| 1 | Genetic analysis and molecular mapping of a presenescing leaf gene psll in rice (Oryza sativa L.)显示文摘A rice psl1 (presenescing leaf) mutant was obtained from a japonica variety Zhonghua 11 via radiation of 60Co-γ in M2 generation. Every leaf of the mutant began to wither after it reached the big-gest length,while the leaves of the wild variety could keep green for 25―35 d. In this study,genetic analysis and gene mapping were carried out for the mutant identified. The SSR marker analysis showed that the mutant was controlled by a single recessive gene (psl1) located on chromosome 2. Fine mapping of the psl1 locus was conducted with 34 new STS markers developed around psl1 anchored region based on the sequence diversity between Nippon-bare and 93-11. The psl1 was further mapped be-tween two STS markers,STS2-19 and STS2-26,with genetic distances of 0.43 and 0.11 cM,respectively,while cosegregated with STS2-25. A BAC contig was found to span the psl1 locus,the region being delim-ited to 48 kb. | 3 | PA1b, a plant peptide, induces intracellular [Ca^(2+)] in- crease via Ca^(2+) influx through the L-type Ca^(2+) channel and triggers secretion in pancreatic β cells显示文摘Using alginic acid to adsorb polypeptides at pH 2.7, we isolated a peptide pea albumin 1b (PA1b) from pea seeds. The PA1b is a single chain peptide consisting of 37 amino acid residues with 6 cysteines which constitutes the cystine-knot structure. Using microfluorometry and patch clamp techniques, we found that PA1b significantly elevated the intracellular calcium level ([Ca2+ ]i) and elicited membrane capacitance increase in the primary rat pancreatic β cells. The PA1b effect on [Ca2+]i elevation was abolished in the absence of extracellular Ca2+ or in the presence of L-type Ca2+ channel blocker, ni- modipine. Interestingly, we found that PA1b significantly depolarized membrane potential, which could lead to the opening of voltage-dependent L-type Ca2+ channels and influx of extracellular Ca2+, and then evoke robust secretion. | 4 | Campus Vibration in Nanwangshan Campus,China University of Geosciences at Wuhan Monitored by Short-Period Seismometers显示文摘Continuous seismic observations can record seismic waveforms, and ambient noise, for the purposes of earthquake researches and other applications. Here we deploy three digital seismometers(EPS-2) in and around the Nanwangshan Campus of the China University of Geosciences(Wuhan). This network was running from April 9 to May 9 of 2018. During this period, the seismometers recorded the May 4, 2018 M6.9 Hawaii earthquake. From the recorded waveforms, we could observe clearly the P and S arrivals, and the corresponding particle motions. Analysis of continuous observations of ambient noise shows obvious fluctuation of vibration intensity inside of the campus. The campus is quietest from 0 to 5 am. From 5 am on, the vibration intensity increases, and reaches the peak of entire day at 12 am. The amplitude then decreases to a very low level at 19:30 to 20:00 pm, and reaches another strong noisy time at 21:00 to 21:30 pm. After 21:30 pm, the intensity goes down slowly. We also observed seismic signals that were generated by the interaction of speed-control hump cars and ground. By taking the envelope and smooth operations, we observe different characteristics for different car speeds, which suggests that seismic monitoring approaches can be used for speed measurement of cars. This kind of small seismic network running in a real time fashion, would greatly help understanding of the sources of ambient noise at high frequency bands in interested areas. | 14 | A New Method to Construct Education Knowledge Graph显示文摘Learning from the Internet is becoming more and more convenient and attracting more and more people. How to obtain knowledge from massive data and construct high quality knowledge graph has become a research hot topic. This paper proposes a new method of knowledge graph construction based on crowd-sourcing. Firstly, learners build the subgraphs to acquire knowledge through the crowd-sourcing task; secondly, we put forward the fusion strategy of knowledge subgraph, in which knowledge graph is converted into the adjacency matrix, and the weight of the knowledge relation is calculated by matrix operations, thus knowledge graph is constructed. | 18 | Optimization design of fertilizer apparatus owned arc gears based on discrete element method显示文摘To improve the uniformity of the flow when fertilizer apparatus discharges fertilizer,a kind of fertilizer apparatus owned arc gears was designed.Also,the design and working principle of its general structure and key components were analyzed theoretically.Aiming at exploring the influence of arc structural parameters to the performance of arc gears discharging fertilizer,DEM simulation was used in this study.Fertilizer apparatus owned arc gears was taken as the research object to analyze the influence of two key components including the arc radius of gears’concave-grooves and the minimal length between two mutual meshing gears.The variation coefficient of fertilizer discharging amount stability and the linear determined coefficient of fertilizer discharging amount in unit time as evaluation index were selected.Two factors and five levels quadratic rotation orthogonal experiment was applied,and Design-Expert 8.0 software was used to achieve data processing and analysis.The results of experiment indicated that the arc radius of gears’concave-grooves had significant effect on the linear determined coefficient of fertilizer discharging amount in unit time,and the minimal length between two mutual meshing gears had significant effect on the variation coefficient of fertilizer discharging amount stability.The optimum structural parameters were 8.54 mm as the arc radius of gears’concave-grooves and 5.22 mm as the minimal length between two mutual meshing gears.Upon this circumstance,the variation coefficient was 0.28 and determination coefficient was 0.9972.The optimum apparatus was selected to conduct the bench test and field simulation test.The results indicated that the variation coefficient is 0.27,and the linear determined coefficient of is 0.9980.The results of simulation experiment were anastomotic basically with the real result,which showed that DEM simulation technology was usable to the experiment of testing fertilizer discharging.The results of this research can provide a reference to the improving of performance in discharging fertilizer. | Guoqiang Dun Zhiyong Gao Yuxuan Liu Wenyi Ji Ning Mao Xingpeng Wu Wenhui Liu | 2021 | International Journal of Agricultural and Biological Engineering2021,14,2: | 1 |
