| 1 | PTPL1 suppresses lung cancer cell migration via inhibiting TGF-β1-induced activation of p38 MAPK and Smad 2/3 pathways and EMT显示文摘Epithelial-mesenchymal transition(EMT)enables dissemination of neoplastic cells and onset of distal metastasis of primary tumors.However,the regulatory mechanisms of EMT by microenvironmental factors such as transforming growth factor-β(TGF-β)remain largely unresolved.Protein tyrosine phosphatase L1(PTPL1)is a non-receptor protein tyrosine phosphatase that plays a suppressive role in tumorigenesis of diverse tissues.In this study we investigated the role of PTPL1/PTPN13 in metastasis of lung cancer and the signaling pathways regulated by PTPL1 in terms of EMT of non-small cell lung cancer(NSCLC)cells.We showed that the expression of PTPL1 was significantly downregulated in cancerous tissues of 23 patients with NSCLC compared with adjacent normal tissues.PTPL1 expression was positively correlated with overall survival of NSCLC patients.Then we treated A549 cells in vitro with TGF-β1(10 ng/mL)and assessed EMT.We found that knockdown of PTPL1 enhanced the migration and invasion capabilities of A549 cells,through enhancing TGF-β1-induced EMT.In nude mice bearing A549 cell xenografts,knockdown of PTPL1 significantly promoted homing of cells and formation of tumor loci in the lungs.We further revealed that PTPL1 suppressed TGF-β-induced EMT by counteracting the activation of canonical Smad2/3 and non-canonical p38 MAPK signaling pathways.Using immunoprecipitation assay we demonstrated that PTPL1 could bind to p38 MAPK,suggesting that p38 MAPK might be a direct substrate of PTPL1.In conclusion,these results unravel novel mechanisms underlying the regulation of TGF-βsignaling pathway,and have implications for prognostic assessment and targeted therapy of metastatic lung cancer. | Ning Zhu Xiu-juan Zhang Hai Zou Yuan-yuan Zhang Jing-wen Xia Peng Zhang You-zhi Zhang Jing Li Liang Dong Gulinuer Wumaier Sheng-qing Li | 2021 | Acta Pharmacologica Sinica2021,42,8: | 5 |

| 8 | 1H-NMR based metabolomics approach to evaluate effect of Cydonia oblanga Miller total flavonoid on atherosclerosis apoE(-/-) mice显示文摘OBJECTIVE ~1H-NMR-based metabolomics approach was conducted to holistically explore the effect and mechanisms of Cydonia oblanga Mill flavonoids(COMF) on high-fat diet induced Atherosclerosis(AS) apoE-/-mice.METHODS AS model was established on the apolipoprotein e knockout mice by high-fat diet.The ApoE-/-mice were split into 6 groups including control group,AS model group,COMF High dose(COMF-H) group,COMF medium dose(COMF-M) group,COMF Low dose(COMF-L) group and Simvastatin group as the positive control group.Serum samples from all groups were analyzed by ~1H-NMR technology and the OPLS-DA was conducted to distinguish the metabolic phenotypes.RESULTS Compared to the control group,serum levels of cholesterol,VLDL,leucine,isoleucine,valine,blood lipid,citrulline,methylamine,glucose,glycine,glycerol,myo-inositol,fructose,phenylalanine,unsaturated lipid,urea and other metabolites content significantly increased,while HDL,lactate,alanine,glutamate,glutamine,pyruvate,carnitine,citrate,choline content signifi.cantly decreased and the difference was statistically significant(P<0.05).The trend of metabolites in serum samples of COMF low,medium and high group was opposite to that of atherosclerosis model group and the difference was statistically significant(P<0.05).CONCLUSION Through functional analysis of these biomakers,amino acid metabolism,lipid metabolism,cholesterol metabolism,energy metabolism and inflammation reaction were considered as the most relevant pathological biomakers in the serum of AS mice.This study also demonstrates that COMF had the therapeutic effectiveness on AS through partly reversing the lipid,cholesterol,amino acid,energy metabolism and Inflammation reaction. | Ainiwaer WUMAIER Adili ABUDOUREHEMAN Wen-ting ZHOU | 2018 | 中国药理学与毒理学杂志2018,32,4: | 1 |

| 17 | Perspective Material for Photoenergetics on the Basis of Silicon with Binary Elementary Cells显示文摘The paper proposes a scientifically grounded - principally new approach to managing the fundamental parameters of the basic material of electronic engineering as like silicon. The essense of the proposed approach is that the formation of binary elementary cells in the silicon lattice involving elements III (B, Al, Ga, Zn) and V (P, As, Sb) groups in the form of Si2GaAs, Si2GaSb, etc., taking into account electrical and chemical parameters of these impurity atoms, as well as their diffusion parameters in Si, are determined by the most suitable pairs of atoms of groups III and V that allow obtaining silicon with the necessary composition and structure of binary elementary cells, as well as their more complex associations, up to the formation of nanocrystals of semiconductor connections AIIIBV. It is shown that by controlling the composition and structure, as well as the concentration of binary elementary cells, it is possible to significantly expand the spectral sensitivity of silicon, both in the IR and hλ> Eg directions. The formation of nanoclusters of AIIIBV semiconductor compounds in the silicon lattice significantly changes the emissivity of the material. It is established that the successive diffusion of elements of groups III and V in silicon and additional low-temperature annealing under certain thermodynamic conditions makes it possible to ensure the maximum participation of the impurity atoms introduced in the formation of binary elementary cells. It is shown that silicon with binary elementary cells involving atoms of groups III and V is a new class of semiconductor material with unique functionality for modern optoelectronics and photoenergetics. | M.K. | 19 | NDRG1 promotes endothelial dysfunction and hypoxia-induced pulmonary hypertension by targeting TAF15显示文摘Background:Pulmonary hypertension(PH)represents a threatening pathophysiologic state that can be induced by chronic hypoxia and is characterized by extensive vascular remodeling.However,the mechanism underlying hypoxia-induced vascular remodeling is not fully elucidated.Methods and Results:By using quantitative polymerase chain reactions,western blotting,and immunohistochemistry,we demon-strate that the expression of N-myc downstream regulated gene-1(NDRG1)is markedly increased in hypoxia-stimulated endothelial cells in a time-dependent manner as well as in human and rat endothelium lesions.To determine the role of NDRG1 in endothelial dysfunction,we performed loss-of-function studies using NDRG1 short hairpin RNAs and NDRG1 over-expression plasmids.In vitro,silencing NDRG1 attenuated proliferation,migration,and tube formation of human pulmonary artery endothelial cells(HPAECs)un-der hypoxia,while NDRG1 over-expression promoted these behaviors of HPAECs.Mechanistically,NDRG1 can directly interact with TATA-box binding protein associated factor 15(TAF15)and promote its nuclear localization.Knockdown of TAF15 abrogated the effect of NDRG1 on the proliferation,migration and tube formation capacity of HPAECs.Bioinformatics studies found that TAF15 was involved in regulating PI3K-Akt,p53,and hypoxia-inducible factor 1(HIF-1)signaling pathways,which have been proved to be PH-related pathways.In addition,vascular remodeling and right ventricular hypertrophy induced by hypoxia were markedly alleviated in NDRG1 knock-down rats compared with their wild-type littermates.Conclusions:Taken together,our results indicate that hypoxia-induced upregulation of NDRG1 contributes to endothelial dysfunction through targeting TAF15,which ultimately contributes to the development of hypoxia-induced PH. | Chengwei Li Junzhu Lv Gulinuer Wumaier Yu Zhao Liang Dong Yuzhen Zeng Ning Zhu Xiujuan Zhang Jing Wang Jingwen Xia Shengqing Li | 2023 | Precision Clinical Medicine2023,6,4: | 0 |
