| 1 | Effects of penehyclidine hydrochloride in small intestinal damage caused by limb ischemia-reperfusion显示文摘AIM:To investigate the protective effect of penehyclidine hydrochloride post-conditioning in the damage to the barrier function of the small intestinal mucosa caused by limb ischemia-reperfusion(LIR) injury. METHODS:Male Wistar rats were randomly divided into three groups(36 rats each) :the sham-operation group(group S) ,lower limb ischemia-reperfusion group(group LIR) ,and penehyclidine hydrochloride postconditioning group(group PHC) .Each group was divided into subgroups(n=6 in each group) according to ischemic-reperfusion time,i.e.immediately 0 h(T1) ,1 h(T2) ,3 h(T3) ,6 h(T4) ,12 h(T5) ,and 24 h(T6) .Bilateral hind-limb ischemia was induced by rubber band application proximal to the level of the greater trochanter for 3 h.In group PHC,0.15 mg/kg of penehyclidine hydrochloride was injected into the tail vein immediately after 3 h of bilateral hind-limb ischemia.The designated rats were sacrificed at different time-points of reperfusion;diamine oxidase(DAO) ,superoxide dismutase(SOD) activity,myeloperoxidase(MPO) of small intestinal tissue,plasma endotoxin,DAO,tumor necrosis factor-α(TNF-α) ,and interleukin(IL) -10 in serum were detected in the rats. RESULTS:The pathological changes in the small intestine were observed under light microscope.The levels of MPO,endotoxin,serum DAO,and IL-10 at T1-T6,and TNF-αlevel at T1-T4 increased in groups LIR and PHC(P<0.05) compared with those in group S,but tissue DAO and SOD activity at T1-T6 decreased(P<0.05) .In group PHC,the tissue DAO and SOD activity at T2-T6,and IL-10 at T2-T5 increased to higher levels than those in group LIR(P<0.05) ;however,the levels of MPO,endotoxin,and DAO in the blood at T2-T6,and TNF-αat T2 and T4 decreased(P<0.05) . | 4 | Oxidative stress and hypoxia-induced factor 1α expression in gastric ischemia显示文摘AIM:To investigate the relation of reactive oxygen species (ROS) to hypoxia induced factor 1α (HIF-1α) in gastric ischemia. METHODS:The animal model of gastric ischemia reperfusion was established by placing an elastic rubber band on the proximal part of the bilateral lower limb for ligature for 3 h and reperfusion for 0,1,3,6,12 or 24 h. Ischemic post-conditioning,three cycles of 30-s reperfusion and 30-s femoral aortic reocclusion were conducted before reperfusion. Histological and immunohistochemical methods were used to assess the gastric oxidative damage and the expression of HIF1-α in gastric ischemia. The malondialdehyde (MDA) content and superoxide dismutase (SOD),xanthine oxidase (XOD) and myeloperoxidase (MPO) activities were determined by colorimetric assays. RESULTS:Ischemic post-conditioning can reduce post-ischemic oxidative stress and the expression of HIF-1α of gastric tissue resulting from limb ischemia reperfusion injury. MDA,SOD,XOD and MPO were regarded as indexes for mucosal injuries from ROS,and ROS was found to affect the expression of HIF-1α under gastric ischemic conditions. CONCLUSION:ROS affects HIF-1α expression under gastric ischemic conditions induced by limb ischemia reperfusion injury. | 5 | Total polysaccharides of the Sijunzi decoction attenuate tumor necrosis factor-α-induced damage to the barrier function of a Caco-2 cell monolayer via the nuclear factor-κB-myosin light chain kinase-myosin light chain pathway显示文摘AIM To explore the protective effects and underlying mechanisms of total polysaccharides of the Sijunzi decoction(TPSJ) on the epithelial barriers in vitro. METHODS Caco-2 cell monolayers were treated with or without TPSJ in the presence or absence of TNF-α, and paracellular permeability and transepithelial electrical resistance(TEER) were measured to evaluate the epithelial barrier function. Immunofluorescence and western blotting were respecti-vely used to evaluate the distribution and expression of the tight junction proteins claudin 1, claudin 2, zo3, and occludin in Caco-2 cells. western blotting was also used to evaluate the cellular expression of myosin light chain(MLC), phosphorylated MLC(pM LC), MLC kinase(MLCK), and nuclear factor(NF)-κB p65. RESULTS TPSJ promoted the proliferation of Caco-2 cells and inhibited TNF-α-induced secretion of pro-inflammatory cyto-kines. Furthermore, TPSJ significantly ameliorated both the reduction of TEER and the increased paracellular permeability observed in tumor necrosis factor(TNF)-α-damaged Caco-2 monolayers. Furthermore, TPSJ remarkably attenuated TNF-α-induced morphological changes, downregulated the expression of claudin 1, claudin 2, zo3, and occludin, and markedly suppressed TNF-α-mediated upregulation of p-MLC and MLCK expression. Finally, TPSJ inhibited the activation and expression of NF-κB p65. | 6 | Velocity forecasts using a combined deep learning model in hybrid electric vehicles with V2V and V2I communication显示文摘Vehicle velocity forecast is an important clue in improving the performance of energy management in hybrid electric vehicles(HEV). This paper presents a new combined model for predicting vehicle’s velocity time series. The main features of the model are to combine the feature extraction capability of deep restricted Boltzmann machines(DBM) and sequence pattern predicting capability of bidirectional long short-term memory(BLSTM). Hence, the model is named as DBMBLSTM. In addition, the DRMBLSTM model utilizes the vehicle driving information and roadside infrastructure information provided respectively through vehicle-to-vehicle(V2V) and vehicle-to-infrastructure(V2I) communication channels to predict vehicle velocity at various length of prediction horizon. Furthermore, the predictions results of this study are compared with the state of the art of vehicle velocity forecasts. The root mean square error(RMSE) is used as an evaluation criteria of predictions accuracy. Finally,these compared prediction model are applied in model predictive control(MPC) energy management strategy for the verifications of fuel economy improvement of a HEV. Simulation results confirm that the proposed combined deep learning model performs better than other five prediction methods. | 7 | A LC-ESI-MS method for the simultaneous determination of madecassoside and its metabolite madecassic acid in rat plasma: comparison pharmacokinetics in normal and collagen-induced arthritic rats显示文摘To develop a simple and highly sensitive high performance liquid chromatography with electrospray ionization mass spectrometric(LC-ESI-MS) method for the simultaneous determination of madecassoside and its major metabolite madecassic acid in rat plasma, and compare the pharmacokinetics of the two compounds in normal and collagen-induced arthritis(CIA) rats. Glycyrrhetinic acid was used as the internal standard(IS). Chromatographic separation was accomplished on an Inertsil ODS-3 column, using a gradient elution with the mobile phase composed of acetonitrile and water acidified with 0.1%(V/V) formic acid. Detection was achieved by ESI-MS under the negative selected ion monitoring(SIM) mode. In normal and CIA rats, madecassoside(30 mg·kg-1) was orally administered for 21 consecutive days from the day of arthritis onset. For madecassoside, the linear range was 10–1 000 ng·mL-1 with the square regression coefficient(r) of 0.998 9, while for madecassic acid, the linear range was 10–500 ng·mL-1 with the square regression coefficient(r) of 0.996 1. The lower limit of quantification was 10 ng·mL-1 for both analytes. The intra- and inter-day precision ranged from 1.78% to 13.42% for madecassoside and 2.30% to 14.90% for madecassic acid, and the accuracy was between –0.95% and 6.30% for madecassoside and between –1.48% and 5.34% for madecassic acid. The average recoveries of madecassoside, madecassic acid and IS from spiked plasma samples were > 81%. The developed method was successfully applied to the pharmacokinetic study of madecassoside and madecassic acid in rats after an oral administration of madecassoside. During initial 7 days of dosing, the cmax and AUC of madecassoside were greatly decreased and Vd/F was markedly increased in CIA rats, and no significant difference was observed on the first day of dosing. In contrast, the T1/2, cmax and AUC of madecassic acid were significantly increased, and Ke of madecassic acid was greatly decreased in CIA rats compared with normal rats. Along with repeated administration of madecassoside, the differences of pharmacokinetic parameters of both madecassoside and madecassic acid between CIA and normal rats gradually subsided. | 10 | Respiratory Response of Dormant Nectarine Vegetative Buds to High Temperature Stress显示文摘High temperature stress (HT) is efficient in breaking endo-dormancy of perennial trees. The effects of HT (50°C) on the respiration of dormant nectarine (Prunus persica var. nectariana cv. Shuguang) vegetative buds were evaluated in the research. We found that bud respiration was transiently inhibited by HT and the pentose phosphate pathway (PPP) and the cytochrome C pathway (CYT) were significantly affected. On the substrate level, PPP was activated in the HT-treated buds compared with the control group. However, the activation did mot occur until hours after HT treatment. The tricarboxylic acid cycle (TCA) in both the HT-treated buds and in the control group proceeded at a low level most of the time compared with total respiration. On the electron transfer level, CYT was transiently inhibited by HT but became significantly active in the later stage. CYT operation in the control group exhibited an attenuation process. The alternative pathway (ALT) fluctuated both in the HT-treated samples and in the control. | 19 | Therapeutic effects of EGF-modified curcumin/ chitosan nano-spray on wound healing显示文摘Dermal injury,including trauma,surgical incisions,and burns,remain the most prevalent socio-economical health care issue in the clinic.Nanomedicine represents a reliable administration strategy that can promote the healing of skin lesions,but the lack of effective drug delivery methods can limit its effectiveness.In this study,we developed a novel nano-drug delivery system to treat skin defects through spraying.We prepared curcumin-loaded chitosan nanoparticles modified with epidermal growth factor(EGF)to develop an aqueous EGF-modified spray(EGF@CCN)for the treatment of dermal wounds.In vitro assays showed that the EGF@CCN displayed low cytotoxicity,and that curcumin was continuously and slowly released from the EGF@CCN.In vivo efficacy on wound healing was then evaluated using full-thickness dermal defect models in Wistar rats,showing that the EGF@CCN had significant advantages in promoting wound healing.On day 12 post-operation,skin defects in the rats of the EGF@CCN group were almost completely restored.These effects were related to the activity of curcumin and EGF on skin healing,and the high compatibility of the nano formulation.We therefore conclude that the prepared nano-scaled EGF@CCN spray represents a promising strategy for the treatment of dermal wounds. | Yue Li QingQing Leng XianLun Pang Huan Shi YanLin Liu SuSu Xiao Ling Zhao Ping Zhou ShaoZhi Fu | 2021 | Regenerative Biomaterials2021,8,2: | 1 |
