1Expression of TFF2 and Helicobacter pylori infection in carcinogenesis of gastric mucosa显示文摘AIM: To investigate the expression of TFF2 and Helicobacter pyloriinfection in carcinogenesis of gastric mucosa.METHODS: The expression of TFF2 was immunohistochemically analyzed in paraffin-embedded samples from 119 patients with endoscopic biopsy and subtotal gastrectomy specimens of gastric mucosal lesions, including 16 cases of chronic superficial gastritis (CSG), 20 chronic atrophic gastritis (CAG),35 intestinal metaplasia (IN), 23 gastric epithelial dysplasia (GED) and 25 gastric carcinoma (CA), and Helicobacter pylori infection was detected by Warthin-Starry staining.RESULTS: 1:TFF2 was located in the cytoplasm of gastrk mucous neck cell. The expression of TFF2 was 100 %,100 %, 0, 56.5 % and 0 in CSGs, CAGs, INs, GEDs and CAs, respectively. 2: The value of TFF2 positive cell density in CSG with Helicobacter pyloriinfection was higher than that without Helicobacter pyloriinfection. (52.89±7.27vs46.49±13.04, P>0.05); But the value of TFF2 positive cell density in CAG and GED with Helicobacter pyloriinfection was significantly lower than that without Helicobacter pylori infection (18.17±4.09 vs 37.93±13.80, P<0.01 and 14.44±9.32 vs 24.84±10.22, P<0.05).CONCLUSION: Increase of TFF2 expression in CSG is perhaps associated with the protective mechanism after gastric mucosal injury. Decrease of TFF2 expression in CAG possibly attributes to the decrease in the number of gastric gland cell expressing TFF2. Re-expression of TFF2 in gastric epithelial dysplasia implies that TFF2 possibly contributes to the initiation of gastric carcinoma. 2Genetic and Antibiotic Resistance Characteristics ofCampylobacterjejuni Isolated from Diarrheal Patients,Poultry and Cattle in Shenzhen显示文摘Objective To investigate genetic and antibiotic resistance characteristics of Campylobacter jejuni(C. jejuni) isolated from Shenzhen. Methods Multilocs sequence typing and agar dilution methods were used to define the genotype and antibiotic resistance of C. jejuni, respectively. Results In total, 126 C. jejuni strains were isolated. The prevalence of C. jejuni was 5.3% in diarrheal patients. The prevalence in poultry meat(36.5%) was higher than that in cattle meat(1.1%). However, the prevalence in poultry cloacal swabs(27.0%) was lower than that in cattle stool(57.3%). Sixty-two sequence types were obtained, among which 27 of the STs and 10 alleles were previously unreported. The most frequently observed clonal complexes were ST-21(11.9%), ST-22(10.3%), and ST-403(7.1%). ST-21, ST-45, ST-354, ST-403, and ST-443 complexes overlapped between isolates from patients and cattle, whereas ST-45 and ST-574 complexes overlapped between isolates from patients and poultry. All C. jejuni were resistant to at least one antibiotic. The highest resistance rate was toward ciprofloxacin(89.7%), followed by tetracycline(74.6%), and nalidixic acid(69.0%). Conclusion This is the first report of the genotypes and antibiotic resistance of C. jejuni in Shenzhen. 6Three-dimensional label-free morphology of CD8+T cells as a sepsis biomarker显示文摘Sepsis is a dysregulated immune response to infection that leads to organ dysfunction and is associated with a high incidence and mortality rate.The lack of reliable biomarkers for diagnosing and prognosis of sepsis is a major challenge in its management.We aimed to investigate the potential of three-dimensional label-free CD8+T cell morphology as a biomarker for sepsis.This study included three-time points in the sepsis recovery cohort(N=8)and healthy controls(N=20).Morphological features and spatial distribution within cells were compared among the patients'statuses.We developed a deep learning model to predict the diagnosis and prognosis of sepsis using the internal cell morphology.Correlation between the morphological features and clinical indices were analysed.Cell morphological features and spatial distribution differed significantly between patients with sepsis and healthy controls and between the survival and non-survival groups.The model for predicting the diagnosis and prognosis of sepsis showed an area under the receiver operating characteristic curve of nearly 100%with only a few cells,and a strong correlation between the morphological features and clinical indices was observed.Our study highlights the potential of three-dimensional label-free CD8+T cell morphology as a promising biomarker for sepsis.This approach is rapid,requires a minimum amount of blood samples,and has the potential to provide valuable information for the early diagnosis and prognosis of sepsis.MinDong Sung Jong Hyun Kim Hyun-Seok Min Sooyoung Jang JaeSeong Hong Bo Kyu Choi JuHye Shin Kyung Soo Chung Yu Rang Park 2023Light(Science & Applications)2023,12,12:0
7The role of extracellular matrix on unfavorable maternal-fetal interface:focusing on the function of collagen in human fertility显示文摘Extracellular matrix(ECM)is characterized as widespread,abundant,and pluripotent.Among ECM members,collagen is widely accepted as one of the most prominent components for its essential structural property that can provide a scaffold for other components of ECM and the rich biological functions,which has been extensively used in tissue engineering.Emerging evidence has shown that the balance of ECM degradation and remodeling is vital to regulations of maternal-fetal interface including menstrual cycling,decidualization,embryo implantation and pregnancy maintenance.Moreover,disorders in these events may eventually lead to failure of pregnancy.Although the improvement of assisted conception and embryo culture technologies bring hope to many infertile couples,some unfavorable outcomes,such as recurrent implantation failure(RIF),recurrent pregnancy loss(RPL)or recurrent miscarriage(RM),keep troubling the clinicians and patients.Recently,in vitro three-dimensional(3D)model mimicking the microenvironment of the maternal-fetal interface is developed to investigate the physiological and pathological conditions of conception and pregnancy.The progress of this technology is based on clarifying the role of ECM in the endometrium and the interaction between endometrium and conceptus.Focusing on collagen,the present review summarized the degradation and regulation of ECM and its role in normal menstruation,endometrium receptivity and unsatisfying events occurring in infertility treatments,as well as the application in therapeutic approaches to improve pregnancy outcomes.More investigations about ECM focusing on the maternal-fetal interface interaction with mesenchymal stem cells or local immunoregulation may inspire new thoughts and advancements in the clinical application of infertility treatments.Rang Liu Mengyuan Dai Guidong Gong Mei Chen Canhui Cao Tianren Wang Zhenhui Hou Yu Shi Junling Guo Yaoyao Zhang Xi Xia 2022Journal of Leather Science and Engineering2022,4,1:0
8Thermal Stability of Magnetron Sputtered Low Hydrogen Content Diamondlike Carbon显示文摘Response to vacuum annealing at temperatures up to 600℃ for 3h for low hydrogen content diamondlike carbon(LHDLC)films prepared by magnetron sputtering of pure graphite in pure Ar atmosphere has been studied.Optical properties of the as-deposited and annealed films have been characterized by ellipsometry and infrared spectroscopy.No change was found for the films annealed at up to 400℃.The LHDLC begins losing hydrogen after annealing above 400℃ and the sp2 hybrid structure increases.The refractive index of the films increases with increasing annealing temperatures above 500℃.Annealing at 600℃ results in graphitization and the loss of IR transparency.CHENG Degang LÜFanxiu SUN Qinbiao YU Wenxiu YANG Rang 1994Chinese Physics Letters1994,11,5:0
