| 2 | Differential protein expression in substantia nigra induced by 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine in a mouse model of chronic Parkinson’s disease显示文摘BACKGROUND: To date, a complete protein expression profile of the midbrain substantia nigra in a mouse model of chronic Parkinson’s disease, induced by 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP), does not exist. In addition, there are no reports of analysis of differential protein expression. OBJECTIVE: To separate and evaluate MPTP-induced differential protein expression through the use of proteomics in the substantia nigra of a mouse model of chronic Parkinson’s disease. DESIGN: Randomized controlled animal study. SETTING: Department of Neurology, the First Affiliated Hospital, Chongqing Medical University. MATERIALS: Sixteen 8–10-week old, healthy, male, C57BL mice, weighing 20–25 g, and of clean grade, were provided by the Experimental Animal Center of Chongqing Medical University. The experimental animals were disposed according to ethical criteria. MPTP was provided by Sigma Company, USA; Pdquest 2D image analysis software and gelatum/irradiance image analysis system (ChemiDoc XRS) by Bio-Rad, USA; and Voyager DE-PROMALDI-TOF-MS mass spectroscopy analyzer by ABI Company, USA. METHODS: This study was performed in Chongqing Neurological Laboratory between November 2006 and July 2007. Mice were randomly divided into model and control groups, with 8 mice in each group. Mice in the model group were received a subcutaneous injection of MPTP (25 mg/kg), twice a week, for five successive weeks, to establish a chronic Parkinson’s disease model. Mice in the control group received the same volume of a subcutaneous saline injection at the same time points. Mice were sacrificed by anesthesia to rapidly obtain the midbrain for protein separation of the substantia nigra. MAIN OUTCOME MEASURES: (1) 2-ED handbook (Bio-Rad Company) was referenced for two-dimensional electrophoresis. (2) PDQUEST8.0 analytical electrophoresis pattern was adopted to evaluate differential protein expression. (3) Peptide mass finger print map and data were retrieved on http://www.prospector.ucsf.edu to compare differential substantia nigral protein expression in the two groups. RESULTS: Two-dimensional gel electrophoresis of substantia nigra tissue indicated that there were 33 differential protein expressions between the two groups. Three new proteins were evaluated, including α-enolase, which exhibited regulated expression, tumor necrosis factor ligand superfamily member 4, and cyclin-dependent kinase inhibitor 1B. | 3 | Influence of mandrel-cores filling on size effect of cross-section distortion of bimetallic thin-walled composite bending tube显示文摘Two new size factors of cross-section hollow coefficient and bending degree are introduced to reveal the size effect of bending forming of bimetallic composite tube.Hollow coefficient and bending degree can limit the commonly used bent tube to the size description range of(0,2.00).The evolution laws of the cross-section distortion forms in the hollow coefficient-bending degree interval are revealed as well as the action of the mandrel-cores on the size effect.Results show the mandrel-cores filling can expand the forming limit of the bent tube,but also bring two other forming defects of wrinkle and rupture.The identification factor(hollow coefficient multiply bending degree)provides a method for querying the cross-section distortion forms of all composite bending tubes.In the identification factor interval(0,1.00),the distribution area of bending forming defects of the composite tube is continuous.The thin-walled composite bending tube collapses when identification factor in(0,0.39),wrinkles when identification factor in[0.39,0.50),and ruptures when identification factor in[0.50,1.00).The mathematical model of size effect is derived,by which the average cross-section distortion rate is found to distribute like a radial leaf in the hollow coefficient-bending degree qualified forming space.The best forming zone is hollow coefficient 0.46-0.68,and bending degree 0.25-0.47. | Yingxia ZHU Miaomiao WAN Wei CHEN Yun WANG Wenbin TU Fan XU | 2023 | Chinese Journal of Aeronautics2023,36,3: | 0 |
| 4 | Cerebrospinal fluid diagnostic markers for two-dimensional electrophoresis-mass spectrometry in Parkinson’s disease patients显示文摘BACKGROUND: Previous studies have confirmed the existence of specific proteins in body fluid of Parkinson’s disease (PD) patients. However, the existing research has contained several interference factors with poor reproducibility and has not focused on patients grouped according to disease duration. OBJECTIVE: To verify differential expression of proteins in cerebrospinal fluid of PD patients grouped in order of disease severity through the use of two-dimensional electrophoresis-mass spectrometry methods. DESIGN, TIME AND SETTING: The proteomic-based, case-control study was performed between September 2008 and June 2009 at the Key Laboratory of Neurology in the First Affiliated Hospital of Chongqing Medical University. PARTICIPANTS: A total of 52 outpatients and/or inpatients, who were admitted to the Department of Neurology in the First Affiliated Hospital of Chongqing Medical University between 2008 and 2009, were randomized into the present study. Among them, 27 PD patients served as the PD group and were assigned to three subgroups according to modified Webster, Hoehn, and Yahr rating scales: 14 = mild, 8 = moderate, and 5 = severe; non-PD group of 16 patients included 5 cases of viral meningitis, 3 cases of acute myelitis, 1 case of Guillain-Barre syndrome, 2 cases of tuberculous meningitis, 2 cases of restless legs syndrome, and 3 cases of essential tremor; control group (n = 9) consisted of muscular tension headache in 6 cases, as well as syncope, trigeminal neuralgia, idiopathic orthostatic hypotension in 1 case. METHODS: Cerebrospinal fluid was collected from the involved patients using the lumbar puncture method. Proteins in the cerebrospinal fluid were separated by two-dimensional electrophoresis. MAIN OUTCOME MEASURES: Characteristics of protein electrophoresis patterns were analyzed, differentially expressed proteins were detected using matrix-assisted laser desorption ionization time of flight mass spectrometry, and protein data were analyzed in the Mascot database. RESULTS: Five protein electropherograms were analyzed by PDQuest 8.0, and (789 ± 32) protein spots were observed. There were significant differences in four protein spots in each of the PD sub-groups compared with the non-disease and control groups. Expression was down-regulated in three protein spots and up-regulated in one protein spot; 100% repetition rate was observed in four protein spots. According to the Mascot database, protein spots with down-regulated expression were as follows: DNA-guided RNA polymerase III subunit RPC5 (score: 50 points); double serine, threonine, and tyrosine protein kinase (score: 64 points, P < 0.05); activity-regulated cytoskeleton-associated protein (score: 58 points, P < 0.05). However, G2 mitotic-specific cyclin was up-regulated (score: 84 points, P < 0.05). | 6 | USEVis:Visual analytics of attention-based neural embedding in information retrieval显示文摘Neural attention-based encoders,which effectively attend sentence tokens to their associated context without being restricted by long-term distance or dependency,have demonstrated outstanding performance in embedding sentences into meaningful representations(embeddings).The Universal Sentence Encoder(USE)is one of the most well-recognized deep neural network(DNN)based solutions,which is facilitated with an attention-driven transformer architecture and has been pre-trained on a large number of sentences from the Internet.Besides the fact that USE has been widely used in many downstream applications,including information retrieval(IR),interpreting its complicated internal working mechanism remains challenging.In this work,we present a visual analytics solution towards addressing this challenge.Specifically,focused on semantics and syntactics(concepts and relations)that are critical to domain clinical IR,we designed and developed a visual analytics system,i.e.,USEVis.The system investigates the power of USE in effectively extracting sentences’semantics and syntactics through exploring and interpreting how linguistic properties are captured by attentions.Furthermore,by thoroughly examining and comparing the inherent patterns of these attentions,we are able to exploit attentions to retrieve sentences/documents that have similar semantics or are closely related to a given clinical problem in IR.By collaborating with domain experts,we demonstrate use cases with inspiring findings to validate the contribution of our work and the effectiveness of our system. | Xiaonan Ji Yamei Tu Wenbin He Junpeng Wang Han-Wei Shen Po-Yin Yen | 2021 | Visual Informatics2021,5,2: | 0 |
