| 1 | AMF siRNA treatment of keloid through inhibition signaling pathway of RhoA/ROCK1显示文摘A keloid(KD)is a benign dermal fibrotic tumor.Treatment of KDs is challenging and the recurrence rate is high;thus,there is an unmet need to explore new target sites and new treatment methods.As a tumor-associated cytokine,autocrine motility factor(AMF)can effectively stimulate the random and directional movement of cells.We first found that AMF was overexpressed in keloid fibroblasts(KFs)and the proliferation and migration of KFs were promoted by AMF stimulation.After treatment with Y-27632,RhoA kinase inhibitor,the proliferation and migration capacity of KFs declined significantly,and type I collagen protein,active RhoA and ROCK1 also were downregulated.In addition,a KD transplantation model was established under the skin of nude mice,with KD intramural injection AMF siRNA,we found that the weight of the KD was smaller than in the control group(P<0.05),KD tissue sections stained by HE and Masson showed that fibers became loose and the blood vessels were visibly reduced.In conclusion,AMF siRNA is expected to be a novel strategy to treat KD by inhibiting signaling pathway of RhoA/ROCK1. | Yi Tian Lan Jin Wenhong Zhang Zumeng Ya Yuan Cheng Hongyun Zhao | 2019 | Genes & Diseases2019,6,2: | 4 |
| 2 | Non-suicidal self-injury in Shanghai inner bound middle school students显示文摘Background Non-suicidal self-injury (NSSI) behaviour is very common among adolescents. Its prevalence and behavioural characteristics may vary according to regional and cultural differences. Investigation of NSSI locations and diagnosis of adolescents with NSSI are relatively lacking in China. Aims The study objective was to determine the prevalence and features of NSSI among middle school students in Shanghai. Methods The participants were from grade 6 to 8 selected from three junior schools in Jing’an District. Consenting students completed the Ottawa Self-Injury Inventory to determine the prevalence and characteristics of NSSI. Those who indicated NSSI within the past month were administered the Mini-International Neuropsychiatric Interview for Children and Adolescents to assess for emotional disorders. Result The result shows 510 participants (21.7%;mean age 13.51 (0.97) years old;56.7% female) reported at least one instance of NSSI during the previous 12 months. NSSI was significantly more common in girls than boys (24.9% vs 18.5%;χ^2=14.03, p=0.00). Commonly reported reasons for NSSI were for internal and external emotion regulation (87.9%, 82.3%) and social influence (57.8%). Anxiety disorders were the most common (28.9%) disorder associated with NSSI. Conclusion The rate of NSSI of middle school students in Shanghai inner bound is similar to those reported in North American and European youth. | 3 | Development of a psychological first-aid model in inpatients with COVID-19 in Wuhan,China显示文摘Background Facing the social panic and substantial shortage of medical resources during the coronavirus disease 2019(COVID-19)outbreak,providing psychological first-aid to inpatients is essential for their rehabilitation and the orderly operating of medical systems.However,the closed-ward environment and extreme shortage of onsite mental health workers have limited the use of traditional face-to-face diagnosis and psychological interventions.Aim To develop a mental health intervention model for inpatients that can be applied during a widespread epidemic,such as COVID-19.Methods In a medical team stationed in Leishenshan Hospital,Wuhan,China,we integrated onsite and online psychological support resources to implement a graded psychological intervention system.The onsite psychiatrist established trust with the patients and classified them into categories according to their symptom severity.While face-to-face evaluation and intervention are critical for effective online support,the online team effectively extended the scope of the'first-aid'to all patients.Conclusion This integrated onsite and online approach was effective and eficient in providing psychological interventions for inpatients during the crisis.Our model provides a realistic scheme for healthcare systems in or after the COVID-19 epidemic and also could be adopted in areas of the world with insufficient mental healthcare resources. | Wenhong Cheng Fang Zhang Yingqi Hua Zhi Yang Jun Liu | 2020 | General Psychiatry2020,33,3: | 3 |

| 7 | SARS-CoV-2 helicase NSP13 hijacks the host protein EWSR1 to promote viral replication by enhancing RNA unwinding activity显示文摘Background:Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)emerged in December 2019 and has led to a global coronavirus disease 2019(COVID-19)pandemic.Currently,incomplete understanding of how SARS-CoV-2 arrogates the host cell to establish its life cycle has led to slow progress in the development of effective drugs.Results:In this study,we found that SARS-CoV-2 hijacks the host protein EWSR1(Ewing Sarcoma breakpoint region 1/EWS RNA binding protein 1)to promote the activity of its helicase NSP13 to facilitate viral propagation.NSP13 is highly conserved among coronaviruses and is crucial for virus replication,providing chemical energy to unwind viral RNA replication intermediates.Treatment with different SARS-CoV-2 NSP13 inhibitors in multi-ple cell lines infected with SARS-CoV-2 effectively suppressed SARS-CoV-2 infection.Using affinity-purification mass spectrometry,the RNA binding protein EWSR1 was then identified as a potent host factor that physically associated with NSP13.Furthermore,silencing EWSR1 dramatically reduced virus replication at both viral RNA and protein levels.Mechanistically,EWSR1 was found to bind to the NTPase domain of NSP13 and potentially enhance its dsRNA unwinding ability.Conclusions:Our results pinpoint EWSR1 as a novel host factor for NSP13 that could potentially be used for drug repurposing as a therapeutic target for COVID-19. | Hongxiang Zeng Xiang Gao Gang Xu Shengyuan Zhang Lin Cheng Tongyang Xiao Wenhong Zu Zheng Zhang | 2022 | Infectious Medicine2022,1,1: | 0 |
