| 4 | Genome Array on Differentially Expressed Genes of Skin Tissue in Cashmere Goat at Early Anagen of Cashmere Growth Cycle Using DNA Microarray显示文摘In order to study the molecular mechanism involved in cashmere regeneration, this study investigated the gene expression profile of skin tissue at various stages of the cashmere growth cycle and screen differentially expressed genes at proangen in 10 cashmere goats at 2 years of age using agilent sheep oligo microarray. Significance analysis of microarray(SAM) methods was used to identify the differentially expressed genes, Hierarchical clustering was performed to clarify these genes in association with different cashmere growth stages, and GO(Gene ontology) and the pathway analyses were con-ducted by a free web-based Molecular Annotation System3.0(MAS 3.0). Approximately 10 200 probe sets were detected in skin tissue of 2-yr-old cashmere goat. After SAM analysis of the microarray data, totally 417 genes were shown to be differentially expressed at different cashmere growth stages, and 24 genes are significantly up-regulated(21) or down-regulated(3) at proangen concurrently compared to angen and telogen. Hierarchical clustering analysis clearly distinguished the differentially expressed genes of each stage. GO analysis indicated that these altered genes at proangen were predominantly involved in collagen fibril organization, integrin-mediated signaling pathway, cell-matrix adhesion, cell adhesion, transforming growth factor-β(TGF-β) receptor signaling pathway, regulation of cell growth. Kyoto encyclopedia of genes and genomes(KEGG) analysis showed that the significant pathways involved mainly included focal adhesion and extracellular matrixc(ECM)-receptor interaction. Some important genes involved in these biological processes, such as COL1A1, COL1A2, COL3A1, SPARC, CYR61 and CTGF, were related to tissue remolding and repairing and detected by more than one probe with similar expression trends at different stages of cashmere growth cycle. | 15 | Hyperthermia promotes degradation of the acute promyelocytic leukemia driver oncoprotein ZBTB16/RARα显示文摘The acute promyelocytic leukemia(APL)driver ZBTB16/RARαis generated by the t(11;17)(q23;q21)chromosomal translocation,which is resistant to combined treatment of all-trans retinoic acid(ATRA)and arsenic trioxide(ATO)or conventional chemotherapy,resulting in extremely low survival rates.In the current study,we investigated the effects of hyperthermia on the oncogenic fusion ZBTB16/RARαprotein to explore a potential therapeutic approach for this variant APL.We showed that Z/R fusion protein expressed in HeLa cells was resistant to ATO,ATRA,and conventional chemotherapeutic agents.However,mild hyperthermia(42℃)rapidly destabilized the ZBTB16/RARαfusion protein expressed in HeLa,293T,and OCI-AML3 cells,followed by robust ubiquitination and proteasomal degradation.In contrast,hyperthermia did not affect the normal(i.e.,unfused)ZBTB16 and RARαproteins,suggesting a specific thermal sensitivity of the ZBTB16/RARαfusion protein.Importantly,we found that the destabilization of ZBTB16/RARαwas the initial step for oncogenic fusion protein degradation by hyperthermia,which could be blocked by deletion of nuclear receptor corepressor(NCoR)binding sites or knockdown of NCoRs.Furthermore,SIAH2 was identified as the E3 ligase participating in hyperthermia-induced ubiquitination of ZBTB16/RARα.In short,these results demonstrate that hyperthermia could effectively destabilize and subsequently degrade the ZBTB16/RARαfusion protein in an NCoR-dependent manner,suggesting a thermal-based therapeutic strategy that may improve the outcome in refractory ZBTB16/RARα-driven APL patients in the clinic. | Qian-qian Wang Liaqat Hussain Pei-han Yu Chang Yang Chen-ying Zhu Ya-fang Ma Si-chun Wang Tao Yang Yuan-yuan Kang Wen-juan Yu Yasen Maimaitiyiming Hua Naranmandura | 2023 | Acta Pharmacologica Sinica2023,44,4: | 1 |
| 16 | Economy, environmental assessment and energy conservation for separation of isopropanol/diisopropyl ether/water multi-azeotropes via extractive distillation coupled pervaporation process显示文摘This wok proposed the extraction distillation coupled pervaporation(ED+PV) technology process using two different solvents to separate isopropanol(IPA) and diisopropyl ether(DIPE) from DIPE/IPA/H_(2)O ternary heterogeneous azeotropes in industrial wastewater from the synthesis of isopropanol in this study.Based on strict design specifications, simulation and sequential iteration methods are used for process design and optimization. Compared to the ethylene glycol(EG)-EG+H_(2)O process and the 1,3-propanediol(PDO)-IPA+H_(2)O process, the total annual cost(TAC) of the EG-IPA+H_(2)O process decreased by 20.76% and 7.86%(PDO). Compared to the EG-EG+H_(2)O process, the TAC of the PDO-IPA+H_(2)O process reduced 14%, but the global warming potential(GWP) and human toxicity of the PDO-IPA+H_(2)O process increased 11.3% and 4.07% respectively. Compared to the PDO-IPA+H_(2)O process, the EG-IPA+H_(2)O process saves 7.86%(TAC), 9.78%(GWP) and 9.85%(human toxicity). The ED+PV process with EG is superior to PDO in factors of TAC, energy consumption, human toxicity and environment. The EG-IPA+H_(2)O process changed the separation order of the products of the multi-azeotropic system, reduced the cost and energy conservation of the system, and enhanced the environmental protection evaluation of the process, is the best process through life cycle assessment for analyzing the economy, energy conservation, environmental assessment and human toxicity, designing cleaner products, controlling waste discharge, and promoting the chemical purification industry.
