| 1 | The crosstalk between the caspase family and the cGAS-STING signaling pathway显示文摘Cytosolic nucleic acid sensors are critical for sensing nucleic acids and initiating innate immunity during microbial infections and/or cell death.Over the last decade,several key studies have characterized the conserved mechanism of cyclic guanosine monophosphate-adenosine monophosphate synthase(cGAS)and the downstream signaling adaptor stimulator of interferon genes(STING)initiating the innate immune signaling pathways.Aside from its primary involvement in microbial infections and inflammatory diseases,there is growing interest in the alternate roles of cGAS-STING-mediated signaling.Caspase family members are powerful functional proteins that respond to cellular stress,including cell death signals,inflammation,and innate immunity.Recent studies have uncovered how the caspase family cooperates with the cGAS-STING signaling pathway.Most caspase family members negatively regulate the cGAS-STING signaling pathway.In turn,some caspase family members can also be modulated by cGAS-STING.This review gives a detailed account of the interplay between the caspase family and the cGAS-STING signaling pathway,which will shed light on developing novel therapeutics targeting the caspase family and cGAS-STING signaling in antiviral innate immunity,cancer,inflammatory,and autoimmunity. | Yongai Xiong Yan-Dong Tang Chunfu Zheng | 2021 | Journal of Molecular Cell Biology2021,13,10: | 2 |

| 11 | MHC Class I Assembly Function and Intracellular Transport Routes for Hepatitis B Virus Antigen Cross-presentation by Heat Shock Protein gp96显示文摘Background:During hepatitis B virus(HBV)infection,virus-infected hepatocytes directly cross-present viral antigens and regulate T cell response within the liver microenvironment.However,little is known regarding the regulatory pathways involved in viral antigen presentation in HBV-infected hepatocytes.This study investigated the underlying mechanism of antigen assembly and the HBV antigen-presenting function of major histocompatibility complex(MHC)class I molecules using heat shock protein gp96.Methods:First,western blotting,flow cytometry,co-immuno precipitation,GST pull-down,and confocal microscopic assays were performed to determine whether endogenous gp96 affects MHC-I levels via an antigen presentation pathway.Second,the B3Z assay and an AAV/HBV-infected hepatocyte-specific gp96-deficient mouse model were used to determine whether gp96 knockout functionally impaired peptide cross-presentation and produced a weakened antiviral cytotoxic T cell(CTL)response both in vivo and in vitro.Finally,confocal microscopic analysis and the B3Z assay were employed to show that exogenous gp96-associated peptide was present in MHC-I molecules via the endoplasmic reticulum(ER)-Golgi secretory pathway.Results:Compared with the control,gp96 knockdown significantly reduced the cell surface levels of MHC-I by approximately 75%(P<0.01).Endogenous gp96 interacts with MHC-I and is involved in antigen presentation.Moreover,a weakened antiviral CTL response(34%compared to control mice)has been observed in hepatocyte-specific gp96-deficient mice following HBV infection.gp96 directed exogenous antigen to the ER,and the exogenous gp96-chaperoned peptide was endosome-and proteasome-dependent but not transporter associated with antigen processing dependent.Conclusions:Cellular gp96 promotes the assembly and antigen presentation of MHC class I molecules.In addition,extracellular gp96 served as a natural adjuvant to induce a CTL response in a concerted and regulated manner within different cellular compartments.Our results elucidate the mechanism of assembly of MHC class I molecules by gp96,which may be beneficial for the design of immunotherapy and vaccines. | Lijuan Qin Yongai Liu Yuxiu Xu Yang Li Jun Hu Ying Ju Yu Zhang Shuo Wang Zihai Li Changfei Li Xin Li Songdong Meng | 2022 | Infectious Diseases & Immunity2022,2,3: | 0 |
| 12 | Low Fertility Trap:Theories,Facts and Implications显示文摘The low Fertility Trap Hypothesis proposes that there are three self-reinforcing mechanism--demographic,sociological and economic,working towards a downward spiral in future fertility.Once TFR drops below 1.5,it will be difficult to recover.The fertility recovery emerging in many countries across the world has changed the lowfertility trap theory and also the justification of the theory itself.So low-fertility trap is more likely a pattern summarized from a short-time phenomenon than a generalized social law.As with China,the current fertility level is above 1.5 births per woman,and China is not in the 'Low-fertility trap'.Moreover,most recent surveys suggests that current fertility intention in China is above 1.8 births per woman and more than60 percent of people would have a second child if there is an adjustment of fertility policy.In addition,birth postponement is still playing an important role in reducing TFR in China.Thus,with the high fertility intention,adjustment of family planning policy and decreasing tempo effect,TFR in China will experience upturn but not further decline in the near future.It is lack of evidence to conclude that China has already been or is going to be trapped in low fertility. | Jin Yongai | 2014 | China Population Today2014,31,3: | 0 |

| 14 | Diverging destinies:changing trends of induced abortion in China显示文摘In the era of the second demographic transition,“diverging destinies”theory states that women with the most economic opportunities often obtain gains while women with the least economic opportunities suffer from losses.Using nationally represent-ative data from the 2017 China Fertility Survey,we attempted to examine if diverg-ing destinies theory can explain differences in the incidence of induced abortion among women with differing economic opportunities.We found that women with good economic opportunities(i.e.,well-educated and holding urban hukou)were more likely to have induced abortions between 1980 and 2010 and less likely to do so between 2015 and 2017.Moreover,younger cohorts were far more likely to have induced abortions at a younger age and before marriage.These findings provide sup-portive evidence for diverging destinies in induced abortion scenarios in China.We thus advocate that sexual and reproductive health policy making shows an overarch-ing concern for the interests of disadvantaged groups. | Yongai Jin Wenbo Hu | 2023 | China Population and Development Studies2023,7,1: | 0 |

| 16 | Cellular gp96 upregulates AFP expression by blocking NR5A2 SUMOylation and ubiquitination in hepatocellular carcinoma显示文摘Alpha-fetoprotein (AFP) is the most widely used biomarker for the diagnosis of hepatocellular carcinoma (HCC). However, asubstantial proportion of HCC patients have either normal or marginally increased AFP levels in serum, and the underlyingmechanisms are not fully understood. In the present study, we provided in vitro and in vivo evidence that heat shock protein gp96promoted AFP expression at the transcriptional level in HCC. NR5A2 was identified as a key transcription factor for the AFP gene, andits stability was enhanced by gp96. A further mechanistic study by co-immunoprecipitation, GST pull-down, and molecular dockingshowed gp96 and the SUMO E3 ligase RanBP2 competitively binding to NR5A2 at the sites spanning from aa 507 to aa 539. Thebinding of gp96 inhibited SUMOylation, ubiquitination, and subsequent degradation of NR5A2. In addition, clinical analysis of HCCpatients indicated that gp96 expression in tumors was positively correlated with serum AFP levels.
