| 1 | Arenobufagin is a novel isoform-specific probe for sensing human sulfotransferase 2A1显示文摘Human cytosolic sulfotransferase 2 A1(SULT2A1) is an important phase II metabolic enzyme. The detection of SULT2A1 is helpful for the functional characterization of SULT2A1 and diagnosis of its related diseases. However, due to the overlapping substrate specificity among members of the sulfotransferase family, it is difficult to develop a probe substrate for selective detection of SULT2A1. In the present study, through characterization of the sulfation of series of bufadienolides, arenobufagin(AB) was proved as a potential probe substrate for SULT2A1 with high sensitivity and specificity. Subsequently, the sulfation of AB was characterized by experimental and molecular docking studies. The sulfate-conjugated metabolite was identified as AB-3-sulfate.The sulfation of AB displayed a high selectivity for SULT2A1 which was confirmed by in vitro reaction phenotyping assays. The sulfation of AB by human liver cytosols and recombinant SULT2A1 both obeyed Michaelis-Menten kinetics, with similar kinetic parameters. Molecular docking was performed to understand the interaction between AB and SULT2A1, in which the lack of interaction with Met-137 and Tyr-238 of SULT2A1 made it possible to eliminate substrate inhibition of AB sulfation. | 8 | DeepOCL:A deep neural network for Object Constraint Language generation from unrestricted nature language显示文摘Object Constraint Language(OCL)is one kind of lightweight formal specification,which is widely used for software verification and validation in NASA and Object Management Group projects.Although OCL provides a simple expressive syntax,it is hard for the developers to write correctly due to lacking knowledge of the mathematical foundations of the first-order logic,which is approximately half accurate at the first stage of devel-opment.A deep neural network named DeepOCL is proposed,which takes the unre-stricted natural language as inputs and automatically outputs the best-scored OCL candidates without requiring a domain conceptual model that is compulsively required in existing rule-based generation approaches.To demonstrate the validity of our proposed approach,ablation experiments were conducted on a new sentence-aligned dataset named OCLPairs.The experiments show that the proposed DeepOCL can achieve state of the art for OCL statement generation,scored 74.30 on BLEU,and greatly outperformed experienced developers by 35.19%.The proposed approach is the first deep learning approach to generate the OCL expression from the natural language.It can be further developed as a CASE tool for the software industry. | Yilong Yang Yibo Liu Tianshu Bao Weiru Wang Nan Niu Yongfeng Yin | 2024 | CAAI Transactions on Intelligence Technology2024,9,1: | 0 |

| 10 | Preliminary Evaluation of Hemodynamic Effects of Fontan Palliation on Renal Artery Using Computational Fluid Dynamics显示文摘Background:The assessment of renal function is important to the prognosis of patients needing Fontan palliation due to the reconstructed compromised circulation.To know the relationship between the kidney perfusion and hemodynamic characteristics during surgical design could reduce the risk of acute kidney injury(AKI)and the postoperative complications.However,the issue is still unsolved because the current clinical evaluation methods are unable to predict the hemodynamic changes in renal artery(RA).Methods:We reconstructed a three-dimensional(3D)vascular model of a patient requiring Fontan palliation.The technique of computational fluid dynamics(CFD)was utilized to explore the changes of RA hemodynamics under different possible blood flow rates.The relationship between the kidney perfusion and hemodynamic characteristics was investigated.Results:The calculated results indicated the declined tendency of the pressure and pressure drop as the flow rate decreased.When the flow rate decreased to two-thirds of its baseline,both the pressure of left renal artery(LRA)and the pressure of right renal artery(RRA)dipped below 50%,and the pressure of RRA fell more quickly than that of LRA.Uneven distribution of WSS was observed on the trunk of RA,and the lowest WSS was found at the distal of RA.The average WSS in RA dropped to around 50%as the flow rate reached one-third of its baseline.Conclusions:As a promising approach,CFD can be utilized to quantitatively evaluate the hemodynamic characteristics of RA and contribute to offsetting the drawbacks of clinical assessments of renal function,to help realize better prognosis for the patients with Fontan palliation. | Jinlong Liu Jing Shi Weiru Luo Zhirong Tong Lefei Yang Peixuan Sun Tianyi Li Jun Du Qian Wang | 2023 | Congenital Heart Disease2023,18,1: | 0 |
