1Palcernuine, the first [5/6/6/6]-cernuane-type alkaloid from Palhinhaea cernua f.sikkimensis显示文摘One novel(named palcernuine, 1) and five known cernuane-type(2–6) alkaloids were isolated from the whole plant of Palhinhaea cernua f. sikkimensis. The structure of 1, possessing an unprecedented [5/6/6/6]-tetracyclic ring system containing two nitrogen atoms, was established on the basis of spectroscopic methods, and its absolute configuration was determined by comparison of the experimental and calculated electronic circular dichroism(ECD) spectra. 5Tumor-specific and photothermal-augmented chemodynamic therapy by ferrocene-carbon dot-crosslinked nanoparticles显示文摘Extensive research have been devoted to the exploration of multifunctional theranostic agents for cancer,but the poor tumor specificity and unsatisfactory treatment efficacy are some of the critical obstacles for their clinical translations.Herein,ferrocene-carbon dot-crosslinked nanoparticles(Fc-CD NPs)were designed and fabricated for achieving highly specific and photothermal-augmented chemodynamic therapy(CDT).The Fc-CD NPs were found not only to inherit the immanent fluorescence,photoacoustic,and photothermal properties of carbon dots(CDs),but also be endowed with CDT that could occur selectively in tumor microenvironment(TME)due to the presence of Fc for triggering Fenton reaction.Moreover,the enlarged particle size of Fc-CD NPs facilitated their effective accumulation at tumor sites,thus realizing great improvement for antitumor treatment outcomes.Once docking at tumor and being exposed to 660 nm laser irradiation,significantly amplified CDT effect of Fc-CD NPs was observed due to heataccelerating generation of reactive oxygen species(ROS).More interestingly,since the produced ROS could in turn alleviate the thermal-resistance of photothermal therapy(PTT),the therapeutic efficiency of integrated PTT and CDT was synergized to the maximum extent.This study on the one hand provides a facile approach to fabricate CDs-based multifunctional theranostic nanoplatform with enhanced tumor accumulation and specificity,on the other hand emphasizes the merits of synergizing mutually beneficial therapeutic modalities for more efficient cancer therapy.Shan Sun Qiao Chen Yike Li Yao Yu Zhongjun Li Hengwei Lin 2022SmartMat2022,3,2:1

7Miocene Crustal Anatexis of Paleozoic Orthogneiss in the Zhada Area,Western Himalaya显示文摘Granitic gneiss(orthogneiss)and Himalayan leucogranite are widely distributed in the Himalayan orogen,but whether or not the granitic gneiss made a contribution to the Himalayan leucogranite remains unclear.In this study,we present the petrological,geochronological and geochemical results for orthogneisses and leucogranites from the Zhada area,Western Himalayas.Zhada orthogneiss is composed mainly of quartz,plagioclase,K-feldspar,biotite and muscovite,with accessory zircon and apatite.Orthogneiss zircon cathodoluminescence(CL)images show that most grains contain a core with oscillatory zoning,which indicates an igneous origin.Sensitive high-resolution ion microprobe(SHRIMP)U-Pb dating of the zircon cores in the orthogneiss shows a weighted ^(206)Pb/^(238)U age of 515±4 Ma(early Paleozoic),with sponge-like zircon rims of 17.9±0.5 Ma(Miocene).Zhada leucogranite shows^(206)Pb/^(238)U ages ranging from 19.0±0.4 Ma to 12.4±0.2 Ma,the weighted average age being 16.2±0.4 Ma.The leucogranites have a low Ca content(<1 wt%),FeOt content(<1 wt%),Rb content(67.0-402 ppm),Sr content(<56.6 ppm),Ba content(3.35-238 ppm)and Rb/Sr ratio(0.5-14.7),which are similar to the geochemical characteristics of the Himalayan leucogranite derived from muscovite dehydration partial melting of metasediments and representative of most Himalayan leucogranites.The highly variable Na_(2)O+K_(2)O(4.33 wt%-9.13 wt%),Al_(2)O_(3)(8.44 wt%-13.51 wt%),ΣREE(40.2-191.0 ppm),Rb(67.0-402 ppm)and Nb(8.23-26.4 ppm)contents,^(87)Sr/^(86)Sr(t)ratios(0.7445-0.8605)andεNd(t)values(−3.6 to−8.2)indicate that the leucogranite is derived from a heterogenetic source.The nonradiogenic Nd isotope values of the studied Zhada leucogranite and orthogneiss range from−8.2 to−3.6 and from−8.7 to−4.1,respectively.Therefore,the general mixing equation was used to perform the Sr and Nd isotope mixing calculations.The results indicate that the heterogenetic source was the Tethyan Himalayan Sequence(THS)/Higher Himalayan Crystalline(HHC)metasediments and Zhada orthogneiss.The Zhada area experienced crustal anatexis during the Miocene and the heterogenetic source of the orthogneiss and metasediment may have experienced crustal anatexis controlled by muscovite dehydration.The Zhada leucogranite inherited not only the geochemical characteristics of the Himalayan metasediment(muscovite dehydration melting),but also the trace elements and Sr-Nd isotopic characteristics of the Zhada orthogneiss.These results indicate that the Paleozoic Zhada orthogneiss was involved in crustal anatexis at 17.9±0.5 Ma(Miocene)and that the muscovite dehydration of the metasediments in the heterogenetic source produced fluid,which may have caused the orthogneiss solidus lines to decline,triggering a partial melting of the Zhada orthogneiss.It is therefore proposed that Himalayan leucogranite is a crust-derived granite rather than a S-type granite,as previously hypothesized.CHEN Han HU Guyue ZENG Lingsen YU Xuhui LI Yike 2022Acta Geologica Sinica(English Edition)2022,96,3:1

14Bioinspired porous microspheres for sustained hypoxic exosomes release and vascularized bone regeneration显示文摘Exosomes derived from mesenchymal stem cells(MSCs)have demonstrated regenerative potential for cell-free bone tissue engineering,nevertheless,certain challenges,including the confined therapeutic potency of exosomes and ineffective delivery method,are still persisted.Here,we confirmed that hypoxic precondition could induce enhanced secretion of exosomes from stem cells from human exfoliated deciduous teeth(SHEDs)via comprehensive proteomics analysis,and the corresponding hypoxic exosomes(H-Exo)exhibited superior potential in promoting cellular angiogenesis and osteogenesis via the significant up-regulation in focal adhesion,VEGF signaling pathway,and thyroid hormone synthesis.Then,we developed a platform technology enabling the effective delivery of hypoxic exosomes with sustained release kinetics to irregular-shaped bone defects via injection.This platform is based on a simple adsorbing technique,where exosomes are adsorbed onto the surface of injectable porous poly(lactide-co-glycolide)(PLGA)microspheres with bioinspired polydopamine(PDA)coating(PMS-PDA microspheres).The PMS-PDA microspheres could effectively adsorb exosomes,show sustained release of H-Exo for 21 days with high bioactivity,and induce vascularized bone regeneration in 5-mm rat calvarial defect.These findings indicate that the hypoxic precondition and PMS-PDA porous microsphere-based exosome delivery are efficient in inducing tissue regeneration,hence facilitating the clinical translation of exosome-based therapy.Yike Gao Zuoying Yuan Xiaojing Yuan Zhuo Wan Yingjie Yu Qi Zhan Yuming Zhao Jianmin Han Jianyong Huang Chunyang Xiong Qing Cai 2022Bioactive Materials2022,7,8:1

18Ovalbumin-loaded paramagnetic nano-triangles for enhanced dendritic cell stimulation,T_(1)-MR imaging,and antitumor immunity显示文摘Vaccine-based cancer immunotherapy has demonstrated a significant potential for cancer treatment in clinics.Although the efficiencies of vaccines are limited,they can be enhanced by a well-designed antigen delivery system that promotes sufficient antigen presentation of dendritic cells(DCs)for initiating high T cell immunity.Herein,antigen-loaded manganese oxide(Mn_(3)O_(4))triangular-shaped ultrasmall nanoparti-cles were prepared to stimulate DC-based immunotherapy under the guidance of T_(1)magnetic resonance imaging.The FDA-approved triblock copolymer Pluronic^(■)F-68 wasused not onlyto transferthe phase from hydrophobic to hydrophilic but also to enrich antigen loading and improve the biocompatibility of the prepared nanoparticles.Ovalbumin(OVA),a model antigen,was adsorbed on the surface of polymer-coated nanoparticles through electrostatic interaction to form Mn_(3)O_(4)@PF68-OVA nanoparticle-antigen complexes to stimulate DC-based immunization and antigen-specific T cell immunity.The Mn_(3)O_(4)@PF68-OVA nanovaccine(NV)induces negligible toxicity effects against 4T1 and bone marrow-derived dendritic cells(BMDCs)by conventional methods supports the proliferation of intestine organoids,which are an innovative three-dimensional cytotoxicity evaluation system,thereby indicating their potential safety for in vivo cancer therapies.The designed paramagnetic nanovaccine possessed excellent OVA delivery to dendritic-regulated antigen-specific T cells in vitro by stimulating the maturation level of BMDCs.In ad-dition,Mn_(3)O_(4)@PF68-OVA NVs enhance immunity in vivo by increasing the T-cells and M1 macrophages,which suggests improved immunity.Excitingly,vaccination with Mn_(3)O_(4)@PF68-OVA offer complete pro-tection in the prophylactic group and significant tumor inhibition in the therapeutic group against B16-OVA tumor.In addition,the designed nanovaccine demonstrated high T_(1)-MR imaging in the tumor,fur-ther justifying enhanced tumor accumulation and capability to real-time monitor the treatment proce-dure.This study presents a promising nanosystem to design an effective nanovaccine for T_(1)-MR imaging-guided tumor immunotherapy.Yike Hou Zhe Tang Jabeen Farheen Madiha Saeed Lijia Luo Wenzi Ren Dandan Luo Asim Mushtaq Ruibo Zhao Jian Ge Zhangsen Yu Yao Li M.Zubair Iqbal Xiangdong Kong 2023Journal of Materials Science & Technology2023,,17:0
