1A miRNA-HERC4 pathway promotes breast tumorigenesis by inactivating tumor suppressor LATS1显示文摘The E3 ligase HERC4 is overexpressed in human breast cancer and its expression levels correlated with the prognosis of breast cancer patients. However, the roles of HERC4 in mammary tumorigenesis remain unclear. Here we demonstrate that the knockdown of HERC4 in human breast cancer cells dramatically suppressed their proliferation, survival, migration, and tumor growth in vivo, while the overexpression of HERC4 promoted their aggressive tumorigenic activities. HERC4 is a new E3 ligase for the tumor suppressor LATS1 and destabilizes LATS1 by promoting the ubiquitination of LATS1 miRNA-136-5p and miRNA-1285-5p, expression of which is decreased in human breast cancers and is inversely correlated with the prognosis of breast cancer patients, are directly involved in suppressing the expression of HERC4. 2Preliminary analysis of distribution and variation of perennialmonthly mean water masses in the Bohai Sea,the Huanghai Sea and the East China Sea显示文摘On the basis of perennial monthly mean temperature and salinity data, the classification of monthly water masses at the surface and the bottom in the Bohai Sea, the Huanghai Sea and the East China Sea, has been made by using the method of fuzzy cluster from the modified characteristic of water masses in the shallow water area. 3Shake table study on precast segmental concrete double-column piers显示文摘Seismic capacity,including the ultimate load-carrying capacity and ultimate deformation capacity of precast segmental concrete double-column(PSCDC)piers with steel sleeve(SS)connection or grouted corrugated-metal duct(GCMD)connection,has been verified to be similar to those of cast-in-place(CIP)piers by quasi-static tests.However,the lack of knowledge of seismic response characteristics and damage process of PSCDC piers has limited their application in high-intensity seismic areas.Therefore,shake table tests,using variable types and intensities of seismic ground motions,were performed to investigate the seismic behavior of connection joints and to evaluate the seismic performance of PSCDC piers with SS and GCMD connections.Also,a finite element analysis(FEA)model was developed to study the influence of design parameters on the seismic behavior of the piers.The results showed that the main damage in PSCDC piers was caused by the cyclic opening and closing of connection joints.Under high-intensity ground motions,the PSCDC piers had a lower seismic performance than the CIP piers due to a significant decrease of their integrity and stiffness.The seismic performance of PSCDC piers is comparable to CIP piers when using an appropriate initial stress of the prestressing tendons.Xia Zhanghua Ge Jiping Lin Youqin Qiu Faqiang 2020Earthquake Engineering and Engineering Vibration2020,19,3:5

6Hydrogen sulfide system in the pathogenesis of renovascular hypertension in rats显示文摘Objective To investigate the role of hydrogen sulfide (H2S) synthases / H2S pathway in the pathogenesis of renovascular hypertension. Methods Wistar rats were subdivided into 4 groups: (1) 2-kidney, 1-clip (2K-1C group, n=7), (2) control (n=7), (3) sham (n=7), and (4) 2K-1C plus sodium hydrosulfide (NaHS) (NaHS-treated group, n=7). The systolic blood pressure (SBP) was measured by a tail-cuff method using a pulse transducer once a week. Four weeks later, all rats were killed and the concentration of plasma hydrogen sulfide (H2S), the activity of the H2S synthases in the kidneys on both sides, the plasma angiotensin Ⅱconcentration, and the left-to-whole heart weight ratio were measured. Results The SBP was significantly increased in the 2K-1C group (185.4±14.0mmHg) comparing with those in the sham group ( 112.9±6.5mmHg, , or the NaHS-treated group(134.8±9.5mmHg) (both P<0.01). At 4 weeks, the angiotensin Ⅱ concentration in the plasma was increased in the 2K-1C and NaHS-treated group, comparing with the control and the sham group (306.92±7.03 pg/ml and 240.73±13.22 pg/ml vs 122.6±25.49 pg/ml and 125.95±10.55 pg/ml, respectively, both P<0.05). The plasma H2S concentration and the activity of H2S synthases in the left kidney were decreased in the 2K-1C group comparing with those in the sham and the control groups. There was no difference of the activity of the H2S synthases in the right kidneys among the 4 groups. The left-to-whole heart weight ratio was increased in the 2K-1C and the NaHS-treated group camparing with that in the sham and natural control groups. Conclusions Dysfunction of the H2S synthases/H2S pathway was involved in the 2K-1C-induced renovascular hypertension in rat. Exogenous administration of H2S donor can attenuate the development of hypertension. 9Heat Shock Protein 72 Protects Retinal Ganglion Cells in Rat Model of Acute Glaucoma显示文摘Purpose: To investigate whether the induction of heat shock protein (HSP)72 by heat stress (HS) or zinc (Zn2+ ) administration can increase survival of retinal ganglion cells (RGC) in rat model of acute experimental glaucoma.Methods: Acute glaucoma model was made by intracameral irrigation with BSS at 102 mmHg for two hours in right eyes of male Wistar rats. Glaucoma model rats were treated with HS once a week (six rats) or intraperitoneal injection of zinc sulfate (24.6 mg/kg) every two weeks (six rats), and were referred to as HS group and zinc group, respectively. Untreated model rats served as damage group (six rats). In control groups, quercetin (400 mg/kg) was intraperitoneally injected to inhibit the induction of heat shock proteins 6 hours before HS or zinc administration, and were referred to as HS+que group (six rats) and zinc+que group (six rats), respectively. Subsequent to 16 days of IOP elevation, the rats were sacrificed. Eyes were quickly enucleated, and the retinas were dissected. RGC were labeled with Nissl staining and counted under microscope.Results: The average RGC density in normal Wistar rats was (2504±181) cells/mm2. In damage group, it decreased to (2015±111 ) cells/mm2. The RGC densities at 1,2, and 3 mm from the center of the optic nerve head were (2716±215), (2496±168), and (2317±171) cells/mm2, respectively, for normal rats and (2211±133), (1969±154),and (1872±68) cells/mm2, respectively, for damage group. The latter was significantly lower at all locations compared with the former (P=0.027 for each, Mann-Whitney test).The average RGC densities were (2207±200) cells/mm2 for HS group, (2272±155) cells/mm2 for zinc group, (1964±188) cells/mm2 for HS+que group, (2051 ±214) cells/mm2 for zinc+que group and (2015±111 ) cells/mm2 for damage group. There were significant differences in density of labeled RGCs among the five groups (P=0.040,Kruskal-Wallis test). Both HS and zinc group had higher RGC densities than damage group (P =0.036 between HS and damage group,P=0.019 between zinc and damage group, Mann-Whitney test). There was no significant difference in RGC densitiy between control groups and damage group(P=0.260 between HS+que and damage group,P=0.748 between zinc+que and damage group, Mann-Whitney test).Conclusions: The results demonstrated that the induction of HSP72 in RGCs by HS or zinc administration plays an important role in the survival of RGCs in rat model of acute glaucoma.
