1Improved Brassica rapa reference genome by single-molecule sequencing and chromosome conformation capture technologies显示文摘Brassica rapa comprises several important cultivated vegetables and oil crops.Current reference genome assemblies of Brassica rapa are quite fragmented and not highly contiguous,thereby limiting extensive genetic and genomic analyses.Here,we report an improved assembly of the B.rapa genome(v3.0)using single-molecule sequencing,optical mapping,and chromosome conformation capture technologies(Hi-C).Relative to the previous reference genomes,our assembly features a contig N50 size of 1.45 Mb,representing a~30-fold improvement.We also identified a new event that occurred in the B.rapa genome~1.2 million years ago,when a long terminal repeat retrotransposon(LTR-RT)expanded.Further analysis refined the relationship of genome blocks and accurately located the centromeres in the B.rapa genome.The B.rapa genome v3.0 will serve as an important community resource for future genetic and genomic studies in B.rapa.This resource will facilitate breeding efforts in B.rapa,as well as comparative genomic analysis with other Brassica species.Lei Zhang Xu Cai Jian Wu Min Liu Stefan Grob Feng Cheng Jianli Liang Chengcheng Cai Zhiyuan Liu Bo Liu Fan Wang Song Li Fuyan Liu Xuming Li Lin Cheng Wencai Yang Mai-he Li Ueli Grossniklaus Hongkun Zheng Xiaowu Wang 2018Horticulture Research2018,5,1:10
2A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. 4Deubiquitinase JOSD2 stabilizes YAP/TAZ to promote cholangiocarcinoma progression显示文摘Cholangiocarcinoma(CCA)has emerged as an intractable cancer with scanty therapeutic regimens.The aberrant activation of Yes-associated protein(YAP)and transcriptional co-activator with PDZ-binding motif(TAZ)are reported to be common in CCA patients.However,the underpinning mechanism remains poorly understood.Deubiquitinase(DUB)is regarded as a main orchestrator in maintaining protein homeostasis.Here,we identified Josephin domain-containing protein 2(JOSD2)as an essential DUB of YAP/TAZ that sustained the protein level through cleavage of polyubiquitin chains in a deubiquitinase activity-dependent manner.The depletion of JOSD2 promoted YAP/TAZ proteasomal degradation and significantly impeded CCA proliferation in vitro and in vivo.Further analysis has highlighted the positive correlation between JOSD2 and YAP abundance in CCA patient samples.Collectively,this study uncovers the regulatory effects of JOSD2 on YAP/TAZ protein stabilities and profiles its contribution in CCA malignant progression,which may provide a potential intervention target for YAP/TAZ-related CCA patients.Meijia Qian Fangjie Yan Weihua Wang Jiamin Du Tao Yuan Ruilin Wu Chenxi Zhao Jiao Wang Jiabin Lu Bo Zhang Nengming Lin Xin Dong Xiaoyang Dai Xiaowu Dong Bo Yang Hong Zhu Qiaojun He 2021Acta Pharmaceutica Sinica B2021,11,12:5

6Microstructural Characterization and Hardness Behavior of a Biological Saxidomus purpuratus Shell显示文摘The microstructures of the Saxidomus purpuratus shell were observed.It was found that the inner and middle layers of the shell are composed of crossed lamellae,while the outer layer exhibits porous structures.With the characteristic structure of each layer,the hardness of inner layer with narrow domains in crossed lamellar structure is the highest,and that of middle layer with wide domains is lower,while the outer layer has the lowest hardness.The damage morphologies of the indentations change a lot,depending not only upon the magnitude of the indentation load,but also upon the orientation between the indentation direction and the crossed lamellae in the microstructure of the shell,which illustrates the anisotropy in mechanical properties of such shells.Wen Yang Guangping Zhang Huasai Liu Xiaowu Li 2011Journal of Materials Science & Technology2011,27,2:3

17Comparison of the meat quality and fatty acid profile of muscles in finishing Xiangcun Black pigs fed varied dietary energy levels显示文摘To study the effects of dietary energy level on the meat quality of different muscles in finishing pigs,400Xiangcun Black pigs(BW=79.55±4.77 kg)were randomly assigned to 5 treatments with varied calculated digestive energy(DE)at 3,050,3,100,3,150,3,200 and 3,250 kcal/kg,respectively.Each treatment had 8replicates with 10 pigs per replicate.Meat quality,amino acid and fatty acid composition were tested in this study.No differences in average daily gain,average daily feed intake or feed-to-gain ratio(P>0.05)were observed among dietary treatments.Glycogen concentrations of longissimus dorsi(LD)muscle in DE3150was higher than those in other groups(P<0.05).The crude fat concentration of biceps femoris(BF)muscle in DE3250 tended to be higher than that in DE3150 and DE3100 groups(P<0.05).Pigs in DE3250 and DE3200 had higher fiber density and smaller cross-sectional area of BF muscle than those in DE3150(P<0.05).Pigs in DE3150 had the highest Cu concentration in LD muscle compared with those in DE3200,DE3250(P<0.05).The C16:1 proportion of LD muscle was lower(P<0.01)and C20:1 was higher(P<0.05)in DE3050 than that in the other dietary treatments.The C18:3n6 and C20:3n6 proportions of BF muscle in DE3150 were higher than those in DE 3050,DE3200 and DE3250(P<0.05).For LD muscle,m RNA expressions of type I and IIa MyHC in group DE3150 were higher than other treatments(P<0.01).The LD muscle in DE3150 expressed higher PPARd than in other groups(P<0.01).Pigs in DE3100 expressed higher FOX1 than in DE3200 and DE3250(P<0.05).Sterol-regulatory element binding proteins(SREBPa)mRNA expression decreased linearly when dietary energy level increased in BF muscle(P<0.01).In conclusion,a200 kcal/kg decrease in digestible energy for 4 consecutive weeks did not affect growth performance of Xiangcun Black pigs.Furthermore,LD and BF muscle respond differently to dietary energy level,and meat quality was improved by the medium energy level during the finishing phase.Can Yang Wenlong Wang Xiaowu Tang Ruilin Huang Fengna Li Wenxuan Su Yulong Yin Chaoyue Wen Jian Liu 2022Animal Nutrition2022,,4:1
18Structural changes of a bacteriophage upon DNA packaging and maturation显示文摘Dear Editor,Tailed,double-stranded DNA(dsDNA)bacteriophages,which belong to the order of Caudovirales,have a tail attached to a pentameric vertex of the icosahedral capsid shell(head)through a 12-fold portal(Johnson and Chiu,2007).The phages package genomic dsDNA into a round procapsid using the portal in complex with an ATP-dependent terminase complex as the motor.During packaging,the procapsid shell expands to a more angular intermediate to match the size of the viral genome(Guo et al.,2014).When the phage head is full,the portal detects internal pressure and conveys a signal from the inner capsid to the exterior,which triggers a sequence of events-the terminase complex cleaving mature DNA genome from a multi-genome concatemer,the release of the terminase complex from the portal,and the attachment of the tail complex-in the completion of phage assembly(Lander et al.,2006;Johnson and Chiu,2007).At the beginning of phage infection,the tail is responsible for receptor recognition,and the portal and tail act as a tunnel for DNA delivery into the host cytoplasm(Johnson and Chiu,2007).These mechanisms of DNA packaging and ejection may also be conserved in many other DNA viruses,including herpesvirus(Wang et al.,2020;Yang et al.,2020).Wenyuan Chen Hao Xiao Xurong Wang Shuanglin Song Zhen Han Xiaowu Li Fan Yang Li Wang Jingdong Song Hongrong Liu Lingpeng Cheng 2020Protein & Cell2020,11,5:1

20FeP nanoparticles derived from metal-organic frameworks/GO as high-performance anode material for lithium ion batteries显示文摘Double carbon coated Fe P composite(Fe P@NC@r GO)was in situ fabricated via the phosphorization process of the as-prepared Prussian blue@graphene oxide(PB@GO)precursor.The Fe P nanocrystals were successfully embedded in the nitrogen-doped porous carbon matrix.When used as the anode for lithium ion batteries(LIBs),the Fe P@NC@r GO anode shows superior lithium storage properties,delivering a high specific capacity of 830 m A h g^(-1)after 100 cycles at 100 m A g^(-1)and excellent rate capability of 359 m A h g^(-1)at 5 A g^(-1).The outstanding performance mainly ascribes to the synergistic effect of the double carbon coating and porous structure design.The introduction of porous carbon and graphene coating on Fe P nanoparticles greatly enhance the electronic conductivity of the active material and well accommodates the large volume variation of Fe P during the cycling process.Man Gao Xiaowu Liu Hai Yang Yan Yu 2018Science China Chemistry2018,61,9:1
