4A deep learning-enabled portable imaging flow cytometer for cost-effective, highthroughput, and label-free analysis of natural water samples显示文摘We report a deep learning-enabled field-portable and cost-effective imaging flow cytometer that automatically captures phase-contrast color images of the contents of a continuously flowing water sample at a throughput of 100 mL/h.The device is based on partially coherent lens-free holographic microscopy and acquires the diffraction patterns of flowing micro-objects inside a microfluidic channel.These holographic diffraction patterns are reconstructed in real time using a deep learning-based phase-recovery and image-reconstruction method to produce a color image of each micro-object without the use of external labeling.Motion blur is eliminated by simultaneously illuminating the sample with red,green,and blue light-emitting diodes that are pulsed.Operated by a laptop computer,this portable device measures 15.5 cm×15 cm×12.5 cm,weighs 1 kg,and compared to standard imaging flow cytometers,it provides extreme reductions of cost,size and weight while also providing a high volumetric throughput over a large object size range.We demonstrated the capabilities of this device by measuring ocean samples at the Los Angeles coastline and obtaining images of its micro-and nanoplankton composition.Furthermore,we measured the concentration of a potentially toxic alga(Pseudo-nitzschia)in six public beaches in Los Angeles and achieved good agreement with measurements conducted by the California Department of Public Health.The cost-effectiveness,compactness,and simplicity of this computational platform might lead to the creation of a network of imaging flow cytometers for largescale and continuous monitoring of the ocean microbiome,including its plankton composition.Zoltán Gӧrӧcs Miu Tamamitsu Vittorio Bianco Patrick Wolf Shounak Roy Koyoshi Shindo Kyrollos Yanny Yichen Wu Hatice Ceylan Koydemir Yair Rivenson Aydogan Ozcan 2018Light(Science & Applications)2018,7,1:14

6Effect of human autologous serum and fetal bovine serum on human corneal epithelial cell viability,migration and proliferation in vitro显示文摘AIM: To analyze the concentration-dependent effects of autologous serum(AS) and fetal bovine serum(FBS) on human corneal epithelial cell(HCEC) viability, migration and proliferation.METHODS: AS was prepared from 13 patients with nonhealing epithelial defects Dulbecco's modified eagle medium/Ham's F12(DMEM/F12) with 5% FBS, 0.5% dimethyl sulphoxide(DMSO), 10 ng/m L human epidermal growth factor, 1% insulin-transferrin-selenium, then were incubated in serum media: DMEM/F12 supplemented by 5%, 10%, 15% or 30% AS or FBS. HCEC viability was analyzed using cell proliferation kit XTT, migration using a wound healing assay, proliferation by the cell proliferation enzyme-linked immunosorbent assay(ELISA) Brd U kit. Statistical analysis was performed using the generalized linear model, the values at 30% AS or 30% FBS were used as the baselines.RESULTS: HCEC viability was the highest at 30% AS or 15% FBS and the lowest at 10% AS or 30% FBS application. HCEC migration was the quickest through 30% AS or 30% FBS and the slowest through 5% AS or 5% FBS concentrations. Proliferation was the most increased through 15% AS or 5% FBS and the least increased through 30% AS or 30% FBS concentrations. HCEC viability at 10% and 15% AS was significantly worse(P=0.001, P=0.023) compared to baseline and significantly better at 15% FBS(P=0.003) concentrations. HCEC migration was significantly worse(P≤0.007) and HCEC proliferation significantly better(P<0.001) in all concentration groups compared to baseline.CONCLUSION: For the best viability of HCEC 30% AS or 15% FBS, for HCEC migration 30% AS or 30% FBS, for proliferation 15% AS or 5% FBS should be used. 8Neuroprotective effects of INT-777 against Aβ1-42-induced cognitive impairment, neuroinflammation, apoptosis, and synaptic dysfunction in mice显示文摘OBJECTIVE To investigate the neuroprotective effects of TGR5 agonist INT-777 in the Aβ_(1-42)-treated mouse model of acute neurotoxicity.METHODS Cognitive impairment was induced by single intracerebroventricular injection of aggregated Aβ_(1-42)(410 pmol per mouse;5 μL)into the mouse brain in normal ICR mice.After3 d,INT-777(0.75,1.5 or 3.0 μg per mouse) was infused into the same site.Another 3 d later,the cognition function was evaluated by Morris water maze(MWM),novel objective recognition(NOR),and Y maze tests.Furthermore,the levels of TGR5,TNF-α,IL-1β,IL-6,NF-κB p65,Iba1,caspase 3,Bcl-2,Bax,PSD95,and synaptophysin in the hippocampus and frontal cortex were detected using Western blotting.Microglia activation in the hippocampal CA1,CA3 and DG regions and frontal cortex were detected by immunohistochemistry.TUNEL staining was used to detect the apoptotic cells.The number of spines in the hippocampal CA1,CA3 and DG regions and frontal cortex was detected by GolgiCox staining.RESULTS INT-777(1.5 or 3.0 μg per mouse) significantly ameliorated memory impairment in the Aβ_(1-42)-treated mice,evidenced by increased the time spent in the target quadrant(P<0.05),and the number of target crossings(P<0.05) in the MWM test,increased the discrimination index in the NOR test(P<0.05),and increased the number of correct choices(P<0.05,P<0.01) and decreased the latency(P<0.05) to enter the shock-free compartment in the Y-maze test.Importantly,this treatment reversed Aβ_(1-42)-induced TGR5 down-regulation(P<0.05,P<0.01),suppressed the increase of nuclear NF-κB p65(P<0.05,P<0.01) and mitigated neuroinflammation,evidenced by lower proinflammatory cytokines such as TNF-α(P<0.05),IL-1β(P<0.05),IL-6(P<0.05,P<0.01),and less Iba1-positive cells in the hippocampal CA1,CA3 and DG regions and frontal cortex(P<0.05,P<0.01) as well as the protein level of Iba1.INT-777(1.5 or3.0 μg per mouse) also pronouncedly suppressed apoptosis through the reduction of TUNEL-positive cells(P<0.05,P<0.01),decreased activation of caspase 3(P<0.05),increased ratio of Bcl-2/Bax(P<0.05,P<0.01),and ameliorated synaptic dysfunction via promoting dendritic spines generation(P<0.05,P<0.01) with the upregulation of PSD95(P<0.05) and synaptophysin proteins(P<0.05).CONCLUSION INT-777 has potent neuroprotective effects against Aβ_(1-42)-induced neurotoxicity,which suggests that the activation of TGR5 could be a novel and promising strategy for the treatment of AD.WU Xian LYU Yang-ge DU Yi-feng CHEN Fang Miranda N REED HU Mei Vishnu SUPPIRAMANIAM TANG Su-su HONG Hao 2018中国药理学与毒理学杂志2018,32,9:6
9Air quality monitoring using mobile microscopy and machine learning显示文摘Rapid,accurate and high-throughput sizing and quantification of particulate matter(PM)in air is crucial for monitoring and improving air quality.In fact,particles in air with a diameter of≤2.5μm have been classified as carcinogenic by the World Health Organization.Here we present a field-portable cost-effective platform for high-throughput quantification of particulate matter using computational lens-free microscopy and machine-learning.This platform,termed c-Air,is also integrated with a smartphone application for device control and display of results.This mobile device rapidly screens 6.5 L of air in 30 s and generates microscopic images of the aerosols in air.It provides statistics of the particle size and density distribution with a sizing accuracy of~93%.We tested this mobile platform by measuring the air quality at different indoor and outdoor environments and measurement times,and compared our results to those of an Environmental Protection Agency–approved device based on beta-attenuation monitoring,which showed strong correlation to c-Air measurements.Furthermore,we used c-Air to map the air quality around Los Angeles International Airport(LAX)over 24 h to confirm that the impact of LAX on increased PM concentration was present even at 47 km away from the airport,especially along the direction of landing flights.With its machinelearning-based computational microscopy interface,c-Air can be adaptively tailored to detect specific particles in air,for example,various types of pollen and mold and provide a cost-effective mobile solution for highly accurate and distributed sensing of air quality.Yi-Chen Wu Ashutosh Shiledar Yi-Cheng Li Jeffrey Wong Steve Feng Xuan Chen Christine Chen Kevin Jin Saba Janamian Zhe Yang Zachary Scott Ballard Zoltán Göröcs Alborz Feizi Aydogan Ozcan 2017Light(Science & Applications)2017,6,1:5

11Th1 cytokines promote T-cell binding to antigen-presenting cells via enhanced hyaluronan production and accumulation at the immune synapse显示文摘Hyaluronan(HA)production by dendritic cells(DCs)is known to promote antigen presentation and to augment T-cell activation and proliferation.We hypothesized that pericellular HA can function as intercellular‘glue’directly mediating T cell–DC binding.Using primary human cells,we observed HA-dependent binding between T cells and DCs,which was abrogated upon pre-treatment of the DCs with 4-methylumbelliferone(4-MU),an agent which blocks HA synthesis.Furthermore,T cells regulate HA production by DCs via T cell-derived cytokines in a T helper(Th)subset-specific manner,as demonstrated by the observation that cell-culture supernatants from Th1 but not Th2 clones promote HA production.Similar effects were seen upon the addition of exogenous Th1 cytokines,IL-2,interferon c(IFN-c)and tumor necrosis factor a(TNF-a).The critical factors which determined the extent of DC–T cell binding in this system were the nature of the pre-treatment the DCs received and their capacity to synthesize HA,as T-cell clones which were pre-treated with monensin,added to block cytokine secretion,bound equivalently irrespective of their Th subset.These data support the existence of a feedforward loop wherein T-cell cytokines influence DC production of HA,which in turn affects the extent of DC–T cell binding.We also document the presence of focal deposits of HA at the immune synapse between T-cells and APC and on dendritic processes thought to be important in antigen presentation.These data point to a pivotal role for HA in DC–T cell interactions at the IS.Paul L Bollyky Stephen P Evanko Rebecca P Wu Susan Potter-Perigo S Alice Long Brian Kinsella Helena Reijonen Kelly Guebtner Brandon Teng Christina K Chan Kathy R Braun John A Gebe Gerald T Nepom Thomas N Wight 2010Cellular & Molecular Immunology2010,7,3:3
