1Anti-bacterial activity and brine shrimp lethality bioassay of methanolic extracts of fourteen different edible vegetables from Bangladesh显示文摘Objective:To investigate the antibacterial and cytotoxic activity of fourteen different edible vegetables methanolic extract from Bangladesh.Methods:The antibacterial activity was evaluated using disc diffusion assay method against 12 bacteria(both gram positive and gram negative).The plant extracts were also screened for cytotoxic activity using the brine shrimp lethality bioassay method and the lethal concentrations(LC_(50))were determined at confidence intervals by analyzing the data on a computer loaded with'Finney Programme??Results:All the vegetable extracts showed low to elevated levels of antibacterial activity against most of the tested strains(zone of inhibition=5-28 mm).The most active extract against all bacterial strains was from Xanthium indicum which showed remarkable antibacterial activity having the diameter of growth inhibition zone ranging from 12 to 28 mm followed by Alternanthera sessilis(zone of inhibition=6-21 mm).All extracts exhibited considerable general toxicity towards brine shrimps.The LC_(50)value of the tested extracts was within the range of 8.447 to 60.323μg/mL with respect to the positive control(vincristine sulphate)which was 0.91μg/mL.Among all studied extracts,Xanthium indicum displayed the highest cytotoxic effect with LC_(50)value of 8.447μg/mL.Conclusions:The results of the present investigation suggest that most of the studied plants are potentially good source of antibacterial and anticancer agents.M.Obayed Ullah Mahmuda Haque Kaniz Fatima Urmi Abu Hasanat Md.Zulfiker Elichea Synthi Anita Momtaj Begum Kaiser Hamid 2013Asian Pacific Journal of Tropical Biomedicine2013,3,1:5

5Cloning,expression,purification and bioinformatic analysis of 2-methylcitrate synthase from Mycobacterium tuberculosis显示文摘Objective:To clone,express and purify 2—methylcitrale synthase(Rv1131) gene of Mycobacterium tuberculosis(M.tuberculosis) and to study its structural characteristics using various bioinformatics tools.Methods:Rv1131 gene was amplified In polymerase chain reaction using M.tuberculosis H37 Rv genomic DNA and cloned into pGEM-T easy vector and sequenced.The gene was sub-cloned in pET28 c vector,expressed in Escherichia coli RL22(E.coli BL21)(DE3) cells and the recombinant protein was identified by Western blotting.The protein was purified using Nickel affinity chromatography and the structural characteristics like sub-cellular localization.presence of transmembrane helices and secondary structure of the protein were predicted by bioinformaties tools.Tertiary structure of the protein and phylogenetic analysis was aiso established by in silico analysis.Results:The expression of the recombinant protein(Rv1131) was confirmed by western blotting using anti-HIS antibodies and the protein was purified from the soluble fraction.In silico analysis showed that the protein contains no signal peptide and transmembrane helices,Active site prediction showed that the protein has histidine and aspartie acid residues at 242.281 & 332 positions respectively.Phylogenetic analysis showed100%homology with major mycobacterial species.Secondary structure predicts 2-methyleitrate synthase contain 51.9%;alpha-helk.8.7%extended strand and 39.4%random coils.Tertiary structure of the protein was also established.Conclusions:The enzyme 2-methylcitrate synthase from M.tuberculosis H37 Rv has been successfully expressed and purified.The purified protein will further be utilized to develop assay methods lor screening new inhibitors.Kasamy Eniyan Urmi Bajpai 2015Asian Pacific Journal of Tropical Medicine2015,8,1:1

18Transcriptional co-activators: emerging roles in signaling pathways and potential therapeutic targets for diseases显示文摘Specific cell states in metazoans are established by the symphony of gene expression programs that necessitate intricate synergic interactions between transcription factors and the co-activators.Deregulation of these regulatory molecules is associated with cell state transitions,which in turn is accountable for diverse maladies,including developmental disorders,metabolic disorders,and most significantly,cancer.A decade back most transcription factors,the key enablers of disease development,were historically viewed as‘undruggable’;however,in the intervening years,a wealth of literature validated that they can be targeted indirectly through transcriptional co-activators,their confederates in various physiological and molecular processes.These co-activators,along with transcription factors,have the ability to initiate and modulate transcription of diverse genes necessary for normal physiological functions,whereby,deregulation of such interactions may foster tissue-specific disease phenotype.Hence,it is essential to analyze how these co-activators modulate specific multilateral processes in coordination with other factors.The proposed review attempts to elaborate an in-depth account of the transcription co-activators,their involvement in transcription regulation,and context-specific contributions to pathophysiological conditions.This review also addresses an issue that has not been dealt with in a comprehensive manner and hopes to direct attention towards future research that will encompass patient-friendly therapeutic strategies,where drugs targeting co-activators will have enhanced benefits and reduced side effects.Additional insights into currently available therapeutic interventions and the associated constraints will eventually reveal multitudes of advanced therapeutic targets aiming for disease amelioration and good patient prognosis.Priyanka Dey Talukdar Urmi Chatterji 2023Signal Transduction and Targeted Therapy2023,8,12:0
