of spheres.The developed model was validated through comparing with experimental measurements and then used to simulate the cyclic uniaxial compression test,based on which the dynamic modulus and phase angle were calculated from the output stress-strain relationship.The effects of air void content,aggregate stiffness and volumetric fraction on AC modulus were further investigated.The experimental results show that the 3D DEM model is able to accurately predict both dynamic modulus and phase angle of AC across a range of temperature and loading frequencies.The user-defined 3D model also demonstrated significant improvement over the general existing two-dimensional models.陈俊 PAN Tongyan CHEN Jingya HUANG Xiaoming LU Yang 2012Journal of Wuhan University of Technology(Materials Science)2012,27,2:4
3Meiotic nuclear divisions 1(MND1)fuels cell cycle progression by activating a KLF6/E2F1 positive feedback loop in lung adenocarcinoma显示文摘Background:Considering the increase in the proportion of lung adenocarcinoma(LUAD)cases among all lung cancers and its considerable contribution to cancer-related deaths worldwide,we sought to identify novel oncogenes to provide potential targets and facilitate a better understanding of the malignant progression of LUAD.Methods:The results from the screening of transcriptome and survival analyses according to the integrated Gene Expression Omnibus(GEO)datasets and The Cancer Genome Atlas(TCGA)data were combined,and a promising risk biomarker called meiotic nuclear divisions 1(MND1)was selectively acquired.Cell viability assays and subcutaneous xenograftmodelswere used to validate the oncogenic role ofMND1 in LUADcell proliferation and tumor growth.Aseries of assays,including mass spectrometry,co-immunoprecipitation(Co-IP),and chromatin immunoprecipitation(ChIP),were performed to explore the underlying mechanism.Results:MND1 up-regulation was identified to be an independent risk factor for overall survival in LUAD patients evaluated by both tissue microarray staining and third party data analysis.In vivo and in vitro assays showed that MND1 promoted LUAD cell proliferation by regulating cell cycle.The results of the Co-IP,ChIP and dual-luciferase reporter assays validated that MND1 competitively bound to tumor suppressor Kruppel-like factor 6(KLF6),and thereby protecting E2F transcription factor 1(E2F1)from KLF6-induced transcriptional repression.Luciferase reporter and ChIP assays found that E2F1 activated MND1 transcription by binding to its promoter in a feedback manner.Conclusions:MND1,KLF6,and E2F1 form a positive feedback loop to regulate cell cycle and confer DDP resistance in LUAD.MND1 is crucial for malignant progression and may be a potential therapeutic target in LUAD patients.Quanli Zhang Run Shi Yongkang Bai Lijuan Meng Jingwen Hu Hongyu Zhu Tongyan Liu Xiaomeng De Siwei Wang Jie Wang Lin Xu Guoren Zhou Rong Yin 2021Cancer Communications2021,41,6:2
4Research and development of a novel subunit vaccine for the currently circulating pseudorabies virus variant in China显示文摘Pseudorabies(PR)is a devastating viral disease which leads to fatal encephalitis and respiratory disorders in pigs.Commercial gE-deleted live pseudorabies virus(PRV)vaccine has been widely used to control this disease in China.However,the new-emerging variants of PRV compromises the protection provided by current vaccines and lead to the outbreak of PR in vaccinated pig herds.Several killed and live vaccine candidates based on current PRV variants have been reported to be effective to control the disease.A subunit vaccine based on gB protein,one major PRV glycoprotein which elicits strong humoral and cellular immune responses,however,was never evaluated for protection against the current circulating PRV variants.In this study,full-length PRV gB protein was successfully expressed in baculovirus/insect cells in the soluble format and was tested on 3-week-old piglets as a subunit vaccine.Compared with unvaccinated pigs,the gB-vaccinated pigs developed specific antibody-mediated responses and were protected from the virulent PRV HN1201 challenge.All vaccinated pigs survived without showing any PRV-specific respiratory and neurological signs,but all unvaccinated pigs died within 7 days after HN1201 challenge.Hence,this novel gB-based vaccine could be applied as an effective subunit vaccine to control PRV variant in China.Yuzhou WANG Tongyan WANG He YAN Fanli YANG Linghua GUO Qingyuan YANG Xule HU Feifei TAN Yan XIAO Xiangdong LI Kegong TIAN 2015Frontiers of Agricultural Science and Engineering2015,2,3:2
5LncRNA LINC00525 suppresses p21 expression viamRNA decay and triplex-mediated changes in chromatin structure in lung adenocarcinoma显示文摘Background:Emerging evidence suggests that long noncoding RNAs(lncRNAs)play crucial roles in various cancers.In the present study,we aim to investigate the function and molecular mechanism of an up-regulated and survivalassociated lncRNA,LINC00525,in lung adenocarcinoma(LUAD).Methods:The expression level of LINC00525 in tissueswas determined by quantitative reverse transcription polymerase chain reaction(RT-qPCR)and in situ hybridization(ISH).The functional role of LINC00525 in LUAD was investigated using gain-and loss-of-function approaches,both in vivo and in vitro.RNA pull-down,RNA immunoprecipitation(RIP),chromatin immunoprecipitation(ChIP),triplex-capture assay,dual-luciferase assay,gene expression microarray,and bioinformatics analysis were used to investigate the potential underlying mechanisms involved.Results:LINC00525 is highly expressed in LUAD cells and tissues.Survival analysis indicated that upregulation of LINC00525 was associated with poor prognosis in patients with LUAD patients.Knockdown of LINC00525 inhibited cell proliferation and cell cycle progression in vitro.In xenograft models,LINC00525 knockdown suppressed tumor growth and tumorigenesis of tumorbearing mice.Mechanistically,LINC00525 epigenetically suppressed p21 transcription by guiding Enhancer Of Zeste 2 Polycomb Repressive Complex 2 Subunit(EZH2)to the p21 promoter through an formation of RNA-DNA triplex with the p21 promoter,leading to increased trimethylation of lysine 27 on histone 3(H3K27me3)of the p21 promoter.In addition,LINC00525 repressed p21 expression post-transcriptionally by enhancing p21mRNA decay.LINC00525 promoted p21mRNAdecay by competitively binding toRNABindingMotif Single Stranded Interacting Protein 2(RBMS2).Conclusion:Our findings demonstrate that LINC00525 promotes the progression of LUAD by reducing the transcription and stability of p21 mRNA in concert with EZH2 and RBMS2,thus suggesting that LINC00525 may be a potential therapeutic target for clinical intervention in LUAD.Panqi Fang Hao Chen Zhifei Ma Chencheng Han Wenda Yin Siwei Wang Hongyu Zhu Wenjia Xia Jie Wang Lin Xu Tongyan Liu Rong Yin 2021Cancer Communications2021,41,7:2
