| 2 | Role of CD56-expressing immature biliary epithelial cells in biliary atresia显示文摘AIM: To analyze the clinical and pathological parameters and expression of the neural cell adhesion molecule(CD56) in patients with biliary atresia(BA).METHODS: Established clinical laboratory markers of hepatic function, including enzyme activity, protein synthesis, and bilirubin metabolism, were evaluated in patients with BA and compared with those in patients with choledochal cysts and neonatal hepatitis. Pathological changes in tissue morphology and fibrosis were examined by histological and tissue collagen staining. Immunohistochemical staining for the biliary epithelial cell markers CD56 and CK19 together with the Notch signaling related molecules Notch1 and Notch2 was performed in the context of alterations in the structure of intrahepatic biliary ducts.RESULTS: Differences in some clinical laboratoryparameters among the three diseases examined were observed, but they did not correlate with the pathological classification of fibrosis in BA. Immunohistochemical staining showed the presence of CD56-positive immature bile ducts in most patients(74.5%) with BA but not in patients with choledochal cysts or neonatal hepatitis. The number of CD56-expressing cells correlated with disease severity, with more positive cells present in the later stages of liver damage(81.8% vs 18.2%). Furthermore, bile plugs were mainly found in CD56-positive immature biliary ducts. Notch signaling was a key regulatory pathway in biliary duct formation and played a role in tissue fibrosis. Notch1 was co-expressed in CD56-positive cells, whereas Notch2 was found exclusively in blood vessels in the portal area of patients with BA. | 3 | Mycotoxin detection——Recent trends at global level显示文摘Mycotoxin contamination in agro-food systems has been a serious concern globally during the last few decades. Mycotoxins are toxic secondary metabolites produced by fungi when they grow in agro-food products and feedstuff. Several detection techniques have been developed in recent years to detect mycotoxins in the food and feed effectively. HPLC based techniques are very common in usage in the laboratories for the testing of mycotoxins. In recent years,immuno-based assays is widely used and have been reported at large due to its sensitivity and limited detection time. Immuno assay-based kits were developed effectively to be used in the fields and in storage systems to detect the mycotoxin levels. Microarray-based immunoassays developed in the recent years could simultaneously detect aflatoxin,ochratoxin,and zearalenone with the higher sensitivity. Aptamer-based assays could target the detection of ochratoxin and aflatoxins and fumonisins at high specificity in food products. In recent years,several assays reported for the simultaneous multiple detection of different mycotoxin was based on HPLC and LC-MS/MS. | 4 | Downregulation of Hes1 expression in experimental biliary atresia and its effects on bile duct structure显示文摘AIM To analyze the expression and function of the notchsignaling target gene Hes1 in a rhesus rotavirusinduced mouse biliary atresia model. METHODS The morphologies of biliary epithelial cells in biliary atresia patients and in a mouse model were examined by immunohistochemical staining. Then, the differential expression of Notch signaling pathway-related molecules was investigated. Further, the effects of the si RNAmediated inhibition of Hes1 expression were examined using a biliary epithelial cell 3 D culture system.RESULTS Both immature(Ep CAM+) and mature(CK19+) biliary epithelial cells were detected in the livers of biliary atresia patients without a ductile structure and in the mouse model with a distorted bile duct structure. The hepatic expression of transcripts for most Notch signaling molecules were significantly reduced on day 7 but recovered to normal levels by day 14, except for the target molecule Hes1, which still exhibited lower m RNA and protein levels. Expression of the Hes1 transcriptional co-regulator, RBP-Jκ was also reduced. A 3 D gel culture system promoted the maturation of immature biliary epithelial cells, with increased expression of CK19+ cells and the formation of a duct-like structure. The administration of Hes1 si RNA blocked this process. | 5 | Formula for calculating spatial similarity degrees between point clouds on multi-scale maps taking map scale change as the only independent variable显示文摘The degree of spatial similarity plays an important role in map generalization, yet there has been no quantitative research into it. To fill this gap, this study first defines map scale change and spatial similarity degree/relation in multi-scale map spaces and then proposes a model for calculating the degree of spatial similarity between a point cloud at one scale and its generalized counterpart at another scale. After validation, the new model features 16 points with map scale change as the x coordinate and the degree of spatial similarity as the y coordinate.Finally, using an application for curve fitting, the model achieves an empirical formula that can calculate the degree of spatial similarity using map scale change as the sole independent variable, and vice versa. | 6 | Calculation of Ecological Footprint Based on Modified Method and Quantitative Analysis of Its Impact Factors——A Case Study of Shanghai显示文摘Ecological footprint(EF) has attracted much attention internationally as a potential indicator for sustainable development.In China,EF has also gained recognition in related fields and disciplines.Moreover,some scholars in these fields have proposed alternative methods to calculate EF and have made some progress.This paper,therefore,begins with the introduction and development of EF in China.However,the established methods of EF calculation hold some limitations,such as indicator variance and result abnormality.In order to resolve those problems,the authors make a further modification considering the demand of EF as a comprehensive indicator:1) More accurate analysis has been done to divide EF into several parts:imported EF,exported EF,and producible EF,which is the solution to the problem of abnormality in original EF results and can explain abnormal phenomena reasonably.2) Considering the actual situa-tion of Shanghai,amended equivalence factor is brought forward and a matrix is formed with equivalence factors.The measure can reduce the deviation between the fact and the results.3) The calculation compares local yield with global average yield to analyze the effects of yields.And based on local yields in different years,the results are more accurate.Finally,the calculation method is applied to calculating EF of Shanghai from 1980 to 2003,and the subsequent detailed analysis is presented.Available data and results suggest a statistically significant correlation coefficient between EF and GDP,population density and urbanization level.Through analyzing the process of calculating EF and its results,it can be seen that EF,as a macro-indicator,can not exactly indicate whether development within a region can meet the re-quests of sustainable development,which can be explained by the fact that the result of EF is impacted greatly by sub-jective factors including national policy,available technology,population,etc.Nevertheless,EF can demonstrate,at least to some degree,the regional status in terms of resources and energy consumption,as well as developmental potential.The calculation of EF,therefore,deserves further research to achieve more far-reaching significance in application. | GAO Chengkang JIANG Dahe WANG Dan YAN Jonathan | 2006 | Chinese Geographical Science2006,16,4: | 4 |
| 7 | Geochemistry, Geochronology and Lu-Hf Isotopes of Peraluminous Granitic Porphyry from the Walegen Au Deposit, West Qinling Terrane显示文摘Walegen Au deposit is closely correlated with granitic intrusions of Triassic age, which are composed of granite and quartz porphyries. Both granite porphyry and quartz porphyry consist of quartz, feldspar and muscovite as primary minerals. Weakly peraluminous granite porphyry(A/CNK=1.10–1.15) is enriched in LREE, depleted in HREE with Nb-Ta-Ti anomalies, and displays subduction-related geochemistry. Quartz porphyry is strongly peraluminous(A/CNK=1.64–2.81) with highly evolved components, characterized by lower TiO_2, REE contents, Mg~#, K/Rb, Nb/Ta, Zr/Hf ratios and higher Rb/Sr ratios than the granite porphyry. REE patterns of quartz porphyry exhibit lanthanide tetrad effect, resulting from mineral fractionation or participation of fluids with enriched F and Cl. LAICP-MS zircon U-Pb dating indicates quartz porphyry formed at 233±3 Ma. The ages of relict zircons from Triassic magmatic rocks match well with the detrital zircons from regional area. In addition, ε_(Hf)(t) values of Triassic magmatic zircons from the granite and quartz porphyries are -14.2 to -9.1(with an exception of +4.1) and -10.8 to -8.6 respectively, indicating a crustal-dominant source. Regionally, numerous Middle Triassic granitoids were previously reported to be formed under the consumption of Paleotethyan Ocean. | 10 | Silurian Sedimentation in the South Qilian Belt: Arc-Continent Collision-related Deposition in the NE Tibet Plateau?显示文摘The South Qilian belt mainly comprises an early Paleozoic arc-ophiolite complex, accretionary prism, microcontinental block, and foreland basin. These elements represent accretion-collision during Cambrian to Silurian time in response to closure of the Proto-Tethyan Ocean in the NE of the present-day Tibet Plateau. Closure of the Proto-Tethyan Ocean between the Central Qilian block and the Oulongbuluke block and the associated collision took place from NE to SW in a zipper-like style. Sediment would have been dispersed longitudinally SW-ward with a progressive facies migration from marginal alluvial sediments toward slope deep-water and deep-sea turbidites. This migration path indicates an ocean basin that shrank toward the SW. The Balonggongga'er Formation in the western South Qilian belt represents the fill of a latest Ordovician-Silurian remnant ocean basin that separated the Oulongbuluke block from the Central Qilian block, and records Silurian closure of the Proto-Tethyan Ocean and subduction beneath the Central Qilian block. However, alluvial deposits in the Lajishan area were accumulated in a retro-foreland basin, indicating that continent-continent collision in the eastern South Qilian belt occurred at c. 450–440 Ma.
