| 1 | Development of Chinese characterization factors for land use in life cycle impact assessment显示文摘A model with Chinese characterization factors for quantifying the damages to environment by land use in terms of the change in net primary productivity (NPP) of ecosystem was developed in the framework of life cycle assessment (LCA).In this method,the forms of land utilization were divided into two aspects involving long-term use of land (e.g.,for arable farming),namely 'land occupation',and the change of land properties (e.g.,from agricultural area to urban area),namely 'land transformation'.Furthermore,the land use elementary flows (occupation and transformation) and parameters were linked to the impact indicators,and the characterization formulas of the two forms of land utilization were derived respectively according to the ecological theory.Moreover,based on this method,the characterization factors of both land occupation and land transformation were calculated using Chinese empirical information on NPP,which can be incorporated into LCA framework and applied to Chinese LCA case study to fill up the important gap in life cycle impact assessment (LCIA) of land use. | LIU Yu,NIE ZuoRen,SUN BoXue,WANG ZhiHong & GONG XianZheng School of Materials Science and Engineering,Beijing University of Technology,Beijing 100124,China | 2010 | Science China(Technological Sciences)2010,53,6: | 8 |
| 2 | Genome-wide identification and expression analysis of Gossypium RING-H2 finger E3 ligase genes revealed their roles in fiber development,and phytohormone and abiotic stress responses显示文摘Background: RING-H2 finger E3 ligase(RH2FE3) genes encode cysteine-rich proteins that mediate E3 ubiquitin ligase activity and degrade target substrates. The roles of these genes in plant responses to phytohormones and abiotic stresses are well documented in various species, but their roles in cotton fiber development are poorly understood. To date, genome-wide identification and expression analyses of Gossypium hirsutum RH2FE3 genes have not been reported.Methods: We performed computational identification, structural and phylogenetic analyses, chromosomal distribution analysis and estimated K_a/K_s values of G. hirsutum RH2FE3 genes. Orthologous and paralogous gene pairs were identified by all-versus-all BLASTP searches. We predicted cis-regulatory elements and analyzed microarray data sets to generate heatmaps at different development stages. Tissue-specific expression in cotton fiber, and hormonal and abiotic stress responses were determined by quantitative real time polymerase chain reaction(qRT-PCR) analysis.Results: We investigated 140 G. hirsutum, 80 G. arboreum, and 89 G. raimondi putative RH2FE3 genes and their evolutionary mechanisms and compared them with orthologs in Arabidopsis and rice. A domain-based analysis of the G.hirsutum RH2FE3 genes predicted conserved signature motifs and gene structures. Chromosomal localization showed the genes were distributed across al G. hirsutum chromosomes, and 60 duplication events(4 tandem and 56 segmental duplications) and 98 orthologs were detected. cis-elements were detected in the promoter regions of G. hirsutum RH2FE3 genes. Microarray data and qR T-PCR analyses showed that G. hirsutum RH2FE3 genes were strongly correlated with cotton fiber development. Additional y, almost al the identified genes were up-regulated in response to phytohormones(brassinolide, gibberel ic acid(GA), indole-3-acetic acid(IAA), and salicylic acid(SA)) and abiotic stresses(cold, heat,drought, and salt).Conclusions: The genome-wide identification, comprehensive analysis, and characterization of conserved domains and gene structures, as wel as phylogenetic analysis, cis-element prediction, and expression profile analysis of G. hirsutum RH2FE3 genes and their roles in cotton fiber development and responses to plant hormones and abiotic stresses are reported here for the first time. | 3 | Analysis of gene expression patterns with cDNA microarray during late stage of spermatogenesis in mice显示文摘The differentiation process of round spermat-ids to spermatozoa during the late stage of spermatogenesis is called spermiogenesis. To explore spermiogenesis-related genes, cDNA microarray was used to study expression patterns of 1176 genes in pachytene spermatocytes, round spermatids and elongating spermatids of Balb/c mice. The results showed that 208 genes were detected in all the three cell types. Most of them were down-regulated from pachytene spermatocytes to round spermatids and elongating spermatids. However, up-regulation of 7 genes expression in round spermatids and 3 genes in elongating spermatids were found. Expression of 7 differentially expressed genes in cDNA arrays was further confirmed by semi-quantitative RT-PCR study. The RT-PCR results indicated that the expression of 6 genes was consistent with that in cDNA arrays, only one gene did not show differential expression by RT-PCR. | 10 | Abnormal expression of cen-trosome protein (centrin) in spermatozoa of male human infertility显示文摘To study the relations between male infertility and centrosome protein (centrin) and the functions of centrin in spermatogenesis, the matured spermatozoa of 10 normal male people and 18 male infertility patients were stained by immunofluorescence labeling antibody against centrin. The results showed that two fluorescence signal dots appeared in the normal male spermatozoa and were located at the base of flagellum. They are proximal centriole and distal centriole. However, in some spermatozoa of the male infertility, centrin protein was located abnormally at the base of flagellum and its staining signals were spread, the normal proximal and distal centrioles were confused and could not be recognized separately. These results suggest that abnormality of centrosome protein may be related to male infertility. | 13 | GhKLCR1, a kinesin light chain-related gene, induces drought-stress sensitivity in Arabidopsis显示文摘Drought stress results in significant losses in agricultural production, and especially that of cotton. The molecular mechanisms that coordinate drought tolerance remain elusive in cotton. Here, we isolated a drought-response gene GhKLCR1, which is a close homolog of AtKLCR1, which encodes a kinesin light chain-related protein enriched with a tetratrico peptide-repeat region.A subcellular localization assay showed that GhKLCR1 is associated with the cell membrane. A tissue-specific expression profile analysis demonstrated that GhKLCR1 is a cotton root-specific gene. Further abiotic and hormonal stress treatments showed that GhKLCR1 was upregulated during abiotic stresses, especially after polyethylene glycol treatments. In addition, the glucuronidase(GUS) staining activity increased as the increment of mannitol concentration in transgenic Arabidopsis plants harboring the fusion construct PGhKLCR1::GUS. The root lengths of 35 S::GhKLCR1 lines were significantly reduced compared with that of wild type. Additionally, seed germination was strongly inhibited in 35 S::GhKLCR1 lines after 300-mmol L^(-1) mannitol treatments as compared with Columbia-0, indicating the sensitivity of GhKLCR1 to drought.
