A Systematic Characterization of Structural Brain Changes in Schizophrenia显示文摘A systematic characterization of the similarities and differences among different methods for detecting structural brain abnormalities in schizophrenia, such as voxel-based morphometry(VBM),tensor-based morphometry(TBM),and projection-based thickness(PBT),is important for understanding the brain pathology in schizophrenia and for developing effective biomarkers for a diagnosis of schizophrenia. However, such studies are still lacking. Here,we performed VBM,TBM,and PBT analyses on T1-weighted brain MR images acquired from 116 patients with schizophrenia and 116 healthy controls. We found that, although all methods detected wide-spread structural changes, different methods captured different information-only 10.35%of the grey matter changes in cortex were detected by all three methods, and VBM only detected 11.36%of the white matter changes detected by TBM.Further, pattern classification between patients and controls revealed that combining different measures improved the classification accuracy(81.9%),indicating that fusion of different structural measures serves as a better neuroimaging marker for the objective diagnosis of schizophrenia.Wasana Ediri Arachchi Yanmin Peng Xi Zhang Wen Qin Chuanjun Zhuo Chunshui Yu Meng Liang 2020Neuroscience Bulletin2020,36,10:6
The benefit of curative liver resection with a selective bile duct preserving approach for hepatocellular carcinoma with macroscopic bile duct tumor thrombus显示文摘Background:Hepatocellular carcinoma(HCC)presenting with macroscopic bile duct tumor thrombus(BDTT)is an uncommon event. The role of a curative hepatic resection and associated long-term outcomes remain controversial. In addition the necessity for bile duct resection is still unclear. The aim of this study was to evaluate outcomes of hepatectomy with a selective bile duct preservation approach for HCC with BDTT in comparison to outcomes without BDTT.Methods:A total of 22 HCC with BDTT patients who had undergone curative hepatic resection with a selective bile duct preservation approach at our institute were retrospectively reviewed. These were compared to group of 145 HCC without BDTT patients. The impact of curative surgical resection and BDTT on clinical outcomes and survival after surgical resection were analyzed. Results:All HCC with BDTT cases underwent major hepatectomy vs.32.4%in the comparative group.Bile duct preservation rate was 56.5%.The 1-,3-and 5-year survival rates of HCC with BDTT patients in comparison to the HCC without BDTT group were 81.8%,52.8%and 52.8%vs.73.6%,55.6%and 40.7%(P=0.804)respectively.Positive resection margin, tumor size≥5 cm and AFP≥200 IU/mL were significant risk factors regarding overall survival. However,it is unclear whether presence of a bile duct tumor thrombus has an adverse impact on either recurrence free survival or overall survival. Conclusions:Bile duct obstruction from tumor thrombus did not necessarily indicate an advanced form of disease.Tumor size and AFP had greater impact on long-term outcomes than bile duct tumor thrombus.Major liver resection with a selective bile duct preserving approach in HCC with BDTT can achieve favorable outcomes comparable to those of HCC without BDTT in selected patients.Anon Chotirosniramit Akkaphod Liwattanakun Sunhawit Junrungsee Wasana Ko-iam Trichak Sandhu Worakitti Lapisatepun 2020Hepatobiliary Surgery and Nutrition2020,9,6:2

Beyond neutrality:adding habitat filtering to neutral models显示文摘Background:Understanding the processes that structure species is one of the primary focuses in community ecology.Hubbell’s neutral model shows stochastic processes alone can describe the two macro-ecological patterns, species richness and species-area relationship,of the community.Although Hubbell’s neutral model can explain the macro-ecological patterns of the species at large scales,it paid less attention to construct the spatial structure of the community.Previous studies suggest that such spatial structures are mostly due to habitat filtering processes work at the intermediate spatial scales. Therefore,Hubbell’s neutral model does not explain the full picture of the community structuring due to its fully stochastic nature. Results:In this study,we proposed a two-schema model that has the habitat filtering component and the stochastic component to construct the species assemblages seen in the community level. The proposed model uses one additional parameter(i.e.number of individuals in habitat)in addition to Hubbell’s three-parameter neutral model(i.e.fundamental bio-diversity number(θ),dispersal limitation(m)and speciation(v)).The proposed model works at two spatial scales:habitat filtering at the intermediate scales and stochastic processes at the large and very small spatial scales. The model coupled the local community dynamics with the meta-community dynamics. The local community has a fixed area with carrying capacity that is proportional to the local community size. The number of habitats in the proposed model can vary.Individuals are placed into habitats with probabilities according to the habitat suitability.Species richness and species composition in each habitat were calculated. The model is fitted for differentθvalues,m values, and a different number of habitats. Conclusions:We assume that habitat filtering plays an important role together with stochastic processes to structure species in forests. Therefore, the proposed model with only four parameters can explain a large proportion of the species structuring of the communities. We found that more species can be maintained in a heterogeneous environment than a uniform environment. Therefore, habitat conservation is highly important for maintaining species diversity in forest communities.Wasana Abeysinghe Ruwan Punchi-Manage 2020Ecological Processes2020,9,1:0
Genetic variation of sand flies(Diptera:Psychodidae)in Gampaha and Kurunegala districts of Sri Lanka:Complementing the morphological identification显示文摘Objective:To identity the variation of sand flies in the Gampaha and Kurunegala districts of Sri Lanka and to assess DNA barcoding as a complementing method for morphological identification. Methods:A total of 38441 sand flies were collected from selected localities in Gampaha and Kurunegala districts using standard entomological techniques from May 2017 to December 2018.Specimens were identified using morphological features and compared with mitochondrial cytochrome C oxidase subunitⅠgene-based DNA barcoding as an alternative tool. Results:Morphological and molecular identification confirmed the presence of four species under two genera(Phlebotomus and Sergentomyia).Phlebotomus argentipes was the predominant species, followed by Sergentomyia(S.)punjabensis,S.babu insularis, and an unidentified Sergentomyia sp.Phlebotomus argentipes showed a clear genetic differentiation from other species.S.babu insularis and S.punjabensis showed a higher genetic affinity to each other than the unidentified species. The unidentified Sergentomyia species is morphologically similar to S.zeylanica, but differs only in clavate gonostyle. Conclusions:DNA barcoding is an effective technique for the identification of sand flies.Further studies using molecular techniques will improve the knowledge of the cryptic diversity of Sri Lankan sand fly fauna.Establishing a reliable and standardized identification system for sand fly species in Sri Lanka is recommended.Tharaka Wijerathna Nayana Gunathilaka Wasana Rodrigo 2022Asian Pacific Journal of Tropical Medicine2022,15,7:0
Genetic diversity of Leishmania donovani isolates from cutaneous lesions of military personnel in the Mullaitivu and Kilinochchi districts of the Northern Province,Sri Lanka显示文摘Objective:To compare the DNA sequences of Leishmania(L.)donovani isolated from individuals in two districts of the Northern Province with other parts of Sri Lanka and neighboring countries. Methods:Samples were collected from military personnel at the Army Hospital,Narahenpita,Sri Lanka from November 2018 to March 2020.A portion of the samples was fixed, stained with Giemsa and observed under the light microscope. The genomic The DNA was extracted from the remaining portion of the samples using DNEasy blood tissue kit(Qiagen,Germany)and amplified using Leishmania genus-specific primers for molecular diagnosis initially.DNA was amplified using L.donovani species-specific primers by PCR and the amplified product was sequenced for comparison of nucleotide sequences. Results:Out of 76 suspected patients,at least one biological sample of 45(59.2%)was positive for L.amastigotes upon microscopy.Overall,33(43.4%)were positive in Leishmania genus-specific PCR,but only 23(30.3%)were positive in L.donovani specific PCR.The dendrogram indicates that the current sequences clustered together with those from Nepal and Gampaha districts(Western Province),Sri Lanka, while the Indian and Eastern African sequences clustered separately. Conclusions:The genetic diversity was low among the isolates, indicating a single and possibly a local point of origin. However, the similarity of Sri Lankan and Nepal strains indicate a possibility of a shared point of origin, which needs more extensive evidence to confirm.Tharaka Wijerathna Nayana Gunathilaka Saveen Semege Nishantha Pathirana Wasana Rodrigo Deepika Fernando 2022Asian Pacific Journal of Tropical Medicine2022,15,9:0
