22021:A Year of Unprecedented Climate Extremes in Eastern Asia,North America,and Europe显示文摘The year 2021 was recorded as the 6th warmest since 1880.In addition to large-scale warming,2021 will be remembered for its unprecedented climate extremes.Here,a review of selected high-impact climate extremes in 2021,with a focus on China,along with an extension to extreme events in North America and Europe is presented.Nine extreme events that occurred in 2021 in China are highlighted,including a rapid transition from cold to warm extremes and sandstorms in spring,consecutive drought in South China and severe thunderstorms in eastern China in the first half of the year,extremely heavy rainfall over Henan Province and Hubei Province during summer,as well as heatwaves,persistent heavy rainfall,and a cold surge during fall.Potential links of extremes in China to four global-scale climate extremes and the underlying physical mechanisms are discussed here,providing insights to understand climate extremes from a global perspective.This serves as a reference for climate event attribution,process understanding,and high-resolution modeling of extreme events.Tianjun ZHOU Wenxia ZHANG Lixia ZHANG Robin CLARK Cheng QIAN Qinghong ZHANG Hui QIU Jie JIANG Xing ZHANG 2022Advances in Atmospheric Sciences2022,39,10:5
3EGFR TKIs impair lysosome-dependent degradation of SQSTM1 to compromise the effectiveness in lung cancer显示文摘Tyrosine kinase inhibitors for epidermal growth factor receptor(EGFR TKIs)greatly improved clinical outcomes of patients with non-small cell lung cancer(NSCLC).Unfortunately,primary and acquired resistance limits their clinical benefits.To overcome such resistance,new generations of EGFR TKIs have been developed by targeting newly identified mutations in EGFR.However,much less effort has been put into alternative strategies,such as targeting the intrinsic protective responses to EGFR TKIs.In this study,we found that EGFR TKIs,including gefitinib and AZD9291,impaired lysosome-dependent degradation of SQSTM1,thus compromising their anti-cancer efficiency.By accumulating in the lysosome lumen,gefitinib and AZD9291 attenuated lysosomal acidification and impaired autolysosomal degradation of SQSTM1 owing to their intrinsic alkalinity.As a result,SQSTM1 protein was stabilized in response to gefitinib and AZD9291 treatment and conferred EGFR TKI resistance.Depleting SQSTM1 significantly increased the sensitivity of NSCLC cells to gefitinib and AZD9291 both in vitro and in vivo.Furthermore,a chemically modified gefitinib analog lacking alkalinity displayed stronger inhibitory effects on NSCLC cells.Therefore,targeting accumulated SQSTM1 or chemically modified EGFR TKIs may represent new strategies to increase the effectiveness of EGFR targeted therapy.Lixian Yang Shilong Ying Shiman Hu Xiangtong Zhao Muchun Li Miaoqin Chen Yiran Zhu Ping Song Liyuan Zhu Tingting Jiang Huimin An Neelum Aziz Yousafzai Wenxia Xu Zhiguo Zhang Xian Wang Lifeng Feng Hongchuan Jin 2019Signal Transduction and Targeted Therapy2019,4,1:2

7B-cell-derived IL-10 promotes allergic sensitization in asthma regulated by Bcl-3显示文摘Aeroallergen sensitization,mainly mediated by lung epithelium and dendritic cells(DCs),is integral to allergic asthma pathogenesis and progression.IL-10 has a dual role in immune responses,as it inhibits myeloid cell activation but promotes B-cell responses and epithelial cell proliferation.Here,we report a proinflammatory function of B-cell-derived IL-10 modulated by Bcl-3 in allergic asthma.Specifically,Bcl-3^(−/−)mice showed elevated IL-10 levels and were found to be highly vulnerable to allergic asthma induced by house dust mites(HDMs).IL-10 had a positive correlation with the levels of the DC chemoattractant CCL-20 in HDM-sensitized mice and in patients with asthma and induced a selective increase in CCL-20 production by mouse lung epithelial cells.Blockade of IL-10 or IL-10 receptors during sensitization dampened both HDM-induced sensitization and asthma development.IL-10 levels peaked 4 h post sensitization with HDM and IL-10 was primarily produced by B cells under Bcl-3–Blimp-1–Bcl-6 regulation.Mice lacking B-cell-derived IL-10 displayed decreased lung epithelial CCL-20 production and diminished DC recruitment to the lungs upon HDM sensitization,thereby demonstrating resistance to HDM-induced asthma.Moreover,responses to HDM stimulation in Bcl-3^(−/−)mice lacking B-cell-derived IL-10 were comparable to those in Bcl-3^(+/+)mice.The results revealed an unexpected role of B-cell-derived IL-10 in promoting allergic sensitization and demonstrated that Bcl-3 prevents HDM-induced asthma by inhibiting B-cell-derived IL-10 production.Thus,targeting the Bcl-3/IL-10 axis to inhibit allergic sensitization is a promising approach for treating allergic asthma.Guojun Qian Wenxia Jiang Donglin Sun Zhun Sun Anning Chen Hongwei Fang Jingyao Wang Yongzhong Liu Zhinan Yin Haiming Wei Hao Fang Xiaoren Zhan 2023Cellular & Molecular Immunology2023,20,11:1

9Explosive characteristics of nanometer and micrometer aluminum-powder显示文摘The explosive characteristics of aluminum powder have great significance in preventing and controlling aluminum-dust explosion accidents, especially the nano-aluminum powder. The explosion characteris- tics of 100 nm and 75 lm aluminum powders were investigated by using a 20 L spherical explosion cav- ity and a horizontal pipe whose cross-section area is 80 mm 80 mm and length is 8 m. The results show that the maximum explosion pressure and its rising rate of 100 nm aluminum powder gradually increase with increasing concentration of aluminum-powder at the beginning. When aluminum-powder concen- tration is 1 kg/m3, the maximum explosion pressure reaches its maximum, and then gradually decreases. While when the concentration is 1.25 kg/m3, the maximum rate of pressure rise obtains its maximum, and then decreases. After 100 nm aluminum powder is exploded in pipes, the peak overpressure of blast wave first decreases and then increases to the maximum at a distance of 298 cm from the ignition source, and then gradually decreases. The most violent concentration is about 0.4 kg/m3 which is lower than 0.8 kg/m3 of 75 lm aluminum powder, so 100 nm aluminum powders are more easily exploded. The change laws of maximum explosion pressure, maximum rate of pressure rise and blast-wave peak over- pressure of 100 nm aluminum powders with concentration are similar to those of 75 lm aluminum pow- ders, but these values are much higher than 75 lm aluminum powders under the same concentration, so the aluminum-powders explosion of 100 nm will produce more harms. 11FBXW7βloss-of-function enhances FASN-mediated lipogenesis and promotes colorectal cancer growth显示文摘Continuous de novo fatty acid synthesis is required for the biosynthetic demands of tumor.FBXW7 is a highly mutated gene in CRC,but its biological functions in cancer are not fully characterized.Here,we report that FBXW7β,a FBXW7 isoform located in the cytoplasm and frequently mutated in CRC,is an E3 ligase of fatty acid synthase(FASN).Cancer-specific FBXW7βmutations that could not degrade FASN can lead to sustained lipogenesis in CRC.COP9 signalosome subunit 6(CSN6),an oncogenic marker of CRC,increases lipogenesis via interacting with and stabilizing FASN.Mechanistic studies show that CSN6 associates with both FBXW7βand FASN,and antagonizes FBXW7β’s activity by enhancing FBXW7βautoubiquitination and degradation,which in turn prevents FBXW7β-mediated FASN ubiquitination and degradation,thereby regulating lipogenesis positively.Both CSN6 and FASN are positively correlated in CRC,and CSN6-FASN axis,regulated by EGF,is responsible for poor prognosis of CRC.The EGF-CSN6-FASN axis promotes tumor growth and implies a treatment strategy of combination of orlistat and cetuximab.Patient-derived xenograft experiments prove the effectiveness of employing orlistat and cetuximab combination in suppressing tumor growth for CSN6/FASN-high CRC.Thus,CSN6-FASN axis reprograms lipogenesis to promote tumor growth and is a target for cancer intervening strategy in CRC.Wenxia Wei Baifu Qin Weijie Wen Boyu Zhang Haidan Luo Yuzhi Wang Hui Xu Xiaoshan Xie Sicheng Liu Xin Jiang Mengan Wang Qin Tang Jiayu Zhang Runxiang Yang Zongmin Fan Haiwen Lyu Junzhong Lin Kai Li Mong-Hong Lee 2023Signal Transduction and Targeted Therapy2023,8,6:0
12Bcl-3 promotes Wnt signaling by maintaining the acetylation ofβ-catenin at lysine 49 in colorectal cancer显示文摘Wnt/β-catenin signaling plays a critical role in colorectal cancer(CRC)tumorigenesis and the homeostasis of colorectal cancer stem cells(CSCs),but its molecular mechanism remains unclear.B-cell lymphoma 3(Bcl-3),a member of the IκB family,is overexpressed in CRC and promotes tumorigenicity.Here,we report a novel function of Bcl-3 in maintaining colorectal CSC homeostasis by activating Wnt/β-catenin signaling.Silencing Bcl-3 suppresses the self-renewal capacity of colorectal CSCs and sensitizes CRC cells to chemotherapeutic drugs through a decrease in Wnt/β-catenin signaling.Moreover,our data show that Bcl-3 is a crucial component of Wnt/β-catenin signaling and is essential forβ-catenin transcriptional activity in CRC cells.Interestingly,Wnt3a increases the level and nuclear translocation of Bcl-3,which binds directly toβ-catenin and enhances the acetylation ofβ-catenin at lysine 49(Ac-K49-β-catenin)and transcriptional activity.Bcl-3 depletion decreases the Ac-K49-β-catenin level by increasing the level of histone deacetylase 1 to remove acetyl groups fromβ-catenin,thus interrupting Wnt/β-catenin activity.In CRC clinical specimens,Bcl-3 expression negatively correlates with the overall survival of CRC patients.A significantly positive correlation was found between the expression of Bcl-3 and Ac-K49-β-catenin.Collectively,our data reveal that Bcl-3 plays a crucial role in CRC chemoresistance and colorectal CSC maintenance via its modulation of the Ac-K49-β-catenin,which serves as a promising therapeutic target for CRC.Xi Chen Chen Wang Yuhang Jiang Qi Wang Yu Tao Haohao Zhang Yongxu Zhao Yiming Hu Cuifeng Li Deji Ye Dandan Liu Wenxia Jiang Eugene Y.Chin Sheng Chen Yongzhong Liu Mingliang Wang Sanhong Liu Xiaoren Zhang 2020Signal Transduction and Targeted Therapy2020,5,1:0

14MDC-Net:a multi-directional constrained and prior assisted neural network for wood and leaf separation from terrestrial laser scanning显示文摘Wood-leaf separation from terrestrial laser scanning(TLS)is a crucial prerequisite for quantifying many biophysical properties and understanding ecological functions.In this study,we propose a novel multi-directional collaborative convolutional neural network(MDC-Net)that takes the original 3D coordinates and useful features from prior knowledge(prior features)as input,and outputs the semantic labels of TLS point clouds.The MDC-Net contains two key units:(1)a multi-directional neighborhood construction(MDNC)unit to obtain more representative neighbors and enable directionally aware feature encoding in the subsequent local feature extraction,to mitigate occlusion effects;(2)a collaborative feature encoding(CFE)unit is introduced to incorporate useful features from prior knowledge into the network through a collaborative cross coding to enhance the discrimination for thin structures(e.g.small branches and leaf).The MDC-Net is evaluated onfive plots from forests in Guangxi,China,with different branch architectures and leaf distributions.Experimental results showed that the MDC-Net achieved an OA of 0.973 and a mIoU of 0.821 and outperformed other related methods.We believe the MDC-Net would facilitate the usage of TLS in ecology studies for quantifying tree size and morphology and thus promote the development of relevant ecological applications.Wenxia Dai Yiheng Jiang Wen Zeng Ruibo Chen Yongyang Xu Ningning Zhu Wen Xiao Zhen Dong Qingfeng Guan 2023International Journal of Digital Earth2023,16,1:0
