1Compound Danshen injection improves endotoxin-induced microcirculatory disturbance in rat mesentery显示文摘AIM: To investigate the effect of compound Danshen injection on lipopolysaccharide (LPS)-induced rat mesenteric microcirculatory dysfunctions and the underlying possible mechanism by an inverted intravital microscope and high-speed video camera system. METHODS: LPS was continuously infused through the jugular artery of male Wistar rats at the dose of 2 mg/kg per hour. Changes in mesenteric microcirculation,such as diameters of arterioles and venules,velocity of RBCs in venules,leukocyte rolling,adhesion and emigration,free radicals released from post-capillary venules,FITC- albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope assisted with CCD camera and SIT camera. Meanwhile,the expression of adhesion molecules CD11b/CD18 and the production of free radical in neutrophils,and the expression of intercellular adhesion molecule 1 (ICAM-1) in human umbilical vein endothelial cells (HUVECs) were quantified by flow cytometry (FACS) in vitro. RESULTS: The continuous infusion with LPS resulted in a number of responses in microcirculation,including a significant increase in the positive region of venulestained with Monastral blue B,rolling and adhesion of leukocytes,production of oxygen radical in venular wall,albumin efflux and enhanced mast cell degranulation in vivo,all of which,except for the leukocyte rolling,were attenuated by the treatment with compound Danshen injection. Experiments performed in vitro further revealed that the expression of CD11b/CD18 and the production of oxygen free radical in neutrophils,and the expression of ICAM-1 in HUVECs were increased by exposure to LPS,and they were attenuated by compound Danshen injection. 2Atomically dispersed Au_1 catalyst towards efficient electrochemical synthesis of ammonia显示文摘Tremendous efforts have been devoted to explore energy-efficient strategies of ammonia synthesis to replace Haber–Bosch process which accounts for 1.4% of the annual energy consumption. In this study,atomically dispersed Au_1 catalyst is synthesized and applied in electrochemical synthesis of ammonia under ambient conditions. A high NH_4^+ Faradaic efficiency of 11.1% achieved by our Au_1 catalyst surpasses most of reported catalysts under comparable conditions. Benefiting from efficient atom utilization, an NH_4^+ yield rate of 1,305 lg h^(-1) mg_(Au)^(-1) has been reached, which is roughly 22.5 times as high as that by supported Au nanoparticles. We also demonstrate that by employing our Au_1 catalyst, NH4+can be electrochemically produced directly from N_2 and H_2 with an energy utilization rate of 4.02 mmol kJ^(-1). 3High levels of serum interleukin-6 increase mortality of hepatitis B virus-associated acute-on-chronic liver failure显示文摘BACKGROUND Patients with hepatitis B virus-associated acute-on-chronic liver failure(HBVACLF)present a complex and poor prognosis.Systemic inflammation plays an important role in its pathogenesis,and interleukin-6(IL-6)as a pro-inflammatory cytokine is related with severe liver impairment and also plays a role in promoting liver regeneration.Whether serum IL-6 influences HBV-ACLF prognosis has not been studied.AIM To determine the impact of serum IL-6 on outcome of patients with HBV-ACLF.METHODS We performed a retrospective study of 412 HBV-ACLF patients.The findings were analyzed with regard to mortality and the serum IL-6 level at baseline,as well as dynamic changes of serum IL-6 within 4 wk.RESULTS The serum IL-6 level was associated with mortality.Within 4 wk,deceased patients had significantly higher levels of IL-6 at baseline than surviving patients[17.9(7.3-57.6)vs 10.4(4.7-22.3),P=0.011].Patients with high IL-6 levels(>11.8 pg/mL)had a higher mortality within 4 wk than those with low IL-6 levels(≤11.8 pg/mL)(24.2%vs 13.2%,P=0.004).The odds ratios calculated using univariate and multivariate logistic regression were 2.10(95%confidence interval[CI]:1.26-3.51,P=0.005)and 2.11(95%CI:1.15-3.90,P=0.017),respectively.The mortality between weeks 5 and 8 in patients with high IL-6 levels at 4 wk was 15.0%,which was significantly higher than the 6.6%mortality rate in patients with low IL-6 levels at 4 wk(hazard ratio=2.39,95%CI:1.05-5.41,P=0.037).The mortality was 5.0%in patients with high IL-6 levels at baseline and low IL-6 levels at 4 wk,7.5%in patients with low IL-6 levels both at baseline and at 4 wk,11.5%in patients with low IL-6 levels at baseline and high IL-6 levels at 4 wk,and 16.7%in patients with high IL-6 levels both at baseline and at 4 wk.The increasing trend of the mortality rate with the dynamic changes of IL-6 was significant(P for trend=0.023).CONCLUSION A high level of serum IL-6 is an independent risk factor for mortality in patients with HBV-ACLF.Furthermore,a sustained high level or dynamic elevated level of serum IL-6 indicates a higher mortality.Chao Zhou Ning Zhang Ting-Ting He Yao Wang Li-Fu Wang Yong-Qiang Sun Jing Jing Jing-Jing Zhang Shuang-Nan Fu Xuan Wang Xiao-Xiao Liang Xin Li Man Gong Jun Li 2020World Journal of Gastroenterology2020,26,30:15

5Enriched environment enhances histone acetylation of NMDA receptor in the hippocampus and improves cognitive dysfunction in aged mice显示文摘The mechanisms of age-associated memory impairment may be associated with glutamate receptor function and chromatin modification.To observe the effect of an enriched environment on the cognitive function of mice with age-associated memory impairment,3-monthold C57BL/6 male mice('young'mice)were raised in a standard environment,while 24-month-old C57BL/6 male mice with memory impairment('age-associated memory impairment'mice)were raised in either a standard environment or an enriched environment.The enriched environment included a variety of stimuli involving movement and sensation.A water maze test was then used to measure cognitive function in the mice.Furthermore,quantitative real-time polymerase chain reaction and western blot assays were used to detect right hippocampal GluN2B mRNA as well as protein expression of GluN2B and CREB binding protein in all mice.In addition,chromatin immunoprecipitation was used to measure the extent of histone acetylation of the hippocampal GluN2B gene promoters.Compared with the young mice,the water maze performance of age-associated memory impairment mice in the standard environment was significantly decreased.In addition,there were significantly lower levels of total histone acetylation and expression of CREB binding protein in the hippocampus of age-associated memory impairment mice in the standard environment compared with the young mice.There were also significantly lower levels of histone acetylation,protein expression,and mRNA expression of GluN2B in the hippocampus of these mice.In contrast,in the age-associated memory impairment mice with the enriched environment intervention,the water maze performance and molecular biological indexes were significantly improved.These data confirm that an enriched environment can improve cognitive dysfunction in age-associated memory impairment mice,and suggest that the mechanisms may be related to the increased expression of CREB binding protein and the increased degree of total histone acetylation in the hippocampus of age-associated memory impairment mice,which may cause the increase of histone acetylation of GluN2B gene promoter and the enhancement of GluN2B mRNA transcription and protein expression in hippocampus.The animal experiment was approved by the Animal Ethics Committee of Yangzhou University,China(approval No.20170312001)in March 2017.Xin Wang Zhao-Xiang Meng Ying-Zhu Chen Yu-Ping Li Hong-Yu Zhou Man Yang Ting-Ting Zhao Yu-Lai Gong Yi Wu Tao Liu 2020Neural Regeneration Research2020,15,12:8
6Lack of association between apolipoprotein C3 gene polymorphisms and risk of nonalcoholic fatty liver disease in a Chinese Han population显示文摘AIM:To investigate the association between two polymorphisms of apolipoprotein C3(APOC3)and risk of nonalcoholic fatty liver disease(NAFLD)in a Chinese Han population.METHODS:Genotypes for rs2854116 and rs2854117in APOC3 and the known rs738409 in patatin-like phospholipase domain-containing protein 3(PNPLA3)in 390patients with NAFLD and 409 control subjects were determined by sequencing and polymerase chain reaction analysis.Serum lipid profiles were determined using biochemical methods,and an index of insulin resistance(IR,HOMA-IR),serum APOC3 concentrations and total antioxidant status(TAS)were also assessed.RESULTS:No significant differences in genotype and allele frequencies of rs2854116 and rs2854117 were found between the NAFLD population and the controls(P>0.05).The OR for the association between-455Cand-482T allele carriers and the risk of NAFLD were1.06(95%CI:0.72-1.57,P>0.05)and 1.00(95%CI:0.68-1.48,P>0.05),respectively.The variant carriers did not have a significantly increased risk of NAFLD or elevated clinical and biochemical parameters such as APOC3 concentrations,IR(1.42±0.43 vs 1.48±0.52,P>0.05),liver enzymes and TAS(13.94±2.01vs 14.38±1.92,P>0.05)compared with the controls.Moreover,the results were similar when testing was carried out independent of the genetic variation in PNPLA3.CONCLUSION:The two polymorphisms of the APOC3gene are not associated with a risk of NAFLD,or with lipid profiles,IR and oxidative stress in the Chinese Han population.Tong-Hong Niu Man Jiang Yong-Ning Xin Xiang-Jun Jiang Zhong-Hua Lin Shi-Ying Xuan 2014World Journal of Gastroenterology2014,20,13:7
7PNPLA3 I148M variant in nonalcoholic fatty liver disease:Demographic and ethnic characteristics and the role of the variant in nonalcoholic fatty liver fibrosis显示文摘Patatin-like phospholipase domain-containing 3(PNPLA3 or adiponutrin) displays anabolic and catabolic activities in lipid metabolism,and has been reported to be significantly associated with liver fat content.Variousstudies have established a strong link between the 148 isoleucine to methionine protein variant(I148M) of PNPLA3 and liver diseases,including nonalcoholic fatty liver disease(NAFLD).However,detailed demographic and ethnic characteristics of the I148 M variant and its role in the development of nonalcoholic fatty liver fibrosis have not been fully elucidated.The present review summarizes the current knowledge on the association between the PNPLA3 I148 M variant and NAFLD,and especially its role in the development of nonalcoholic fatty liver fibrosis.First,we analyze the impact of demographic and ethnic characteristics of the PNPLA3 I148 M variant and the presence of metabolic syndrome on the association between PNPLA3 I148 M and NAFLD.Then,we explore the role of the PNPLA3 I148 M in the development of nonalcoholic fatty liver fibrosis,and hypothesize the underlying mechanisms by speculating a pro-fibrogenic network.Finally,we briefly highlight future research that may elucidate the specific mechanisms of the PNPLA3 I148 M variant in fibrogenesis,which,in turn,provides a theoretical foundation and valuable experimental data for the clinical management of nonalcoholic fatty liver fibrosis.Li-Zhen Chen Yong-Ning Xin Ning Geng Man Jiang Ding-Ding Zhang Shi-Ying Xuan 2015World Journal of Gastroenterology2015,21,3:6
8Adaptive optical focusing through perturbed scattering media with a dynamic mutation algorithm显示文摘Optical imaging through or inside scattering media, such as multimode fiber and biological tissues, has a significant impact in biomedicine yet is considered challenging due to the strong scattering nature of light. In the past decade, promising progress has been made in the field, largely benefiting from the invention of iterative optical wavefront shaping, with which deep-tissue high-resolution optical focusing and hence imaging becomes possible. Most of the reported iterative algorithms can overcome small perturbations on the noise level but fail to effectively adapt beyond the noise level, e.g., sudden strong perturbations. Reoptimizations are usually needed for significant decorrelation to the medium since these algorithms heavily rely on the optimization performance in the previous iterations. Such ineffectiveness is probably due to the absence of a metric that can gauge the deviation of the instant wavefront from the optimum compensation based on the concurrently measured optical focusing.In this study, a square rule of binary-amplitude modulation, directly relating the measured focusing performance with the error in the optimized wavefront, is theoretically proved and experimentally validated. With this simple rule, it is feasible to quantify how many pixels on the spatial light modulator incorrectly modulate the wavefront for the instant status of the medium or the whole system. As an example of application, we propose a novel algorithm, the dynamic mutation algorithm, which has high adaptability against perturbations by probing how far the optimization has gone toward the theoretically optimal performance. The diminished focus of scattered light can be effectively recovered when perturbations to the medium cause a significant drop in the focusing performance, which no existing algorithms can achieve due to their inherent strong dependence on previous optimizations. 9Cultivation of microalgae for biodiesel production:A review on upstream and downstream processing显示文摘Fluctuating market price of fossil fuel and overwhelming emission of greenhouse gases to the atmosphere have resulted in climate change and have been a global concern in this decade. Hence, biodiesel has become an alternative option to fossil diesel as it is renewable and environmentally friendly. Nevertheless, this alternative fuel that is usually derived from terrestrial oil crops will cause shortage in food supply and deforestation if mass production is realized. In recent years, cultivation of aquatic microorganism(particularly microalgae) to produce biodiesel is considered as a practical solution due to their high growth rate and ability to synthesize large quantity of lipid within their cell. However, the development of energy and cost-efficiency of microalgae cultivation system are the main issues in producing renewable microalgae biodiesel. Of late, wastewater or organic compost has been used as the cultivation medium as it can provide sufficient nutrients to sustain microalgae growth.Microalgae cultivation method and system are vitally important as these factors undoubtedly affect the final microalgae biomass and lipid yield. In this review, the cultivation system of microalgae, nutrients demanded for microalgae production, cell harvesting and drying, microalgae oil extraction, and utilization of microalgae biomass for biodiesel production are introduced and discussed. 10Cytoreductive surgery and hyperthermic intraperitoneal chemotherapy in treatment of gastric cancer with peritoneal carcinomatosis显示文摘Although gastric cancer with peritoneal carcinomatosis is associated with poor prognosis and is generally treated with palliative systemic therapy, recent studies have shown that cytoreductive surgery(CRS) and hyperthermic intraperitoneal chemotherapy(HIPEC) may prove to be an efficacious treatment option. In addition to reviewing the natural history of gastric cancer with peritoneal carcinomatosis, this mini-review examines literature on the efficacy of CRS and HIPEC as compared to chemotherapy and surgical options. Both randomized and nonrandomized studies were summarized with the emphasis focused on overall survival. 122,3,7,8-Tetrachlorodibenzo-p-dioxin Promotes Proliferation of Astrocyte Cells via the Akt/STAT3/Cyclin D1 Pathway显示文摘Objective The compound 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD), a persistent organic pollutant, is harmful to the nervous system, but its effects on the brain are still unclear. This study aimed to investigate the effects of TCDD on astrocytes proliferation and underlying molecular mechanism. Methods The cell proliferation was measured by EdU-based proliferation assay and PI staining by flow cytometry. Protein expression levels were detected by Western blotting. Immunofluorescence, cytoplasmic and nuclear fractions separation were used to assess the distribution of signal transducer and activator of transcription 3(STAT3). Results C6 cells treated with 10 and 50 nmol/L TCDD for 24 h showed significant promotion of the proliferation of. The exposure to TCDD resulted in the upregulation in the expression levels of phosphorylated protein kinase B(p-Akt), phosphorylated STAT3, and cyclin D1 in a dose-and time-dependent manner. The inhibition of Akt expression with LY294002 or STAT3 expression with AG490 abolished the TCDD-induced cyclin D1 upregulation and cell proliferation. Furthermore, LY294002 suppressed the activation of STAT3. Finally, TCDD promoted the translocation of STAT3 from the cytoplasm to the nucleus, and LY294002 treatment blocked this effect. 13Lichtenstein inguinal hernia repairs with porcine small intestine submucosa: a 5- year follow-up. a prospective randomized controlled study显示文摘Porcine small intestine submucosa(SIS)biologic patch has been used in inguinal hernia repair.However,there are lttle data available to assess the long-term effect after repair.This study aimed to explore the long-term effect of SIS patch in open inguinal hernia repair.Sevent-six patients with unilateral inguinal hernia were treated with Lichtenstein tension-free hernia repair using SIS patch(Beijing Datsing Bio-Tech Co,Ltd.)and Surgisis patch(COOK,USA)in Tianjn Union Medical Center and China-Japan Friendship Hospital.In the trial,the long-term efficacy of the treatment group and the control group were compared.A total of 66 patients in both groups received long-term follow-up(>5years)after surgery,with a followup rate of 86.8%.During the follow-up period,there was one case of recurrence,one case of chronic pain in the control group.There was no statistically significant difference(P>0.05)in terms of recurrence,chronic pain,foreign body sensation and infection between the two groups of patients.After long-term observations,it has been found that the porcine small intestinal submucosa(SIS)biological patch is safe and effective for inguinal hernia Lichtenstein repair,and has a low recurrence rate and complication rate.Baoshan Li Xin Zhang Yi Man Jiadong Xie Wei Hu Huang Huang Yinlong Wang Hongguang Ma 2021Regenerative Biomaterials2021,8,1:5
14Comprehensive model for a slag bath in electroslag remelting process with a current-conductive mould显示文摘A mathematical model was developed to describe the interaction of multiple physical fields in a slag bath during electroslag remelting (ESR) process with a current-conductive mould. The distributions of current density, magnetic induction intensity, electromagnetic force, Joule heating, fluid flow and temperature were simulated. The model was verified by temperature measurements during remelting 12CrMoVG steel with a slag of 50wt%-70wt% CaF2, 20wt%-30wt% CaO, 10wt%-20wt% Al2O3, and ≤10wt% SiO2 in a 600 mm diameter current-conductive mould. There is a good agreement between the calculated temperature results and the measured data in the slag bath. The calculated results show that the maximum values of current density, electromagnetic force and Joule heating are in the region between the corner electrodes and the conductivity element. 17Plasma membrane-localized SlSWEET7a and SlSWEET14 regulate sugar transport and storage in tomato fruits显示文摘Sugars,especially glucose and fructose,contribute to the taste and quality of tomato fruits.These compounds are translocated from the leaves to the fruits and then unloaded into the fruits by various sugar transporters at the plasma membrane.SWEETs,are sugar transporters that regulate sugar efflux independently of energy or pH.To date,the role of SWEETs in tomato has received very little attention.In this study,we performed functional analysis of SlSWEET7a and SlSWEET14 to gain insight into the regulation of sugar transport and storage in tomato fruits.SlSWEET7a and SlSWEET14 were mainly expressed in peduncles,vascular bundles,and seeds.Both SlSWEET7a and SlSWEET14 are plasma membrane-localized proteins that transport fructose,glucose,and sucrose.Apart from the resulting increase in mature fruit sugar content,silencing SlSWEET7a or SlSWEET14 resulted in taller plants and larger fruits(in SlSWEET7a-silenced lines).We also found that invertase activity and gene expression of some SlSWEET members increased,which was consistent with the increased availability of sucrose and hexose in the fruits.Overall,our results demonstrate that suppressing SlSWEET7a and SlSWEET14 could be a potential strategy for enhancing the sugar content of tomato fruits.Xinsheng Zhang Chaoyang Feng Manning Wang Tianlai Li Xin Liu Jing Jiang 2021Horticulture Research2021,8,1:4
18Therapeutic potential of traditional Chinese medicine for the treatment of NAFLD:A promising drug Potentilla discolor Bunge显示文摘Nonalcoholic fatty liver disease(NAFLD)is characterized by excessive accumulation of hepatic lipids and metabolic stress-induced liver injury.There are currently no approved effective pharmacological treatments for NAFLD.Traditional Chinese medicine(TCM)has been used for centuries to treat patients with chronic liver diseases without clear disease types and mechanisms.More recently,TCM has been shown to have unique advantages in the treatment of NAFLD.We performed a systematic review of the medical literature published over the last two decades and found that many TCM formulas have been reported to be beneficial for the treatment of metabolic dysfunctions,including Potentilla discolor Bunge(PDB).PDB has a variety of active compounds,including flavonoids,terpenoids,organic acids,steroids and tannins.Many compounds have been shown to exhibit a series of beneficial effects for the treatment of NAFLD,including anti-oxidative and anti-inflammatory functions,improvement of lipid metabolism and reversal of insulin resistance.In this review,we summarize potential therapeutic effects of TCM formulas for the treatment of NAFLD,focusing on the medicinal properties of natural active compounds from PDB and their underlying mechanisms.We point out that PDB can be classified as a novel candidate for the treatment and prevention of NAFLD.Longshan Ji Qian Li Yong He Xin Zhang Zhenhua Zhou Yating Gao Miao Fang Zhuo Yu Robim M.Rodrigues Yueqiu Gao Man Li 2022Acta Pharmaceutica Sinica B2022,12,9:4
19Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. 20Relatedness of Helicobacter pylori populations to gastric carcinogenesis显示文摘Helicobacter pylori(H.pylori) is a Gram-negative bacterium that infects half of the human population.The infection is associated with chronic inflammation of the gastric mucosa and peptic ulcers.It is also a major risk factor for gastric cancer.Phylogenetic analysis of global strains reveals there are seven populations of H.pylori,including hpAfrica1,hpAfrica2,hpEastAsia,hpEurope,hpNEAfrica,hpAsia2 and hpSahul.These populations are consistent with their geographical origins,and possibly result from geographical separation of the bacterium leading to reduced bacterial recombination in some populations.For each population,H.pylori has evolved to possess genomic contents distinguishable from others.The hpEurope population is distinct in that it has the largest genome of 1.65 mbp on average,and the highest number of coding sequences.This confers its competitive advantage over other populations but at the cost of a lower infection rate.The large genomic size could be a cause of the frequent occurrence of the deletion of the cag pathogenicity island in H.pylori strains from hpEurope.The incidence of gastric cancer varies among different geographical regions.This can be attributed in part to different rates of infection of H.pylori.Recent studies found that different populations of H.pylori vary in their carcinogenic potential and contribute to the variation in incidence of gastric cancer among geographical regions.This could be related to the ancestral origin of H.pylori.Further studies are indicated to investigate the bacterial factors contributing to differential virulence and their influence on the clinical features in infected individuals.Quan-Jiang Dong Shu-Hui Zhan Li-Li Wang Yong-Ning Xin Man Jiang Shi-Ying Xuan 2012World Journal of Gastroenterology2012,18,45:3
