| 1 | Genome-wide Dissection of Co-selected UV-B Responsive Pathways in the UV-B Adaptation of Qingke显示文摘Qingke (Tibetan hulless barley) has long been cultivated and exposed to long-term and strong UV-B radiation on the Tibetan Plateau, which renders it an ideal species for elucidating novel UV-B responsive mechanisms in plants. Here we report a comprehensive metabolite profiling and metabolite-based genome-wide association study (mGWAS) using 196 diverse qingke and barley accessions. Our results demonstrated both constitutive and induced accumulation, and common genetic regulation, of metabolites from different branches of the phenylpropanoid pathway that are involved in UV-B protection. A total of 90 significant mGWAS loci for these metabolites were identified in barley-qingke differentiation regions, and a number of high-level metabolite trait alleles were found to be significantly enriched in qingke, suggesting co-selection of various phenylpropanoids. Upon dissecting the entire phenylpropanoid pathway, we identified some key determinants controlling natural variation of phenylpropanoid content, including three novel proteins, a flavone C-pentosyltransferase, a tyramine hydroxycinnamoyl acyltransferase, and a MYB transcription factor. | 3 | Semen Analysis and Fecundity Association Among Women with Polycystic Ovary Syndrome Experiencing Ovulatory Dysfunction Treated by Ovulation Induction显示文摘In this study,normal values of semen analysis were set for a general infertile population of couples among which most women had normal ovulation.The predictive capacity values of sperm quality,including concentration,motile count,progressive motile count,and morphology,are unclear for women with polycystic ovary syndrome(PCOS).A secondary analysis was conducted based on a randomized controlled trial investigating infertility among women with PCOS experiencing ovulatory disorder between 2011 and 2016 in China.A total of 1000 women received ovulation induction(acupuncture and clomiphene).We randomized the women with PCOS in 27 hospitals in China who received one of four interventions(acupuncture plus clomiphene,sham acupuncture plus clomiphene,acupuncture plus placebo,or sham acupuncture plus placebo).Semen analysis was performed for every male partner according to the World Health Organization(WHO)criteria.The outcomes included conception,clinical pregnancy,and live birth.Logistic regression was used to evaluate the predictive value of semen analysis among ovulatory women for conception,clinical pregnancy,and live birth.Among the 1000 couples,the number of couples who attained ovulation,conception,clinical pregnancy,and live birth were 780,320,235,and 205,respectively.Semen volume and motility were applied and used as prediction parameters for conception(area under the curve(AUC)of 0.62(95%confidence interval(CI),0.55–0.69)),clinical pregnancy(AUC of 0.67(95%CI:0.61–0.73)),and live birth(AUC of 0.57(95%CI:0.50–0.64)).No poor calibration was shown for these models in Hosmer–Lemeshow tests.The predictive capacity of semen analysis for treatment outcome in PCOS women with PCOS experiencing with ovulatory dysfunction is limited. | Jingshu Gao Yu Wang Mubai Li Mengyi Zhu Xuekui Liu Hongli Ma Yijuan Cao Lu Li Xinming Yang Xiaoke Wu | 2021 | Engineering2021,7,11: | 1 |
| 4 | CryoEM structure of the tegumented capsid of Epstein-Barr virus显示文摘Epstein-Barr virus(EBV)is the primary cause of infectious mononucleosis and has been shown to be closely associated with various malignancies.Here,we present a complete atomic model of EBV,including the icosahedral capsid,the dodecameric portal and the capsid-associated tegument complex(CATC).Our in situ portal from the tegumented capsid adopts a closed conformation with its channel valve holding the terminal viral DNA and with its crown region firmly engaged by three layers of ring-like dsDNA,which,together with the penton flexibility,effectively alleviates the capsid inner pressure placed on the portal cap.In contrast,the CATCs,through binding to the flexible penton vertices in a stoichiometric manner,accurately increase the inner capsid pressure to facilitate the pressure-driven genome delivery.Together,our results provide important insights into the mechanism by which the EBV capsid,portal,packaged genome and the CATCs coordinately achieve a pressure balance to simultaneously benefit both viral genome retention and ejection. | Zhihai Li Xiao Zhang Lili Dong Jingjing Pang Miao Xu Qian Zhong Mu-Sheng Zeng Xuekui Yu | 2020 | Cell Research2020,30,10: | 1 |
| 5 | A telomere-to-telomere reference genome provides genetic insight into the pentacyclic triterpenoid biosynthesis in Chaenomeles speciosa显示文摘Chaenomeles speciosa(2n=34),a medicinal and edible plant in the Rosaceae,is commonly used in traditional Chinese medicine.To date,the lack of genomic sequence and genetic studies has impeded efforts to improve its medicinal value.Herein,we report the use of an integrative approach involving PacBio HiFi(third-generation)sequencing and Hi-C scaffolding to assemble a high-quality telomere-to-telomere genome of C.speciosa.The genome comprised 650.4 Mb with a contig N50 of 35.5 Mb.Of these,632.3 Mb were anchored to 17 pseudo-chromosomes,in which 12,4,and 1 pseudo-chromosomes were represented by a single contig,two contigs,and four contigs,respectively.Eleven pseudo-chromosomes had telomere repeats at both ends,and four had telomere repeats at a single end.Repetitive sequences accounted for 49.5%of the genome,while a total of 45515 protein-coding genes have been annotated.The genome size of C.speciosa was relatively similar to that of Malus domestica.Expanded or contracted gene families were identified and investigated for their association with different plant metabolisms or biological processes.In particular,functional annotation characterized gene families that were associated with the biosynthetic pathway of oleanolic and ursolic acids,two abundant pentacyclic triterpenoids in the fruits of C.speciosa.Taken together,this telomere-to-telomere and chromosome-level genome of C.speciosa not only provides a valuable resource to enhance understanding of the biosynthesis of medicinal compounds in tissues,but also promotes understanding of the evolution of the Rosaceae. | Shaofang He Duanyang Weng Yipeng Zhang Qiusheng Kong Keyue Wang Naliang Jing Fengfeng Li Yuebin Ge Hui Xiong Lei Wu De-Yu Xie Shengqiu Feng Xiaqing Yu Xuekui Wang Shaohua Shu Zhinan Mei | 2023 | Horticulture Research2023,10,10: | 0 |
