2CD137 costimulation enhances the antiviral activity of Vγ9Vδ2-T cells against influenza virus显示文摘Influenza epidemics and pandemics are constant threats to global public health.Although strategies including vaccines and antiviral drugs have achieved great advances in controlling influenza virus infection,the efficacy of these strategies is limited by the highly frequent mutations in the viral genome and the emergence of drug-resistant strains.Our previous study indicated that boosting the immunity of human Vγ9Vδ2-T cells with the phosphoantigen pamidronate could be a therapeutic strategy to treat seasonal and avian influenza virus infections.However,one notable drawback ofγδ-T cell-based immunotherapy is the rapid exhaustion of proliferation and effector responses due to repeated treatments with phosphoantigens.Here,we found that the expression of CD137 was inducible in Vγ9Vδ2-T cells following antigenic stimulation.CD137^(+)Vγ9Vδ2-T cells displayed more potent antiviral activity against influenza virus than their CD137−counterparts in vitro and in Rag2^(-/-)γc^(-/-)mice.We further demonstrated that CD137 costimulation was essential for Vγ9Vδ2-T cell activation,proliferation,survival and effector functions.In humanized mice reconstituted with human peripheral blood mononuclear cells,CD137 costimulation with a recombinant human CD137L protein boosted the therapeutic effects of pamidronate against influenza virus.Our study provides a novel strategy of targeting CD137 to improve the efficacy of Vγ9Vδ2-T cell-based immunotherapy.Yujun Pei Kun Wen Zheng Xiang Chunyu Huang Xiwei Wang Xiaofeng Mu Liyan Wen Yinping Liu Wenwei Tu 2020Signal Transduction and Targeted Therapy2020,5,1:1

5Exposures to temperature beyond threshold disproportionately reduce vegetation growth in the northern hemisphere显示文摘In recent decades,terrestrial vegetation in the northern hemisphere(NH)has been exposed to warming and more extremely high temperatures.However,the consequences of these changes for terrestrial vegetation growth remain poorly quantifed and understood.By examining a satellite-based vegetation index,tree-ring measurements and land-surface model simulations,we discovered a consistent convex patern in the responses of vegetation growth to temperature exposure(TE)for forest,shrub and grass in both the temperate(30°-50°N)and boreal(50°-70°N)NH during the period of 1982-2012.Te response of vegetation growth to TE for the three vegetation types in both the temperate and boreal NH increased convergently with increasing temperature,until vegetation type-dependent temperature thresholds were reached.A TE beyond these temperature thresholds resulted in disproportionately weak positive or even strong negative responses.Vegetation growth in the boreal NH was more vulnerable to extremely high-temperature events than vegetation growth in the temporal NH.Te non-linear responses discovered here provide new insights into the dynamics of northern terrestrial ecosystems in a warmer world.Xiuchen Wu Weichao Guo Hongyan Liu Xiaoyan Li Changhui Peng Craig D.Allen Cicheng Zhang Pei Wang Tingting Pei Yujun Ma Yuhong Tian Zhaoliang Song Wenquan Zhu Yang Wang Zongshan Li Deliang Chen 2019National Science Review2019,6,4:0
6Solute drag-controlled grain growth in magnesium investigated by quasi in-situ orientation mapping and level-set simulations显示文摘Critical properties of metallic materials,such as the yield stress,corrosion resistance and ductility depend on the microstructure and its grain size and size distribution.Solute atoms that favorably segregate to grain boundaries produce a pinning atmosphere that exerts a drag pressure on the boundary motion,which strongly affects the grain growth behavior during annealing.In the current work,the characteristics of grain growth in an annealed Mg-1 wt.%Mn-1 wt.%Nd magnesium alloy were investigated by advanced experimental and modeling techniques.Systematic quasi in-situ orientation mappings with a scanning electron microscope were performed to track the evolution of local and global microstructural characteristics as a function of annealing time.Solute segregation at targeted grain boundaries was measured using three-dimensional atom probe tomography.Level-set computer simulations were carried with different setups of driving forces to explore their contribution to the microstructure development with and without solute drag.The results showed that the favorable growth advantage for some grains leading to a transient stage of abnormal grain growth is controlled by several drivers with varying importance at different stages of annealing.For longer annealing times,residual dislocation density gradients between large and smaller grains are no longer important,which leads to microstructure stability due to predominant solute drag.Local fluctuations in residual dislocation energy and solute concentration near grain boundaries cause different boundary segments to migrate at different rates,which affects the average growth rate of large grains and their evolved shape.Risheng Pei Yujun Zhao Muhammad Zubair Sangbong Yi Talal Al-Samman 2023Journal of Magnesium and Alloys2023,11,7:0
