1Particle size distribution and characteristics of heavy metals in road-deposited sediments from Beijing Olympic Park显示文摘Due to rapid urbanization and industrialization, heavy metals in road-deposited sediments(RDSs) of parks are emitted into the terrestrial, atmospheric, and water environment, and have a severe impact on residents' and tourists' health. To identify the distribution and characteristic of heavy metals in RDS and to assess the road environmental quality in Chinese parks, samples were collected from Beijing Olympic Park in the present study. The results indicated that particles with small grain size(<150 μm) were the dominant fraction. The length of dry period was one of the main factors affecting the particle size distribution, as indicated by the variation of size fraction with the increase of dry days. The amount of heavy metal(i.e., Cu, Zn, Pb and Cd) content was the largest in particles with small size(<150 μm) among all samples. Specifically, the percentage of Cu, Zn, Pb and Cd in these particles was 74.7%, 55.5%, 56.6% and 71.3%, respectively.Heavy metals adsorbed in sediments may mainly be contributed by road traffic emissions. The contamination levels of Pb and Cd were higher than Cu and Zn on the basis of the mean heavy metal contents. Specifically, the geoaccumulation index(I geo) decreased in the order:Cd>Pb>Cu>Zn. This study analyzed the mobility of heavy metals in sediments using partial sequential extraction with the Tessier procedure. 2Efficacy and safety of trastuzumab combined with chemotherapy for first-line treatment and beyond progression of HER2-overexpressing advanced breast cancer显示文摘Objective: To observe the efficacy and safety of trastuzumab combined with chemotherapy in patients with human epidermal growth factor receptor 2(HER2)-overexpressing advanced breast cancer.Methods: A total of 90 patients with HER2-overexpressing advanced breast cancer were enrolled in this study. All patients were diagnosed with ductal invasive breast cancer by pathological analysis, and were aged between 31–73 years with a median of 51 years. HER2-positivity was defined as 3(+) staining in immunochemistry or amplification of fluorescence in situ hybridization(FISH, ratio ≥2.0). Trastuzumab was administered in combination with chemotherapy as first-line treatment and beyond progression as a secondline, third-line, and above treatment in 90, 34, 14, and 6 patients, respectively. The chemotherapy regimen was given according to normal clinical practice. The response rate was evaluated every two cycles, and the primary endpoints were progression-free survival(PFS) and overall survival(OS). Survival curves were estimated by using Kaplan-Meier graphs and were compared by using log-rank test statistics. Multivariate analysis was done using Cox's proportional hazards regression model, and the level of significance was P<0.05.Results: All 90 patients received at least one dose of trastuzumab, and efficacy could be evaluated in 85 patients. The median follow-up was 50 months. In total, 72(80.00%) patients had visceral metastasis, and 43(47.78%) patients had progressed after one or more extensive chemotherapy regimens for metastatic diseases. The median PFS for first-line trastuzumab was 10 months(range, 2–59 months), and the median OS after metastasis or initially local advanced disease was 22 months(range, 2–116 months).Conclusions: Trastuzumab combined with chemotherapy was active and well-tolerated as a first-line treatment and even beyond progression in HER2-overexpressing advanced breast cancer as a second-line or third-line treatment. 4Efficacy of platinum in advanced triple-negative breast cancer with germline BRCA mutation determined by next generation sequencing显示文摘Objective:To compare the efficacy of platinum-and non-platinum-based regimens as first-line treatment for advanced triple-negative breast cancer(TNBC)and analyze the relationship between their efficacy and BRCA gene status.Methods:Retrospectively analyze clinical data of 220 patients diagnosed pathologically with advanced TNBC and treated at the Department of Breast Oncology,Peking University Cancer Hospital from 2013 to 2018 and evaluate the efficacy of chemotherapy.A total of 114 patients had BRCA1/2 gene tested by next generation sequencing(NGS)using peripheral blood,and we analyzed the correlation between their efficacy and BRCA1/2 gene status.Results:Non-platinum-based chemotherapy(NPCT)was administered to 129 and platinum-based chemotherapy(PBCT)to 91 study patients.The clinical benefit rate(CBR)and median progression-free survival(PFS)were not statistically different between NPCT and PBCT groups.The median overall survival(OS)was 30.0 and 22.5 months for PBCT and NPCT group,respectively[P=0.090,hazard ratios(HR)=0.703].BRCA status was assessed in 114 patients,14 of whom had deleterious germline BRCA1/2(g BRCA)mutations(seven in each group).In PBCT group,the CBR was 85.7%and 35.1%for patients with and without deleterious g BRCA mutations,respectively(P=0.039).The median PFS were 14.9 and 5.3 months and median OS were 26.5 and 15.5 months for patients with and without deleterious g BRCA mutations,respectively(P=0.001,P=0.161,respectively).Patients in PBCT group had significantly greater rates of grade 3-4 anemia(5.5%vs.0%)and thrombocytopenia(8.8%vs.0%),whereas palmar-plantar erythrodysesthesia(12.4%vs.0%)and peripheral neuropathy(8.6%vs.1.1%)occurred more frequently in NPCT group.Conclusions:Platinum-based regimens are more effective in patients with deleterious g BRCA mutations,but no difference in patients without BRCA gene mutations,so non-platinum is an option in patients without BRCA gene mutations considering the toxicity and side effect.And we recommend that patients with advanced TNBC should have BRCA gene test.Nan Wang Kun Li Wenfa Huang Weiyao Kong Xiaoran Liu Weijie Shi Feng Xie Hanfang Jiang Guohong Song Lijun Di Quanren Wang Jianjun Yu Huiping Li 2020Chinese Journal of Cancer Research2020,32,2:7
5Integrin-interacting protein Kindlin-2 induces mammary tumors in transgenic mice显示文摘Kindlin-2, an integrin-interacting protein, regulates breast cancer progression. However, currently, no animal model to study the role of Kindlin-2 in the carcinogenesis of mammary gland is available. We established a Kindlin-2 transgenic mouse model using a mammary gland-specific promoter, mammary tumor virus(MMTV) long terminal repeat(LTR). Kindlin-2 was overexpressed in the epithelial cells of the transgenic mice. The mammary gland ductal trees were found to grow faster in MMTV-Kindlin-2 transgenic mice than in control mice during puberty. Kindlin-2 promoted mammary gland growth as indicated by more numerous duct branches and larger lumens, and more alveoli were formed in the mammary glands during pregnancy under Kindlin-2 overexpression. Importantly, mammary gland-specific expression of Kindlin-2 induced tumor formation at the age of 55 weeks on average. Additionally, the levels of estrogen receptor and progesterone receptor were decreased, whereas human epidermal growth factor receptor 2 and β-catenin were upregulated in the Kindlin-2-induced mammary tumors. 6Long-term anodal block stimulation at sacral anterior roots promoted recovery of neurogenic bladder function in a rabbit model of complete spinal cord injury显示文摘A complete spinal cord injury model was established in experimental rabbits using the spinal cord clip compression method.Urodynamic examination was performed 2 weeks later to determine neurogenic bladder status.The rabbits were treated with anodal block stimulation at sacral anterior roots for 4 weeks.Electrical stimulation of sacral anterior roots improved urodynamic parameters of neurogenic bladder in rabbit models of complete spinal cord injury,effectively promoted urinary function,and relieved urinary retention.Immunohistochemistry results showed that a balance was achieved among expression of muscarinic receptor subunits M2,M3,ATP-gated ion channel P2X3 receptors,and β2-adrenergic receptor,and nerve growth factor expression decreased.These results suggested that long-term sacral anterior root stimulation of anodal block could be used to treat neurogenic bladder in a rabbit model of complete spinal cord injury.Xiaoran Wang Qi Gao Xiaoyu Yang Weihua Wang Xinquan Gu Guifeng Liu Peng Yan Ge Gao Xin Yu Yongjie Wang Jihu Lian Chaoling Shi Yao Wang Li Fan 2012Neural Regeneration Research2012,7,5:6
7DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR.Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen 2020Acta Biochimica et Biophysica Sinica2020,52,11:5
8Combined peripheral natural killer cell and circulating tumor cell enumeration enhance prognostic efficiency in patients with metastatic triple-negative breast cancer显示文摘Objective:Triple-negative breast cancer(TNBC)is a heterogeneous disease with poor prognosis.Circulating tumor cells(CTCs)are a promising predictor for breast cancer prognoses but their reliability regarding progression-free survival(PFS)is controversial.We aim to verify their predictive value in TNBC.Methods:In present prospective cohort study,we used the Pep@MNPs method to enumerate CTCs in baseline blood samples from 75 patients with TNBC(taken at inclusion in this study)and analyzed correlations between CTC numbers and outcomes and other clinical parameters.Results:Median PFS was 6.0(range:1.0–25.0)months for the entire cohort,in whom we found no correlations between baseline CTC status and initial tumor stage(P=0.167),tumor grade(P=0.783)or histological type(P=0.084).However,among those getting first-line treatment,baseline CTC status was positively correlated with ratio of peripheral natural killer(NK)cells(P=0.032),presence of lung metastasis(P=0.034)and number of visceral metastatic site(P=0.037).Baseline CTC status was predictive for PFS in first-line TNBC(P=0.033),but not for the cohort as a whole(P=0.118).This prognostic limitation of CTC could be ameliorated by combining CTC and NK cell enumeration(P=0.049).Conclusions:Baseline CTC status was predictive of lung metastasis,peripheral NK cell ratio and PFS in TNBC patients undergoing first-line treatment.We have developed a combined CTC-NK enumeration strategy that allows us to predict PFS in TNBC without any preconditions.Xiaoran Liu Ran Ran Bin Shao Hope S.Rugo Yanlian Yang Zhiyuan Hu Zewen Wei Fengling Wan Weiyao Kong Guohong Song Hanfang Jiang Xu Liang Ruyan Zhang Ying Yan Guobing Xu Huiping Li 2018Chinese Journal of Cancer Research2018,30,3:4
9PhaseⅠdose-escalation and expansion study of PARP inhibitor,fluzoparib(SHR3162),in patients with advanced solid tumors显示文摘Objective:Fluzoparib(SHR3162)is a novel,potent poly(ADP-ribose)polymerases(PARP)1,2 inhibitor that showed anti-tumor activity in xenograft models.We conducted a phaseⅠ,first-in-human,dose-escalation and expansion(D-Esc and D-Ex)trial in patients with advanced solid cancer.Methods:This was a 3+3 phaseⅠD-Esc trial with a 3-level D-Ex at 5 hospitals in China.Eligible patients for DEsc had advanced solid tumors refractory to standard therapies,and D-Ex enrolled patients with ovarian cancer(OC).Fluzoparib was administered orally once or twice daily(bid)at 11 dose levels from 10 to 400 mg/d.Endpoints included dose-finding,safety,pharmacokinetics,and antitumor activity.Results:Seventy-nine patients were enrolled from March,2015 to January,2018[OC(47,59.5%);breast cancer(BC)(16,20.3%);colorectal cancer(8,10.1%),other tumors(8,10.1%)];48 patients were treated in the D-Esc arm and 31 in the D-Ex arm.The maximum tolerated dose(MTD)was 150 mg bid,with a half-life of 9.14 h.Grade 3/4 adverse events included anemia(7.6%)and neutropenia(5.1%).The objective response rate(ORR)was 30%(3/10)in patients with platinum-sensitive OC and 7.7%(1/13)in patients with BC.Among patients treated with fluzoparib≥120 mg/d,median progression-free survival(m PFS)was 7.2[95%confidence interval(95%CI),1.8-9.3]months in OC,9.3(95%CI,7.2-9.3)months in platinum-sensitive OC,and 3.5(range,2.0-28.0)months in BC.In patients with germline BC susceptibility gene mutation(g BRCAMut)(11/43 OC;2/16 BC),m PFS was 8.9 months for OC(range,1.0-23.2;95%CI,1.0-16.8)and 14 and 28 months for BC(those two patients both also had somatic BRCAMut).Conclusions:The MTD of fluzoparib was 150 mg bid in advanced solid malignancies.Fluzoparib demonstrated single-agent antitumor activity in BC and OC,particularly in BRCAMut and platinum-sensitive OC.Huiping Li Rongrui Liu Bin Shao Ran Ran Guohong Song Ke Wang Yehui Shi Jihong Liu Wenjing Hu Fu Chen Xiaoran Liu Gairong Zhang Chuanhua Zhao Ru Jia Quanren Wang Hope S.Rugo Yifan Zhang Guangze Li Jianming Xu 2020Chinese Journal of Cancer Research2020,32,3:4
10Ages and Hf isotopes of detrital zircons from the Permian strata in the engbatu area(Inner Mongolia) and tectonic implications显示文摘The Central Asian Orogenic Belt(CAOB) was built up through protracted accretion and collision of a variety of terranes/micro-continents during Neoproterozoice Mesozoic time. To understand potential links among Paleozoic subduction and accretionary processes that were operative during the development of the southeastern CAOB, we conducted a combined U-Pb and Hf-isotope analysis of detrital zircons from previously defined Devonian, Carboniferous and Early Permian strata in the Bengbatu area,Inner Mongolia. Detrital zircons from(meta-) sandstones in these strata commonly yield major Paleozoic age populations at ca. 300-261 Ma, 351-300 Ma and 517-419 Ma, and also give several Precambrian ages that range from 2687 Ma to 544 Ma. The youngest ages redefine the deposition of all these strata to be in the Middle Permian(Wordiane Capitanian) or later, much younger than previously considered.These ages, coupled with regional magmatic records, support an interpretation of most surrounding areas as possible detritus sources, including the Mongolian arcs to the north, the Northern Accretionary Orogen to the south, and the intervening Erenhote Hegenshan Ophiolite Belt. Zircons with magmatic ages of ca. 500-350 Ma and ca. 300-261 Ma display a large range of εHf(t) values(-13.97 to +15.31),whereas ca. 350-300 Ma zircons are dominated by positive εHf(t) values(+0.14 to +16.00). These results support the occurrence of two significant shifts of the zircon εHf(t) values, which has tectonic implications for the understanding of the Carboniferouse Permian evolution of the southeastern CAOB. A marked shift from mixed to positive zircon εHf(t) values at 350 -330 Ma likely manifests the incipient opening of the Hegenshan Ocean, due to the slab rollback of the subducting Paleo-Asian Oceanic lithosphere. Another shift from positive to mixed zircon εHf(t) values at ca. 300 Ma likely corresponds to a tectonic switch from syn-orogenic subduction-related to post-orogenic extensional setting, genetically related to the tectonic collapse of a formerly overthickened crust.Bing Xu Guochun Zhao Jianhua Li Dongxing Liu Bo Wang Yigui Han Paul R. 13Biopsy-free in vivo virtual histology of skin using deep learning显示文摘An invasive biopsy followed by histological staining is the benchmark for pathological diagnosis of skin tumors.The process is cumbersome and time-consuming,often leading to unnecessary biopsies and scars.Emerging noninvasive optical technologies such as reflectance confocal microscopy(RCM)can provide label-free,cellular-level resolution,in vivo images of skin without performing a biopsy.Although RCM is a useful diagnostic tool,it requires specialized training because the acquired images are grayscale,lack nuclear features,and are difficult to correlate with tissue pathology.Here,we present a deep learning-based framework that uses a convolutional neural network to rapidly transform in vivo RCM images of unstained skin into virtually-stained hematoxylin and eosin-like images with microscopic resolution,enabling visualization of the epidermis,dermal-epidermal junction,and superficial dermis layers.The network was trained under an adversarial learning scheme,which takes ex vivo RCM images of excised unstained/label-free tissue as inputs and uses the microscopic images of the same tissue labeled with acetic acid nuclear contrast staining as the ground truth.We show that this trained neural network can be used to rapidly perform virtual histology of in vivo,label-free RCM images of normal skin structure,basal cell carcinoma,and melanocytic nevi with pigmented melanocytes,demonstrating similar histological features to traditional histology from the same excised tissue.This application of deep learning-based virtual staining to noninvasive imaging technologies may permit more rapid diagnoses of malignant skin neoplasms and reduce invasive skin biopsies.Jingxi Li Jason Garfinkel Xiaoran Zhang Di Wu Yijie Zhang Kevin de Haan Hongda Wang Tairan Liu Bijie Bai Yair Rivenson Gennady Rubinstein Philip O.Scumpia Aydogan Ozcan 2021Light(Science & Applications)2021,10,12:1

20The first two leading modes of the tropical Pacific and their linkage without global warming显示文摘A discrepancy remains in the first two leading empirical orthogonal function(EOF) modes of the tropical Pacific sea surface temperature anomaly(SSTA) based on observations since the 1980s. The EOF1 mode, representing the El Ni?o-Southern Oscillation(ENSO), is a robust result. However, the EOF2 features either El Ni?o Modoki(EM) or ENSO evolution during different periods, which is probably associated with the impacts of global warming. The underlying question is what the EOF2 mode of the tropical Pacific would be without global warming. Using the CMIP5 preindustrial scenario to exclude the influence of global warming, we find that the EOF1 mode of the tropical Pacific SSTA represents ENSO and that the EOF2 mode is not EM. According to the lead–lag correlation between the ENSO and EOF2 modes, the linkage between these two modes is as follows: …El Ni?o → EOF2 → La Ni?a →–EOF2 → El Ni?o…. By analyzing the evolution of sea surface temperature, surface wind, and subsurface ocean temperature anomalies, we find the mechanism linking the ENSO and EOF2 modes is the air–sea interaction associated with the ENSO cycle. This result suggests that the EOF2 mode represents an aspect of ENSO evolution under preindustrial conditions.
