1Expression of vascular endothelial growth factor C and chemokine receptor CCR7 in gastric carcinoma and their values in predicting lymph node metastasis显示文摘AIM:To study the expression of vascular endothelial growth factor C (VEGF-C) and chemokine receptor CCR7 in gastric carcinoma and to investigate their associations with lymph node metastasis of gastric carcinoma and their values in predicting lymph node metastasis.METHODS:The expression of VEGF-C and CCR7 in gastric carcinoma tissues obtained from 118 patients who underwent curative gastrectomy was examined by immunohistochemisty.Among these patients,39 patients underwent multi-slice spiral CT (MSCT) examination.RESULTS:VEGF-C and CCR7 were positively expressed in 52.5 and 53.4% of patients. VEGF-C expression was more frequently found in tumors with lymph node metastasis than those without it (P<0.001).VEGF-C expression was also closely related to lymphatic invasion (P<0.001), vascular invasion (P<0.01),and TNM stage (P<0.001). However,there was no significant correlation between VEGF-C expression and age at surgery, gender, tumor size, tumor location,Lauren classification,and depth of invasion. CCR7 expression was significantly higher in patients with lymph node metastasis compared with those without lymph node metastasis (P<0.001) and was also associated with tumorsize (P<0.01), depth of invasion (P<0.001), lymphatic invasion (P<0.001),and TNM stage (P<0.001).However,the presence of CCR7 had no correlation to age at surgery,gender, tumor location, Lauren classification,and vascular invasion. Among the 39 patients who underwent MSCT examination,only CCR7 expression was related to lymph node metastasis determined by MSCT (P<0.05).In the current retrospective study,the sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy of VEGF-C and CCR7 expression in the diagnosis of lymph node metastasis for patients with gastric carcinoma were 73.8%, 70.2%,72.6%,71.4% and 72.0%,and 82.0%,77.2%,79.4%,80.0% and 79.7%,respectively.After subdivision according to the combination of VEGF-C and CCR7 expression, receiver operating characteristic (ROC) analysis showed that the accuracy of the combined examination of VEGF-C and CCR7 expression in predicting lymph node metastasis was relatively high (area under ROC curve [Az]=0.83).CONCLUSION:The expression of VEGF-C and CCR7 is related to lymph node metastasis of gastric carcinoma and both of them may become new targets for the treatment of gastric carcinoma.Furthermore,the combined examination of VEGF-C and CCR7 expression in endoscopic biopsy specimens may be useful in predicting lymph node metastasis of gastric carcinoma and deciding the extent of surgical lymph node resection.ChaoYan Zheng-GangZhu Ying-YanYu JunJi YiZhang Yu-BaoJi MinYan JunChen Bing-YaLin 2004World Journal of Gastroenterology2004,10,6:54
2Adenoviral transfer of human intedeukin-10 gene in lethal pancreatitis显示文摘AIM: To evaluate the therapeutic effect of adenoviral-vector-delivered human interleukin-10 (hIL-10) gene on severe acute pancreatitis (SAP) rats. METHODS: Healthy Sprague-Dawley (SD) rats were intraperitoneally injected with adenoviral IL-10 gene (AdvhIL-10), empty vector (Adv0) or PBS solution. Blood, liver, pancreas and lung were harvested on the second day to examine hIL-10 level by ELISA and serum amylase by enzymatic assay. A SAP model was induced by retrograde injection of sodium taurocholate through pancreatic duct. SAP rats were then administered with AdvhIL-10, Adv0and PBS solution by a single intraperitoneal injection 20 min after SAP induction. In addition to serum amylase assay, levels of hIL-10 and tumor necrosis factor-α (TNF-α) were detected by RT-PCR, ELISA and histological study. The mortality rate was studied and analyzed by Kaplan-Meier and log rank analysis.RESULTS: The levels of hIL-10 in the pancreas, liver and lung of healthy rats increased significantly after AdvhIL-10 injection (1.42 ng/g in liver, 0.91 ng/g in pancreas); while there was no significant change of hIL-10 in the other two control groups. The concentration of hIL-10 was increased significantly in the SAP rats after AdvhIL-10 injection (1.68 ng/g in liver, 1.12 ng/g in pancreas) compared to the other two SAP groups with blank vector or PBS treatment (P<0.05). The serum amylase levels remained normal in the AdvhIL-10 transfected healthy rats. However, the serum amylase level was significantly elevated in the other two control SAP rats. In contrast, serum amylase was down-regulated in the AdvhIL-10 treated SAP groups.The TNF-α expression in the AdvhIL-10 treated SAP rats was significantly lower compared to the other two control SAP groups. The pathohistological changes in the AdvhIL-10 treated group were better than those in the other two control groups. Furthermore, the mortality of the AdvhIL-10 treated group was significantly reduced compared to the other two control groups (P<0.05). 3Correlation of thymidylate synthase,thymidine phosphorylase and dihydropyrimidine dehydrogenase with sensitivity of gastrointestinal cancer cells to 5-fluorouracil and 5-fluoro-2′-deoxyuridine显示文摘AIM:To determine the expression levels of three metabolic enzymes of fluoropyrimidines:thymidylate synthase (TS),thymidine phosphorylase (TP) and dihydropyrimidine dehydrogenase (DPD) in seven human gastrointestinal cancer cell lines,and to compare the enzyme levels with the sensitivity to 5-fluorouracil (5-FU) and 5-fluoro-2′-deoxyuridine (FdUrd).METHODS:TS,TP and DPD mRNA levels were assessed by semi-quantitative RT-PCR,TP and DPD protein contents were measured by ELISA. Fifty percent inhibitory concentrations of growth (IC50),representing the sensitivity to drugs,were determined by MTT assay.RESULTS:IC50 values ranged from 1.28 to 12.26μM for 5-FU,and from 5.02 to 24.21μM for FdUrd,respectively.Cell lines with lower DPD mRNA and protein levels tended to be more sensitive to 5-FU (P<0.05), but neither TS nor TP correlated with 5-FU IC50 (P>0.05).Only TS mRNA level was sharply related with FdUrd sensitivity (P<0.05),but TP and DPD were not (P>0.05).A correlation was found between mRNA and protein levels of DPD (P<0.05),but not TP (P<0.05).CONCLUSION:DPD and TS enzyme levels may be useful indicators in predicting the antitumor activity of 5-FU or FdUrd,respectively.TaoMa Zheng-GangZhu Yu-BaoJi YiZhang Ying-YanYu Bing-YaLiu Hao-RanYin Yan-ZhenLin 2004World Journal of Gastroenterology2004,10,2:17
4Polymorphisms at cholesterol 7α-hydroxylase,apolipoproteins B and E and low density lipoprotein receptor genes in patients with gallbladder stone disease显示文摘AIM: To investigate the relationship between gallbladder stone disease (GSD) and single nucleotide polymorphisms of cholesterol 7α-hydroxylase (CYP7A) gene promoter,apolipoprotein (APO) B gene exon 26, APOEgene exon 4 or microsatellite polymorphism of low density lipoprotein receptor (LDLR) gene exon 18.METHODS: Genotypes of CYP7A, APOB, APOE and LDLR genes were determined in 105 patients with GSD diagnosed by B-mode ultrasonography and 274 control subjects.Serum lipids were analyzed with HITACHI 7060 automaic biochemical analyzer.RESULTS: Body mass index (BMI) was significantly higher in patients with GSD (24.47±3.09) than in controls (23.50±2.16).Plasma total cholesterol was lower in patients with GSD (4.66±0.92 mmol/L) than in controls (4.91±0.96 mmol/L),P<0.01 after adjusted for age, sex and BMI. The significantly higher frequency of A allele of CYP7A gene polymorphism and X+ allele of APOBgene polymorphism was seen in GSD patients. Percentages of A allele in patients and controls were 62.86% and 54.38% (P <0.05) and those of X+ allele 8.57% and 4.01% (P<0.01). Subjects with A allele had significantly lower plasma total cholesterol and LDL cholesterol than subjects with CC homozygote. In a multiple variable logistic regression model, the BMI (OR=1.13, 95% CI: 1.05-1.22), A allele (OR=1.48, 95% CI: 1.05-2.09) and X+ allele (OR=2.28, 95% CI: 1.14-4.59) were positively associated with GSD (P <0.05). Plasma total cholesterol (OR=0.69, 95% CI: 0.64-0.74) was negatively related to GSD (P<0.05).CONCLUSION: With an association analysis, it was determined that A allele of CYP7A gene and X+ allele of APOB gene might be considered as risk genes for GSD. 7Tetrandrine inhibits Ca^(2+)-activated chloride channel in cultured human umbilical vein endothelial cells显示文摘AIM: To characterize the electrophysiological and kinetic properties of Ca2+-activated chloride channel (CaCC) in cultured human umbilical vein endothelial cell line (HUVEC), and test the inhibitory effects of tetrandrine (Tet) on CaCC. METHODS: Ca2+-activated Cl- currents (ICl,Ca) were recorded by patch-clamp whole cell configurations. [Ca2+]i was measured via intracellular Fura-2 fluorescence intensities. RESULTS: ICl,Ca was activated by increasing [Ca2+]i via direct elevation of intracellular calcium. ICl,Ca showed an apparent outward rectification properties, and it was activated in a voltage- and calcium-dependent mode. Tet dose-dependently inhibited ICl,Ca, the IC50 was (5.2±0.4) μmol/L (n=8 cells). Tet suppressed both voltage-dependent and calcium-dependent activation of ICl,Ca. The activa- tion time constant was (326±12) ms [in the presence of 10 μmol/L Tet, compared to control (175±17) ms, at +100 mV], and Ca2+ concentration for half maximal activation was (387±61) nmol/L for Tet (compared to control (287±36) nmol/L. 8DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR.Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen 2020Acta Biochimica et Biophysica Sinica2020,52,11:5

10Isotopic Geochemistry of Cadmium:A Review显示文摘Cadmium(Cd) is a scarce, but not an extremely rare element in the Earth's crust(crustal average: 0.2 ppm Cd). Geochemically, Cd exhibits thiophile, lithophile, and volatile behavior in different geologic processes. Biologically, it is a nutrient-like element that is closely related to P and Zn and is toxic element to organisms. Presently, Cd isotopes have been successfully utilized to trace Cd sources and nutrient cycling in marine systems in addition to unearthing other geochemical processes. Using published studies and our recent work, this survey summarizes the chemical preparation and mass spectrometry of Cd isotopes. 12The in vitro reconstitution of nucleosome and its binding patterns with HMG1/2 and HMG14/17 proteins显示文摘Using atomic force microscopy (AFM), the dynamic process of the in vitro nucleosome reconstitution followed by slow dilution from high salt to low salt was visualized. Data showed that the histone octamers were dissociated from DNA at 1M NaCl. When the salt concentration was slowly reduced to 650 mMand 300 mM, the core histones bound to the naked DNA gradually. Once the salt concentration was reduced to 50 mM the classic 'beads-on-a-string' structure was clearly visualized. Furthermore, using the technique of the in vitro reconstitution ofnucleosome,the mono- and di- nucleosomes were assembled in vitro with both HS2core (-10681 to -10970 bp) and NCR2 (-372to -194 bp) DNA sequences in the 5'flanking sequence of human b-globin gene. Data revealed that HMG 1/2 and HMG 14/17 proteins binding to both DNA sequences are changeable following the assembly and disassembly of nucleosomes. 13Involvement of the crebellum in sequential finger movement learning:Evidence from functional magnetic resonance imaging显示文摘Whether the cerebellum is involved in volun-tary motor learning or motor performance is the subject of a new debate. Using functional magnetic resonance imaging (fMRI), we examined cerebellar activation in eight volun-teers before and after an extended period of training. Activa-tion volume on both sides of cerebellum after learning was significantly reduced compared to that before learning even under the same motor frequency. Remarkably, while motor frequency for the training sequence was significantly higher than the control sequence after 41 d of learning, activation in the cerebellum for both sequences, with respect to activation loci and volumes, was very similar. These results suggest that the cerebellum was involved in motor learning but not motor performance. 16HBVPathDB: A database of HBV infection-related molecular interaction network显示文摘AIM: To describe molecules or genes interaction between hepatitis B viruses (HBV) and host, for understanding how virus' and host's genes and molecules are networked to form a biological system and for perceiving mechanism of HBV infection.METHODS: The knowledge of HBV infection-related reactions was organized into various kinds of pathways with carefully drawn graphs in HBVPathDB. Pathway information is stored with relational database management system (DBMS), which is currently the most efficient way to manage large amounts of data and query is implemented with powerful Structured Query Language (SQL). The search engine is written using Personal Home Page (PHP) with SQL embedded and web retrieval interface is developed for searching with Hypertext Markup Language (HTML).RESULTS: We present the first version of HBVPathDB,which is a HBV infection-related molecular interaction network database composed of 306 pathways with 1050molecules involved. With carefully drawn graphs, pathway information stored in HBVPathDB can be browsed in an intuitive way. We develop an easy-to-use interface for flexible accesses to the details of database. Convenient software is implemented to query and browse the pathway information of HBVPathDB. Four search page layout options-category search, gene search, description search,unitized search-are supported by the search engine ofthe database. The database is freely available at http://www.bio-inf, net/HBVPathDB/HBV/.CONCLUSION: The conventional perspective HBVPathDB have already contained a considerable amount of pathway information with HBV infection related, which is suitable for in-depth analysis of molecular interaction network of virus and host.
