1cagA and vacA genotype of Helicobacter pylori associated with gastric diseases in Xi'an area显示文摘AIM: To establish stock of clinical Helicobacter pylori (H.pylon) isolates, to perform cagA and vacA typing of these isolates, to evaluate the relationship between genotypes of cagA and vacA and upper gastrointestinal diseases and to assess the association of vacA genotypes with presence of the pathogenicity marker-cagA.METHODS: Clinical H.pylori strains were isolated from the antrum of 259 patients in Clumbia agar. The isolated H.pylori strains were identified by histology, and16SrRNA PCR.CagA genotypes were detected by colony hybridization, the probe was derived from the cloned plasmid PcagA, and digested by EcoRI-HindⅢ and the isolated PcagA DNA fragment was radioactively labelled by the random priming method. vacA genes types (s,m)and subtypes (s1a, s1b,s2) were typed by PCR. Vacuolating toxin was detected with neutral red absorb test. The results were treated statistically by χ2test, ttest, and rank sum test.RESULTS: A total of 192 clinical H. pylori strains were isolated and the stock of Helicobacter pylori was established. The total positive rate of cagA was 87 % in all gastric diseases,and 95 % in gastric cancer group. There was a difference between gastric cancer group and the other groups (P<0.05)except duodenal ulcer group. The expression of type s1 of vacA was more than type s2 (P<0.05), and, the expression of type m1 was equal to type m2. In gastric cancer group,there was a difference between s1a and s1b (P<0.05), and s1a was more than s1b. 2Effects of selenium on peripheral blood mononuclear cell membrane fluidity,interleukin-2 production and interleukin-2 receptor expression in patients with chronic hepatitis显示文摘AIM To study the effect of selenium on peripheral blood mononuclear cell (PBMC) membrane fluidity and immune function in patients with chronic hepatitis.METHODS: PBMCs were pretreated with selenium (1.156x 10.7 mol/L) for 6 h in vitro or extracted directly from patients after administration of selenium-yeast continuously for 8-12 wk (200 pg/d), and then exposed to Con-A for 48 h. The membrane fluidity, interleukin-2 (IL-2) production and interleukin-2 receptor (IL-2R) expression in PBMCs and malondialdehyde (MDA) concentration in medium and lipid peroxide (LPO) in plasma were determined.RESULTS: The PBMC membrane fluidity, IL-2 production and IL-2R expression in patients with chronic hepatitis were significantly lower than those in healthy blood donators (particle adhesive degree R, 0.17±0.01 vs 0.14±0.01,P<0.01; IL-2, 40.26±9.55 vs72.96±11.36, P<0.01, IL-2R,31.05±5.09 vs 60.58±10.56, P<0.01), and the MDA concentration in medium in patients with chronic hepatitis was significantly higher than that in healthy blood donators (1.44±0.08 vs0.93±0.08, P<0.01). 3Analysis of the Global Burden of Disease study highlights the trends in death and disability-adjusted life years of leukemia from 1990 to 2017显示文摘Background:The patterns of leukemia burden have dramatically changed in recent years.This study aimed to estimate the global trends of leukemiarelated death and disability-adjusted life-years(DALYs)from 1990 to 2017.Methods:The data was acquired from the latest version of the Global Burden of Disease(GBD)study.Estimated annual percentage changes(EAPCs)were calculated to estimate the trend of age-standardized rate(ASR)of death and DALYs due to leukemia and its main subtypes from 1990 to 2017.Results:Globally,the numbers of death and DALYs due to leukemia were 347.58×10^(3)(95%uncertainty interval[UI]=317.26×10^(3)-364.88×10^(3))and 11975.35×10^(3)(95%UI=10749.15×10^(3)-12793.58×10^(3))in 2017,with a 31.22% and 0.03% increase in absolute numbers from 1990 to 2017,respectively.Both of their ASR showed decreasing trends from 1990 to 2017 with the EAPCs being−1.04(95%confidence interval[CI]=(−1.10-−0.99)and−1.52(95%CI=−1.59-−1.44),respectively.Globally,the most pronounced decreasing trend of death and DALYs occurred in chronic myeloid leukemia with EAPCs of−2.76(95%CI=−2.88-−2.64)and−2.84(95%CI=−2.97-−2.70),respectively,while the trend increased in acute myeloid leukemia.The death and DALYs of leukemia decreased in most areas and countries with high socio-demographic index(SDI)including Bahrain,Finland,and Australia.Conclusions:The disease burden of death and DALYs due to leukemia decreased globally,and for most regions and countries from 1990 to 2017.However,the leukemia burden is still a substantial challenge globally and required adequate and affordable medical resources to improve the survival and quality of life of leukemia patients.Zejin Ou Danfeng Yu Yuanhao Liang Wenqiao He Yongzhi Li Minyi Zhang Fangfei You Huan He Qing Chen 2020Cancer Communications2020,40,11:4
4High-throughput screening for amyloid-β binding natural small-molecules based on the combinational use of biolayer interferometry and UHPLC−DAD-Q/TOF-MS/MS显示文摘Discovery of drugs rapidly and effectively is an important aspect for Alzheimer's disease(AD).In this study,a novel high-throughput screening(HTS)method aims at screening the small-molecules with amyloid-β(Aβ)binding affinity from natural medicines,based on the combinational use of biolayer interferometry(BLI)and ultra-high-performance liquid chromatography coupled with diode-array detector and quadrupole/time-of-flight tandem mass spectrometry(UHPLC−DAD-Q/TOF-MS/MS)has been firstly developed.Briefly,the components in natural medicines disassociated from biotinylated Aβwere collected to analyze their potential Aβbinding affinity by UHPLC−DAD-Q/TOF-MS/MS.Here,baicalein was confirmed to exhibit the highest binding affinity with Aβin Scutellaria baicalensis.Moreover,polyporenic acid C(PPAC),dehydrotumulosic acid(DTA),and tumulosic acid(TA)in Kai-Xin-San(KXS)were also identified as potent Aβinhibitors.Further bioactivity validations indicated that these compounds could inhibit Aβfibrillation,improve the viability in Aβ-induced PC-12 cells,and decrease the Aβcontent and improve the behavioral ability in Caenorhabditis elegans.The molecular docking results confirmed that PPAC,DTA,and TA possessed good binding properties with Aβ.Collectively,the present study has provided a novel and effective HTS method for the identification of natural inhibitors on Aβfibrillation,which may accelerate the process on anti-AD drugs discovery and development.Minsong Guo Fengdan Zhu Wenqiao Qiu Gan Qiao Betty Yuen-Kwan Law Lu Yu Jianming Wu Yong Tang Chonglin Yu Dalian Qin Xiaogang Zhou Anguo Wu 2022Acta Pharmaceutica Sinica B2022,12,4:3
5siRNA沉默STAT5对食管癌EC9706细胞增殖、凋亡和侵袭的影响(英文)显示文摘Objective:The aim of the study was to explore the effect of STAT5 silenced by siRNA on proliferation,apoptosis and invasion of esophageal carcinoma cell line EC9706.Methods:The siRNA vectors aiming to STAT5 gene were constructed.STAT5 siRNA was transfected into EC9706 cells by Lipofectamine TM 2000.Changes of STAT5,Bcl-2 and Cyclin D1 were analyzed by Western blot and RT-PCR.Effect of STAT5 siRNA on EC9706 cells proliferation was determined by MTT.Effects of STAT5 siRNA on EC9706 cells cycle and apoptosis were detected by the flow cytometry.Boyden chamber was used to evaluate the invasion and metastasis capabilities of EC9706 cells.Results:The double strands oligonucleotide of siRNA aiming to STAT5 was successfully cloned into the pRNAT-U6.1 vector,and the target sequence was coincide with the design.RT-PCR and Western blotting detection demonstrated that the expression levels of STAT5,Bcl-2 and Cyclin D1 genes were obviously decreased in EC9706 cells transfected with STAT5 targeting siRNA expression vectors.STAT5 siRNA could suppress the proliferation of EC9706 cells.The proportions of S and G2/M periods frequency were significantly decreased (P < 0.05),and the proportion of G0/G1 period frequency was significantly increased (P < 0.05).The average amount of cells penetrating Matrigel was significantly decreased (P < 0.05).Silencing the STAT5 induced the apoptosis of esophageal carcinoma cell line EC9706 (P < 0.01).Conclusion:STAT5 silenced by siRNA of esophageal carcinoma cell line EC9706 could induce the apoptosis.And it could suppress the proliferation,invasion and metastasis of tumor cells.Daoxin Jin Wenqiao Zang Tao Wang Min Li Jieru Wan Guoqiang Zhao 2010The Chinese-German Journal of Clinical Oncology2010,9,12:2
6Effective syngas cleanup and reforming using Ni/γ-Al_(2)O_(3) catalyst显示文摘This work reports the performance of a selective Ni-based catalyst in tar removal and syngas reforming.Benzene was used as the model tar to optimize catalytic reaction conditions.Parameters investigated were reaction temperature(700℃ to 900℃),gas residence time(0.1 s to 1.1 s),and catalyst loadings(3%to 21%of the weight of γ-Al_(2)O_(3) support).On the basis of the benzene test,a reaction temperature of 800℃,catalyst loading of 15wt%,and residence time of 0.3 s were chosen as optimum reaction conditions.Testing of these conditions showed that the Ni/γ-Al_(2)O_(3) catalyst removed more than 99%of tars in syngas in the downdraft gasifier and 98%in the updraft gasifier.Concentrations of combustible compounds of syngas also increased significantly.H_(2) concentration increased from 19.96%to 51.78%in the downdraft gasifier and from 23.97%to 37.39%in the updraft gasifier;CO concentration increased from 16.26%to 21.10%in the downdraft gasifier and from 22.95%to 25.64%in the updraft gasifier.The results indicate that the Ni/γ-Al_(2)O_(3) catalyst and associated catalytic conditions could not only effectively eliminate tars but also improve the quality of syngas in biomass gasification.Duo Wang Wenqiao Yuan Wei Ji 2010International Journal of Agricultural and Biological Engineering2010,3,2:2
7A Preliminary Study on the Datong Fault Belt显示文摘The Datong fault belt is a NE trending fault in the northern Qinghai-Xizang (Tibet) Plateau and controls the boundary of the Xining Basin and Datong Basin. It consists of the Maziying-Miaogou (F_1) fault and the Laoye Mountain-Nanmenxia fault (F_2). There is obvious displacement in vertical direction along the belt. The field investigation results show that this belt has long-term activity. There are several meters long crushed zones and veins along the fault side in the basement rock. On the fault section,the Cambria system thrusts over the red-brick-colored Quaternary Period gravel,and there is a fault gouge of several centimeters thick developed on the fault plane. The fault gouge date (ESR) on the fault plane is 610±61ka. The covering deluvial loess is not dislocated,and the OSL result is 14.6±1.5ka. So it can be concluded that the fault belt was active in the middle Pleistocene,but inactive in the late Pleistocene according to the age data and geomorphologic features. Interior formations of the Datong basin features fold with the major axis orienting northwest. According to the relation of fault and fold deformation,Datong fault is a transversal tear,which is due to uneven compression of the folds in different parts and NNE trending regional compressive stress. It is common among the NE trending faults in the northeast of Qinghai-Xizang (Tibet) Plateau. These NE trending faults aren't large,and most are located in the active plate. 9The Signatures of Stable Isotopes δ^(15)N and δ^(13)C in Anadromous and Non-Anadromous Coilia nasus Living in the Yangtze River, and the Adjacent Sea Waters显示文摘Stable isotopes are increasingly used to investigate seasonal migrations of aquatic organisms. This study employed stable isotopes(δ^(13) and δ15N) for Coilia nasus from the lower Yangtze River and the adjacent East China Sea to distinguish different ecotypic groups, ascertain trophic nutrition positions, and reflect environmental influences on C. nasus. δ^(13)C signatures of C. nasus sampled from Zhoushan(ZS), Chongming(CM), and Jingjiang(JJ) waters were significantly higher than those from the Poyang Lake(PYL)(P < 0.05). By contrast, δ^(15)Nsignatures of C. nasus in ZS, CM, and JJ groups were significantly lower than those in PYL group(P < 0.05). Basing on δ^(13) and δ^(15)Nsignatures, we could distinguish anadromous(ZS, CM, and JJ) and non-anadromous(PYL) groups. The trophic level(TL) of anadromous C. nasus ranged from 2.90 to 3.04, whereas that of non-anadromous C. nasus was 4.38. C. nasus occupied the middle and top nutrition positions in the marine and Poyang Lake food webs, respectively. C. nasus in Poyang Lake were significantly more enriched in δ^(15)N but depleted in δ^(13), suggesting that anthropogenic nutrient inputs and terrigenous organic carbon are important to the Poyang Lake food web. This study is the first to apply δ^(15)Nand δ^(13) to population assignment studies of C. nasus in the Yangtze River and its affiliated waters. 11Interpretation of the Spatial Distribution Characteristics of the Co-seismic Landslides Induced by the 1920 Haiyuan M81/2 Earthquake Using Remote Sensing Images显示文摘Analyzing the spatial distribution characteristics of earthquake-induced secondary disasters based on advanced techniques is significantly important,especially in understanding the process of strong earthquakes in the Loess Pateau.Using ArcGIS,this study interprets multi-temporal high-resolution satellite images,field investigation data,and historical seismic records.Major conclusions are obtained as follows:①Landslides induced by the Haiyuan earthquake are mainly distributed in the intersection area of the end of the Haiyuan fault and Liupanshan fault,as indicated by multiple dense distribution centers;②The landslide distribution of the Haiyuan Earthquake is determined by the distance to the fault,topographic relief,slope,lithology,and other factors.In detail,the closer the distance to the fault,the greater the density of the landslide.The greater the slope and relief of the terrain,the greater the density and the smaller the average area of a landslide.Compared with tertiary strata,Quaternary strata has a larger average area,and the density of the landslides is smaller;③The density curve of the death toll in the Haiyuan earthquake can be used as a reference for the distribution of co-seismic landslides.Several Haiyuan co-seismic landslides are distributed in the Tongwei landslide area;however,the major landslides here are induced by the 1718 Tongwei earthquake rather than the 1920 Haiyuan earthquake;④The co-seismic landslides of the Haiyuan earthquake exhibits the“slope effect”in the south-west plate of Haiyuan fault,presenting the dominant sliding direction towards the fault and epicenter;however,the“slope effect”is not evident in the northeast plate of the fault.ZHANG Weiheng XU Yueren DU Peng LI Wenqiao TIAN Qinjian CHEN Lize 2020Earthquake Research in China2020,34,4:1
12Relationship between the Landslides Triggered by the Tongwei M71/2 Earthquake in 1718 AD and the Disappearance of Yongning Ancient Town显示文摘Although China’s historical earthquake documentation is relatively rich,it is not all based on scientific records.Therefore,the verification of the seismic information in historical records can effectively avoid exaggerating or underestimating the damage they produced.In this paper,we analyze the detailed information of the 1718 AD Tongwei M7⅟��earthquake through field surveys,document sorting,and manual visual interpretation of UAV images.Major conclusions are listed as follows:①The low-level terraces of Weihe River between Gangu and Wushan are fully developed with flatted surface,and the residents here are mostly killed by house collapses.In addition,the disappearance of Yongning Ancient Town is not directly related to the earthquakeinduced landslides;②In fact,“Yongning Town is entirely buried by the earthquake”in the historical records describes the phenomenon that loess dust has pervaded the entire Weihe Valley.These dust grains are produced by the sliding of earthquakeinduced landslides.Thus,there is no possibility that large-scale landslides have slipped over Weihe bedrock and buried Yongning Town;③After the earthquake,survivors abandoned the ruins and selected a new site to live.They built a new town named“Pan’an(means always peaceful in the future)”.earthquake-induced landslides may be the reason of burying the residential areas on valley-side slopes,while those locations inside the valley are associated with the amplification effect of ground vibration.On the basis of compiling historical seismic data,scientific methods are used to explore the real meaning of these documents,for the purpose of providing basic data for the risk evaluation of strong historical earthquakes.XU Yueren DU Peng LI Wenqiao TIAN Qinjian CHEN Lize 2020Earthquake Research in China2020,34,4:1
13The Calculation Method for the Volume of Landslides and Dammed Lake Sediments Triggered by a Strong Historical Earthquake in the Loess Plateau:A Case Study of Qiuzigou, Gansu Province, Northwest China显示文摘The quantitative calculation of the volume of large earthquake-triggered landslides and related dammed lake sediments is of great significance in the study of secondary disasters and focal parameters of strong historical earthquakes.In this study,the dammed lake induced by Qishan M7 earthquake(Lingtai County,Gansu Province,Northwest China)is selected as the research object.Based on the information collected from the 4 boreholes in the dammed lake area,we further take advantage of the lowlevel Unmanned Aerial Vehicle(UAV)photogrammetry and the morphology recovery method,to calculate the volume of the dammed lake and landslides,respectively.Finally,major conclusions are obtained as follows:①the AMS-14C age at the bottom of the Qiuzigou Dammed Lake sediments is 2890±30 BP,which coincides with the 780 BC Qishan earthquake;furthermore,the Qiuzigou Landslides seem to have been triggered by the earthquake,forming an enclosed dammed lake deposition environment after the upstream sediments accumulate;②the Qiuzigou landslides are opposite-sliding landslides that have blocked the river valley;in detail,landslide volumes at the right and left banks are 235×104 m3 and 229×104 m3,respectively.The length of the dammed lake is 2.6 km,with a thickness of approximately 43 m near the landslides,and the total sedimentary volume is 573×104 m3;③the erosion rate of Qiuzigou Landslide Dammed Lake is 0.44 mm/a,the accumulation rate is 15.05 mm/a,and the soil erosion modulus is 593 t/(km2/a),characterized as slight erosion.Quantitative research on the formation of landslides and dammed lakes from strong historical earthquakes is vital for increasing our understanding of the vibrational characteristics and surface action processes of these types of earthquakes.DU Peng XU Yueren LI Wenqiao TIAN Qinjian CHEN Lize 2020Earthquake Research in China2020,34,4:1
14The Volume Calculation Method of Rock Collapses and Loess Landslides Triggered by the 1556 AD Huaxian M81/2 Earthquake in Shaanxi Province, China显示文摘Accurate volume calculation of each individual landslide triggered by strong historical earthquakes can help understand the characteristics of the typical earthquake-induced landslides,thus providing significant information for the modification of the focal parameters of historical earthquakes.In this study,we select one rock fall and three loess landslides triggered by the 1556 AD Huaxian M8⅟��earthquake,compute their volumes using the low-altitude high-precision Unmanned Aerial Vehicle(UAV)photogrammetry and landslide profile restoration methods.The results show that:①the whole influencing area of the Huangjiagou Rock Fall is approximately 3.03×105 m2 and the area of the collapsed rock accumulated at the slope foot is 3.33×104 m2,accounting for approximately 10%of the entire influencing range.However,the estimated volume of the collapsed rock is only 0.699×106 m3,indicating a rock fall with large influencing range but limited collapsed rock;②the geological form of thethree loess landslides are preserved intactly,with volumes of 0.283×108 m3,0.074×108 m3,and 0.377×108 m3.These important geological hazard relics reflect the strong vibrations and severe casualties in the meizoseismal area;③loess landslides are the key reason of the serious death toll in the hilly-gully loess area.Our new method can be used to estimate the influencing area and the actual volume of each individual landslide,and rationally evaluate the role of earthquake landslides in the disaster.In addition,quantitative research on secondary disasters triggered by strong historical earthquakes is beneficial for understanding the surface process and focal parameters of the earthquakes.ZHANG Yanbo XU Yueren DU Peng LI Wenqiao CHEN Lize TIAN Qinjian 2020Earthquake Research in China2020,34,4:1

16Terahertz bound states in the continuum with incident angle robustness induced by a dual period metagrating显示文摘Metasurface-empowered bound state in the continuum(BIC)provides a unique route for fascinating functional devices with infinitely high quality factors.This method is particularly attractive to the terahertz community because it may essentially solve the deficiencies in terahertz filters,sensors,lasers,and nonlinear sources.However,most BIC metasurfaces are limited to specified incident angles that seriously dim their application prospects.Here,we propose that a dual-period dielectric metagrating can support multiple families of BICs that originate from guided mode resonances in the dielectric grating and exhibit infinite quality factors at arbitrarily tilted incidence.This robustness was analyzed based on the Bloch theory and verified at tilted incident angles.We also demonstrate that inducing geometric asymmetry is an efficient way to manipulate the leakage and coupling of these BICs,which can mimic the electromagnetically induced transparency(EIT)effect in our dual-period metagrating.In this demonstration,a slow-light effect with a measured group delay of 117 ps was achieved.The incidence-insensitive BICs proposed here may greatly extend the application scenarios of the BIC effect.The high Q factor and outstanding slow-light effect in the metagrating show exciting prospects in realizing high-performance filters,sensors,and modulators for prompting terahertz applications.Wenqiao Shi Jianqiang Gu Xingyuan Zhang Quan Xu Jiaguang Han Quanlong Yang Longqing Cong Weili Zhang 2022Photonics Research2022,10,3:1
17Computational investigation of the effect of biodiesel fuel properties on diesel engine NOx emissions显示文摘A detailed numerical spray atomization,ignition,combustion and nitrogen oxides(NOx)formation model was developed for direct injection diesel engines by using KIVA3V code.Several modified or recalibrated sub-models including a Kelvin-Helmholtz Rayleigh-Taylor(KH-RT)spray breakup model,a Shell ignition model,a single-step kinetic combustion model and a Zel’dovich NOx formation model were incorporated into KIVA3V.This modified model was validated by experimental data obtained from a John Deere 4045T direct injection diesel engine that was fueled with a natural soybean methyl ester,a yellow grease methyl ester,a genetically modified soybean methyl ester and No.2 diesel fuel.Errors between predictions of the brake-specific NOx and measured values were less than 1%at full load.For biodiesel fuels,either the Zel’dovich mechanism overpredicted NOx emissions,the ratio of NO to NOx should be less than diesel fuel,or both.As observed from the modeling results,the higher latent heat of vaporization and higher surface tension of biodiesel relative to diesel fuel did not result in increased NOx emissions.The higher viscosity of biodiesel could be one of the reasons for increased NOx,but its effect was relatively small compared with the effect of decreased spray cone angle and advanced start of injection timing on NOx.Decreased spray cone angle and advanced start of injection were the main reasons for increased NOx emissions of biodiesel.Wenqiao Yuan A.C.Hansen 2009International Journal of Agricultural and Biological Engineering2009,2,2:1
