| 1 | Construct Hairpin RNA to Fight Against Rice Dwarf Virus显示文摘Hairpin RNA (hpRNA) can induce post-transcriptional gene silencing, and was constructed with128-754 bp of segment 8 of rice dwarf virus then placed under the control of the CaMV35S promoter andused to transform rice cultivar 'Zhonghua 11' via Agrobacterium tumefaciens. A total of 12 independentlines containing hpRNA were obtained as demonstrated by Southern blotting analysis. Challenge inocula-tion with rice dwarf virus (RDV) viruliferous leafhoppers (Nephotettix cincticeps) showed that T1 plantscontaining the hpRNA transgene displayed high resistance or delayed and attenuated viral symptoms. Incontrast, transgenic lines expressing sense RNA showed severe symptoms similar to control plants trans-formed with the vector alone. | 2 | Production of transgenic calves by somatic cell nuclear transfer显示文摘Bovine fetal oviduct epithelial cells were transfected with constructed double marker selective vector(pCE-EGFP-IRES-Neo-dNdB) containing the enhanced green fluorescent protein (EGFP) and neomycin-resistant(Neo^r) genes by electroporation, and a transgenic cell line was obtained. Somatic cell nuclear transfer (SCNT) was cartied out using the transgenic cells as nuclei donor. A total of 424 SCNT embryos were reconstructed and 208 (49.1%) of them developed to blastocyst stage. 17 blastocysts on D 7 after reconstruction were transferred to 17 surrogate calves,and 5 (29.4%) recipients were found to be pregnant. Three of them maintained to term and delivered three cloned calves.PCR and Southern blot analysis confirmed the integration of transgene in all of the three cloned calves. In addition, expression of EGFP was detected in biopsy isolated from the transgenic cloned calves and fibroblasts derived from the biopsy. Our results suggest that transgenic calves could be efficiently produced by SCNT using transgenic cells as nuclei donor. | 3 | Erosion Resistance Behaviours of H13 Steel to Molten ADC12 Alloy显示文摘H13 hot work steel samples and surface water vapor oxidized H13 samples were immersed into molten ADC 12 alloy under static resting or dynamic rotating conditions. Weigh-losing method was used to discover the influences of temperature, time and surface conditions on the erosion resistance of H13 steel. The interfaces between the steel and the molten alloy were studied with optical microscopy to compare the different erosion resistance behaviours. The results show that the composite layer outside of the compounds layers changes obviously with increasing temperature, lasting time or sample movement. | 7 | Analysis of Intel-metallic Phases Formed on Surface Vapor Oxidized H13 Hot Work Steels in Molten Aluminum显示文摘In this paper, the author dipped surface vapor oxidized H13 steel specimens into 700 °C molten aluminum liquid for a certain period of time. Analyze the intermetallic phases formed on the H13 samples surface with optical microscope and X-ray diffraction method. The observation of immersion test sample’s cross-section shows that Fe3O4 film will protect die substrate from molten aluminum erosion. The identification of the intermetallic phases reveals that they consist of 2 parts, which is named as the composite layer and the compact layer. Further investigations are made in order to know the phase constituents of the 2 layers, they are A18Fe2Si (outer composite layer), (AlCuMg) and A15Fe2 (compact layer), respectively. The experimental results show that on the same specimen, a convex surface with bigger radius of curvature is more likely to be molten and the melting loss speed is also faster than a flat and smooth surface. The thickness of compact layer on a smooth surface is much bigger than that of the convex surface. | 17 | Experimental study on spinal cord injury treated by embryonic spinal cord transplantation and greater omental transposition显示文摘Objective: To observe the clinical efficacy of the embryonic spinal cellular transplantation and greater omentaltranspesition for treatment of the spinal cord injury in 24 mongrel dogs. Methods: 24 adult mongrel dogs, weighing 10 ~ 13kg, without male and female limit, were employed in this experimental study and randomly divided into 4 groups:①simple injury group(group A) ;②greater omental transposition group (group B) ;③embryonic spinal cellular transplantation group (group C) ;④em-bryonic spinal cellular transplantation and greater omental transposition group (group D). Each group consisted of 6 dogs. SEP(somatosensory evoked potential) and MEP (motor evoked potential) of the spinal cord were examed prior to the spinal cord injuryand 2 months after the treatment to observe the changes of the animals' behavior. All dogs were killed 2 months after surgery andthe spinal cord sections were obtained from TI2 to LI level for pathological analysis and observation under the electron microscope.Re.lilts: There was an obvious difference in the spinal sematosensory evoked potential and the motor evoked potential between thegroup D.and the other three groups (group A, B, and C). Recovery of the behavior was noted. The spinal cells had survived fortwo months following the transplantation.
