Simulation methodology development for rotating blade containment analysis显示文摘An experimental and numerical investigation on the aeroengine blade/case containment analysis is presented. Blade out containment capability analysis is an essential step in the new aeroengine design, but containment tests are time-consuming and incur significant costs; thus, developing a short-period and low-cost numerical method is warranted. Using explicit nonlinear dynamic finite element analysis software, the present study numerically investigated the high-speed impact process for simulated blade containment tests which were carried out on high-speed spin testing facility. A number of simulations were conducted using finite element models with different mesh sizes and different values of both the contact penalty factor and the friction coefficient. Detailed comparisons between the experimental and numerical results reveal that the mesh size and the friction coefficient have a considerable impact on the results produced. It is shown that a finer mesh will predict lower containment capability of the case, which is closer to the test data. A larger value of the friction coefficient also predicts lower containment capability. However, the contact penalty factor has little effect on the simulation results if it is large enough to avoid false penetration.Qing HE Hai-jun XUAN Lian-fang LIAO Wei-rong HONG Rong-ren WU 2012Journal of Zhejiang University-Science A(Applied Physics & Engineering)2012,13,4:9
Alpl prevents bone ageing sensitivity by specifically regulating senescence and differentiation in mesenchymal stem cells显示文摘Mutations in the liver/bone/kidney alkaline phosphatase(Alpl) gene cause hypophosphatasia(HPP) and early-onset bone dysplasia, suggesting that this gene is a key factor in human bone development. However, how and where Alpl acts in bone ageing is largely unknown. Here, we determined that ablation of Alpl induces prototypical premature bone ageing characteristics, including bone mass loss and marrow fat gain coupled with elevated expression of p16^(INK4A)(p16) and p53 due to senescence and impaired differentiation in mesenchymal stem cells(MSCs). Mechanistically, Alpl deficiency in MSCs enhances ATP release and reduces ATP hydrolysis. Then, the excessive extracellular ATP is, in turn, internalized by MSCs and causes an elevation in the intracellular ATP level, which consequently inactivates the AMPKα pathway and contributes to the cell fate switch of MSCs. Reactivating AMPKα by metformin treatment successfully prevents premature bone ageing in Alpl+/-mice by improving the function of endogenous MSCs.These results identify a previously unknown role of Alpl in the regulation of ATP-mediated AMPKα alterations that maintain MSC stemness and prevent bone ageing and show that metformin offers a potential therapeutic option.Wenjia Liu Liqiang Zhang Kun Xuan Chenghu Hu Shiyu Liu Li Liao Bei Li Fang Jin Songtao Shi Yan Jin 2018Bone Research2018,6,4:9
Relationship between higher-order aberrations and myopia progression in schoolchildren: a retrospective study显示文摘AIM: To investigate the relationship between higher- order aberration (HOA) and myopic progression in schoolchildren. ·METHODS: Between April 23 2011 and August 29, 2011 in the children's myopia outpatient clinic of the West China Hospital of Sichuan University, 148 eyes of 74 schoolchildren were reviewed. HOAs for a 6 -mm pupil were measured with an aberrometer. Myopic progression rate was defined according to the change in spherical equivalent refraction (SER) divided by the time span (years). Subjects with myopic progression rate of ≥0.50 diopters (D) were classified as the 'fast'group and the subjects with myopic progression rate of <0.50D were classified as the'slow'group. A retrospective study was conducted to compare HOA between the two groups, using root mean square (RMS) values and Zernike coefficients. ·RESULTS: The RMS values of HOA (t =2.316, P =0.02), HOA without Z 4 0 (t =2.224, P =0.03),third-order aberrations (t' =2.62, P =0.01), and coma (t' =2.49, P =0.01) were significantly higher in the fast group than those in the slow group. The individual Zernike coefficients of Z 3 -1 (t = -2.072, P =0.04) and Z 5 1 (Z =-2.627, P =0.01) displayed statistically significant differences between the two groups. Significant correlations were found between the RMS values of HOA (r =0.193, P =0.019), RMS values of HOA without Z 4 0 (r =0.23, P =0.005), RMS values of coma(r =0.235,P =0.004),RMS values of third-order aberrations (r =0.243, P =0.003), and the progression rate. · CONCLUSION: Our results provide evidence of a relationship between HOA and myopic progression. In a future prospective longitudinal study, we aim to verify whether HOA is a risk factor for myopic progression.Ning Zhang Xu-Bo Yang Wen-Qiu Zhang Long-Qian Liu Guang-Jing Dong Tao-Wen Chen Meng Liao Xuan Liao 2013International Journal of Ophthalmology(English edition)2013,6,3:7
Differences in clinical features of Crohn's disease and intestinal tuberculosis显示文摘AIM: To investigate the clinical features of Crohn's disease(CD) and intestinal tuberculosis(ITB) with a scoring system that we have developed.METHODS: A total of 25 CD and 40 ITB patients were prospectively enrolled from August 2011 to July 2012.Their characteristics and clinical features were recorded. Laboratory, endoscopic, histologic and radiographic features were determined. The features with a high specificity were selected to establish a scoring system. The features supporting CD scored +1, and those supporting ITB scored-1; each patient received a final total score. A receiver operating characteristic(ROC) curve was used to determine the best cut-off value for distinguishing CD from ITB.RESULTS: Based on a high specificity of differentiating between CD and ITB, 12 features, including longitudinal ulcers, nodular hyperplasia, cobblestone-like mucosa, intestinal diseases, intestinal fistula, the target sign, the comb sign, night sweats, the purified protein derivative test, the interferon-γ release assay(T-SPOT.TB), ring ulcers and ulcer scars, were selected for the scoring system. The results showed that the average total score of the CD group was 3.12 ± 1.740, the average total score of the ITB group was-2.58 ± 0.984, the best cutoff value for the ROC curve was-0.5, and the diagnostic area under the curve was 0.997, which was statistically significant(P < 0.001). The patients whose total scores were higher than-0.5 were diagnosed with CD; otherwise, patients were diagnosed with ITB. Overall, the diagnostic accuracy rate and misdiagnosis rate of this scoring system were 97% and 3%, respectively. CONCLUSION: Some clinical features are valuable for CD and ITB diagnosis. The described scoring system is key to differentiating between CD and ITB.Xin Huang Wang-Di Liao Chen Yu Yi Tu Xiao-Lin Pan You-Xiang Chen Nong-Hua Lv Xuan Zhu 2015World Journal of Gastroenterology2015,21,12:6
Pharmacokinetics-based identification of pseudoaldosterogenic compounds originating from Glycyrrhiza uralensis roots(Gancao)after dosing LianhuaQingwen capsule显示文摘LianhuaQingwen capsule, prepared from an herbal combination,is officially recommended as treatment for COVID-19 in China.Of the serial pharmacokinetic investigations we designed to facilitate identifying LianhuaQingwen compounds that are likely to be therapeutically important, the current investigation focused on the component Glycyrrhiza uralensis roots(Gancao).Besides its function in COVID-19 treatment,Gancao is able to induce pseudoaldosteronism by inhibiting renal 11β-HSD2.Systemic and colon-luminal exposure to Gancao compounds were characterized in volunteers receiving LianhuaQingwen and by in vitro metabolism studies.Access of Gancao compounds to 11β-HSD2 was characterized using human/rat,in vitro transport, and plasma protein binding studies, while 11β-HSD2 inhibition was assessed using human kidney microsomes.LianhuaQingwen contained a total of 41 Gancao constituents(0.01-8.56μmol/day).Although glycyrrhizin(1),licorice saponin G2(2),and liquiritin/liquiritin apioside(21/22)were the major Gancao constituents in LianhuaQingwen, their poor intestinal absorption and access to colonic microbiota resulted in significant levels of their respective deglycosylated metabolites glycyrrhetic acid(8),24-hydroxyglycyrrhetic acid(M2_(D);a new Gancao metabolite),and liquiritigenin(27)in human plasma and feces after dosing. These circulating metabolites were glucuronized/sulfated in the liver and then excreted into bile.Hepatic oxidation of 8 also yielded M2_(D).Circulating 8 and M2D,having good membrane permeability, could access(via passive tubular reabsorption)and inhibit renal 11β-HSD2.Collectively,1 and 2 were metabolically activated to the pseudoaldosterogenic compounds 8 and M2_(D).This investigation, together with such investigations of other components, has implications for precisely defining therapeutic benefit of LianhuaQingwen and conditions for its safe use.Xiao-fang Lan Olajide EOlaleye Jun-lan Lu Wei Yang Fei-fei Du Jun-ling Yang Chen Cheng Yan-hong Shi Feng-qing Wang Xue-shan Zeng Nan-nan Tian Pei-wei Liao Xuan Yu Fang Xu Ying-fei Li Hong-tao Wang Nai-xia Zhang Wei-wei Jia Chuan Li 2021Acta Pharmacologica Sinica2021,42,12:6

Penetration of disk fragments following impact on thin plate显示文摘To investigate the ballistic resistance and failure pattern of aeroengine casing following the impact of disk fragments, and to determine the optimum case structure, the phenomena of a 1/3rd disk fragment impact on single and double-layered thin plate targets were simulated using nonlinear dynamical analysis software MSC.Dytran. Strain rate effect was introduced in a Johnson-Cook (JC) material model for the disk fragment and the plate. Impact modeling was based on the Arbitrary Lagrange-Eulerian method, and simulated using explicit finite element method (FEM). Simulation results showed that the major failure pattern of the plate is shearing and tensile fracture with large plastic deformation. It was also concluded that the ballistic limit velocity increases with the standoff distance when it is beyond a certain value, and that greater resistance is obtained when the front plate has either a proportionately low or high thickness. The impact resistance of a double-layered plate may exceed that of a single plate if the thicknesses and standoff distance of the two plates are set appropriately.Juan-juan LI Hai-jun XUAN Lian-fang LIAO Wei-rong HONG Rong-ren WU 2009Journal of Zhejiang University-Science A(Applied Physics & Engineering)2009,10,5:4
Characteristics of mucosa-associated gut microbiota during treatment in Crohn's disease显示文摘BACKGROUND The dysbiosis of the gut microbiome is evident in Crohn's disease(CD) compared with healthy controls(HC), although the alterations from active CD to remission after treatment are unclear.AIM To characterize the mucosa-associated gut microbiota in patients with CD before and after the induction therapy.METHODS The basic information was collected from the subjects and the CD activity index(CDAI) was calculated in patients. A 16S rRNA sequencing approach was applied to determine the structures of microbial communities in mucosal samples including the terminal ileal, ascending colon, descending colon and rectum. The composition and function of mucosa-associated gut microbiota were compared between samples from the same cohort of patients before and after treatment.Differential taxa were identified to calculate the microbial dysbiosis index(MDI)and the correlation between MDI and CDAI was analyzed using Pearson correlation test. Predictive functional profiling of microbial communities was obtained with PICRUSt.RESULTS There were no significant differences in microbial richness among the four anatomical sites in individuals. Compared to active disease, the alpha diversity of CD in remission was increased towards the level of HC compared to the active stage. The principal coordinate analysis revealed that samples of active CD were clearly separated from those in remission, which clustered close to HC. Sixty-five genera were identified as differentially abundant between active and quiescent CD, with a loss of Fusobacterium and a gain of potential beneficial bacteria including Lactobacillus, Akkermansia, Roseburia, Ruminococcus and Lachnospira after the induction of remission. The combination of these taxa into a MDI showed a positive correlation with clinical disease severity and a negative correlation with species richness. The increased capacity for the inferred pathways including Lipopolysaccharide biosynthesis and Lipopolysaccharide biosynthesis proteins in patients before treatment negatively correlated with the abundance of Roseburia,Ruminococcus and Lachnospira.CONCLUSION The dysbiosis of mucosa-associated microbiota was associated with the disease phenotype and may become a potential diagnostic tool for the recurrence of disease.Cong He Huan Wang Wang-Di Liao Chao Peng Xu Shu Xuan Zhu Zhen-Hua Zhu 2019World Journal of Gastroenterology2019,25,18:3
Mapping Toll-Like Receptor Signaling Pathway Genes of Zhikong Scallop(Chlamys farreri) with FISH显示文摘Toll-like receptor(TLR) signaling pathway plays a pivotal role in the innate immune system. Studies on TLR signaling pathway genes in Zhikong scallop(Chlamys farreri) have mainly focused on sequence analysis and expression profiling, no research has been carried out on their localization. The chromosomal position of TLR signaling pathway genes can be valuable for assemblying scallop genome and analysizing gene regulatory networks. In the present study, five key TLR signaling pathway genes(Cf TLR, Cf Myd88, Cf TRAF6, Cf NFκB, and Cf IκB) containing bacterial artificial chromosomes(BACs) were isolated and physically mapped through fluorescence in situ hybridization on five non-homologous chromosome pairs, showing a similar distribution to another five model species. The isolation and mapping of these key immune genes of C. farreri will aid to the research on innate immunity, assignment of interested genes to chromosomes, and integration of physical, linkage and cytogenetic maps of this species.ZHAO Bosong ZHAO Liang LIAO Huan CHENG Jie LIAN Shanshan LI Xuan HUANG Xiaoting BAO Zhenmin 2015Journal of Ocean University of China2015,14,6:3
Effect of Daxx on cholesterol accumulation in hepatic cells显示文摘AIM:To study the effect of Daxx on cholesterol accumulation in hepatic cells. METHODS: Sprague Dawley (SD) rats were fed a normal or high fat diet for 6 wk, and serum lipids and Daxx expression of hepatic tissues were measured by immunoblot assays. HepG2 cells were transfected with the pEGFP-C1/Daxx or pEGFP-C1 plasmid. Cells stably transfected with Daxx were identified by RT- PCR analysis. Total cholesterol levels were determined by high performance liquid chromatography. Activated- SREBP and caveolin-1 were assayed by western blotting. RESULTS: Hepatic Daxx protein was higher in normal rats than in high fat diet-fed rats. Noticeable negative correlations were seen between Daxx and LDL-C (γ = -7.56, P = 0.018), and between Daxx and TC (γ = -9.07, P = 0.01), respectively. The total cholesterol of HepG2/GFP-Daxx cells was lower than that of control cells or HepG2/GFP cells (9.28 ± 0.19 vs 14.36 ± 4.45 or 13.94 ± 2.62, both P < 0.05). Furthermore, in HepG2/GFP cells, the expression of activated SREBP was lower than that of control cells, whereas caveolin-1 expression was higher. CONCLUSION: Overexpression of Daxx in HepG2 cells decreased intracellular cholesterol accumulation, which might be associated with inhibition of SREBP activity and an increase in caveolin-1 expression.Qin-Hui Tuo Lei Liang Bing-Yang Zhu Xuan Cao Duan-Fang Liao 2008World Journal of Gastroenterology2008,14,3:3
Quantitative Analysis of Natural and Human Factors of Oasis Change in the Tail of Shiyang River over the Past 60 Years显示文摘The human and natural factors complicit in the driving forces of oasis change have always received considerable interest from the international research community. In this study, we used principal component analysis of natural and socio-economic statistical factors to quantitatively analyze the causal relationships and their contributions to the observed periodic expansion or shrinkage of the Minqin Oasis over almost 60 years. Our results show that human factors were the dominant factors governing expansion or shrinkage, with average contributions of 69.38% and 76.16%, respectively. Moreover, policy decisions have been the pivotal human factors. Under the influence of various policies, we have found that water resource utilization, land reclamation, population explosion, ecological protection and economic development have each played leading roles in different periods. This study provides a scientific basis for modelling the dynamics of an oasis for sustainable management.CHEN Yong YANG Guojing ZHOU Lihua LIAO Jie WEI Xuan 2020Acta Geologica Sinica(English Edition)2020,94,3:2

Investigation on color variation of Morpho butterfly wings hierarchical structure based on PCA显示文摘Morpho butterfly wings show brilliant blue color, which has a close relationship with the hierarchical micro/nano structures on the surface of scales. When liquids such as methanol, ethanol and isopropanol with different refractive indexes drop onto the scales, the reflectance of wings would decrease. The main peaks of reflectance shift to the right and the scales turn to yel-low-green.After the liquids volatilizes completely, the scales revert to the original blue color. The typical micro/nano struc-tures of Morpho butterfly wing scales are modeled, and the reflectance variation in the course of dropping liquids is simulated.Furthermore, the principal component analysis(PCA) method is employed to analyze the experimental and simulation reflec-tance data, extract principal components and reduce dimensions. As a result, the curves mapping the reflectance variation are clearly shown in the coordinate system consisting of three principal components. The typical color variation of scales during the dropping process could be monitored, and different kinds of liquids could be obviously distinguished according to the dis-tributed regions of mapping points. This study provides guidance to environmental media detecting as well as data processing, and enhances the fabrication and application of the mimic Morpho butterfly wings micro/nano structures.JIANG Xuan SHI TieLin ZUO HaiBo YANG XueFeng WU WenJun LIAO GuangLan 2012Science China(Technological Sciences)2012,55,1:2
