| 1 | Progress in quantitative analysis of plant hormones显示文摘Plant hormones are small molecular natural products that regulate all plant developmental processes at low concentrations. Quantitative analysis of plant hormones is increasingly important for in-depth study of their biosynthesis,transport,metabolism and molecular regulatory mechanisms. Although plant hormone analysis remains a bottleneck in plant scientific research owing to the trace concentrations and complex components in plant crude extracts,much progress has been achieved in the development of extraction,purification and detection techniques in recent years. Solid phase extraction and chromatography/mass spectrometry have been applied widely for purification and quantitative analysis of plant hormones owing to their high selectivity and sensitivity. Purification methods such as liquid partition and immunoaffinity chromatography,and detection methods including immunoassay and electrochemical analysis,are employed. The advantages and disadvantages of these methods are discussed. | 2 | Expression Patterns of ABA and GA Metabolism Genes and Hormone Levels during Rice Seed Development and Imbibition: A Comparison of Dormant and Non-Dormant Rice Cultivars显示文摘Seed dormancy is an important agronomic trait in cereals. Using deep dormant(N22), medium dormant(ZH11), and non-dormant(G46B)rice cultivars, we correlated seed dormancy phenotypes with abscisic acid(ABA) and gibberellin(GA) metabolism gene expression profiles and phytohormone levels during seed development and imbibition. A time course analysis of ABA and GA content during seed development showed that N22 had a high ABA level at early and middle seed developmental stages, while at late developmental stage it declined to the level of ZH11; however, its ABA/GA ratio maintained at a high level throughout seed development. By contrast, G46 B had the lowest ABA content during seed development though at early developmental stage its ABA level was close to that of ZH11, and its ABA/GA ratio peaked at late developmental stage that was at the same level of ZH11. Compared with N22 and G46 B, ZH11 had an even and medium ABA level during seed development and its ABA/GA ratio peaked at the middle developmental stage. Moreover, the seed development time-point having high ABA/GA ratio also had relatively high transcript levels for key genes in ABA and GA metabolism pathways across three cultivars. These indicated that the embryo-imposed dormancy has been induced before the late developmental stage and is determined by ABA/GA ratio. A similar analysis during seed imbibition showed that ABA was synthesized in different degrees for the three cultivars. | 6 | Shake table experimental study of cable-stayed bridges with two different design strategies of H-shaped towers显示文摘As one of the main load-carrying components of cable-stayed bridges,bridge towers are typically required to remain elastic even when subjected to severe ground motions with a 2%-3%probability of exceedance in 50 years.To fulfill this special requirement imposed by current seismic design codes,reinforcement ratios in the bridge towers have to be kept significantly higher than if limited ductility behavior of the tower is allowed.In addition,since the foundation capacity is closely related to the moment and shear capacities of the bridge tower,a large increase in bridge construction cost for elastically designed cable-stayed bridge is inevitable.To further investigate the possibility of limited ductility bridge tower design strategies,a new 1/20-scale cable-stayed bridge model with H-shaped bridge towers designed according to strong strut-weak tower column design was tested.The shake table experimental results are compared with a previous strong tower column-weak strut designed full bridge model.A comparison of the results show that ductility design with plastic hinges located on tower columns,i.e.,strong strut-weak tower column design,is another effective seismic design strategy that results in only small residual displacement at the top of the tower column,even under very severe earthquake excitations. | Xu Yan Cui Cunyu Zeng Zeng Zeng Shijie | 2021 | Earthquake Engineering and Engineering Vibration2021,20,2: | 0 |
