| 1 | Ruminal fermentation and microbial community differently influenced by four typical subtropical forages in vitro显示文摘The present study evaluated the effects of 4 typical subtropical forages on ruminal microbial community composition to formulate a better diet for buffalo.Corn straw silage,elephant grass,cassava residues and sugarcane tail silage were used as substrates for in vitro fermentation.Eight replicates were set up for every substrate,and fermentation was carried out in a 100-m L glass syringe,using buffalo rumen inoculum.Every replicate was anaerobically dispensed with 10 m L of rumen inoculum,20 m L of Mc Dougall's buffer and 200 mg of dried substrate,and placed in a water bath at 39C.Gas production was recorded at 0,2,6,12,24,36,48 and 72 h of incubation.After 24 h,fermentation was ceased for 4 replicates and samples were collected.Volatile fatty acids(VFA) concentrations were measured using gas chromatography.Microbial populations were quantified using quantitative real-time PCR(q RT-PCR),and microbial community was analyzed using high throughput sequencing technology.The results showed,cassava residues as substrate had the highest gas production,acetate,propionate and total VFA concentrations(P < 0.05),and corn straw silage had the lowest acetate:propionate ratio(P < 0.05).The lowest numbers of fungi,Ruminococcus albus and Fibrobacter succinogenes,and the highest number of protozoa were observed with cassava residues(P < 0.05).The least abundances of bacterial phyla Firmicutes,Bacteroidetes and genus Prevotella,and substantially higher abundance of phylum proteobacteria(56%) and genus Succinivibrio(52%) were observed with cassava residues.The most abundances of Methanobrevibacter gottschalkii and Entodinium were observed with cassava residues.Spearman's correlations analysis showed,Succinivibrio had strong positive correlations with propionate,butyrate,Metadinium and M.gottschalkii,indicating fermentation products were related to microbial community.In conclusion,incubation with cassava residues resulted in lower number of fiber degrading microbes but higher protozoal population because of its low fiber contents.The microbial community was highly altered by in vitro incubation with cassava residues,whereas remained similar for the other 3 high fiber containing substrates. | Muhammad W.Iqbal Qin Zhang Yingbai Yang Caixia Zou Lili Li Xin Liang Shengju Wei Bo Lin | 2018 | Animal Nutrition2018,,1: | 6 |
| 2 | Nitraria sibirica cell suspension culture:establishment,characterization and application显示文摘Nitraria sibirica Pall.is a shrub that grows in saline-alkali soil and has traditional medicinal value and potential commercial value.The objectives of this study include induction and multiplication of callus,establishment of a suspension cell line,and isolation of protoplasts from cell suspensions.Murashige and Skoog(MS) medium was used for callus induction from mature seeds of N.sibirica.Seed-derived calluses were further multiplied on MS medium augmented with 0.5 mgL^(-1) 6-benzylaminopurine(6-BA) and 1.0 mgL^(-1) 2,4-dichlorophenoxy(2,4-D) acetic acid.Suspension cultures of N.sibirica were initiated by transferring friable calli to the same liquid multiplication medium.Characterization of the suspension culture was assessed based on fresh mass,dry mass,cell viability and p H value of the culture.A typical growth curve was observed after inoculating 1.5 g of callus in 40 mL liquid medium,including a lag phase,an exponential growth phase,a stationary phase,and a negative acceleration phase.The effect of factors such as pre-plasmolysis,enzyme combination,enzymolysis time and mannitol concentration,on the isolation of cell-derived protoplasts were evaluated to determine the usefulness of suspension cultures.The maximum yield(9.79 9 106 cells/g) and highest viability(79.97%) of protoplast were reached when approximately 1 g of cell suspension(cultured for 6 days) was inoculated for 12 h in cell and protoplast washing solution made of 0.8 molL^(-1) mannitol mixture solution,cellulose onozuka R-10 2%(w/v),hemicellulose 0.2%,macerozyme R-10 1%,and pectolyase Y-23 0.5%.Protoplast yield was significantly influenced by pre-plasmolysis and cellulose onozuka R-10(P<0.05). | Xindi Mei Yingbai Shen | 2017 | Journal of Forestry Research2017,28,5: | 1 |

| 6 | The importance of proleptic branch traits in biomass production of poplar in high-density plantations显示文摘Branch phenotypic traits determine tree crown architecture,which in turn governs leaf display,light interception,and biomass production.Sylleptic and proleptic branches are the obviously different branch phenotypes in the poplar crown.Many studies have focused on the influence of sylleptic branch numbers(SBN)on biomass production,but the research on the influence of proleptic branch phenotypes was only a few.To explore the relationship between proleptic branch traits and biomass generation production in a high-density poplar plantation,we investigated the branch phenotypic traits of three poplar genotypes,all of which have high survival rates in forests(>95%)and significantly different crown architecture and biomass performance in the high-density plantations(1667 stems ha−1).The plantation site was established in 2007.A terrestrial laser scanner was used to measure branch characteristics such as length,angle of origin and termination,and azi-muth angle.A hierarchical cluster analysis performed on branch characteristics showed that SBN,crown depth,and proleptic branch curvature(PBC)were clustered with bio-mass production and leaf area index(LAI).Among all of the monitored traits,PBC played the second most important role in biomass production after SBN and was significantly correlated with SBN,LAI,and biomass production.The positive correlation between PBC and SBN indicated that a larger PBC was associated with more sylleptic branches within the monitored genotypes planted in the high-density plantation,providing greater leaf area and biomass produc-tion.The results of this study will improve the identification of high-production poplar varieties for cultivation in high-density plantations for biofuel production. | Changjun Ding Ningning Wang Qinjun Huang Weixi Zhang Juan Huang Suli Yan Boyi Chen Dejun Liang Yufeng Dong Yingbai Shen Xiaohua Su | 2022 | Journal of Forestry Research2022,33,2: | 1 |

| 14 | Adaptive strategy of Nitraria sibirica to transient salt,alkali and osmotic stresses via the alteration of Na+/K+fluxes around root tips显示文摘Nitraria sibirica Pall.is an important shrub with a strong salt-alkali tolerance,but the mechanism underlying this tolerance remains obscure.In this study,N.sibirica,with salt-sensitive Vigna radiata(Linn.)Wilczek as the control,was subjected to transient salt stress(100 mM NaCl),alkali stress(50 mM Na_(2)CO_(3)),and osmotic stress(175 mM mannitol).The ionic fluxes of Na^(+)and K^(+)in the root apical region were measured.Results show that,under salt and alkali stress,N.sibirica roots exhibited higher capacities to limit Na+influx and reduce K+efflux,thereby resulting in lower Na^(+)/K^(+)ratios compared with V.radiata roots.Alkali stress induced stronger Na^(+)influx and K+efflux in the root salt stress treatment;Na^(+)influx was mainly observed in the root cap,while K^(+)efflux was mainly observed in the elongation zone.While under osmotic stress,N.sibirica roots showed stronger Na+efflux and weaker K+efflux than V.radiata roots.Na+efflux was mainly observed in the root elongation zone,while K+efflux was in the root cap.These results reveal the ionic strategy of N.sibirica in response to transient salt,alkali,and osmotic stresses through the regulation of Na+/K+flux homeostasis. | Xindi Mei Ting Dai Yingbai Shen | 2023 | Journal of Forestry Research2023,34,2: | 0 |
